    Biological compatibility between two temperate lineages of brown dog ticks, Rhipicephalus sanguineus (sensu lato)

    Background: The brown dog tick Rhipicephalus sanguineus (sensu stricto) is reputed to be the most widespread tick of domestic dogs worldwide and has also been implicated in the transmission of many pathogens to dogs and humans. For more than two centuries, Rh. sanguineus (s.s.) was regarded as a single taxon, even considering its poor original description and the inexistence of a type specimen. However, genetic and crossbreeding experiments have indicated the existence of at least two distinct taxa within this name: the so-called "temperate" and "tropical" lineages of Rh. sanguineus (sensu lato). Recent genetic studies have also demonstrated the existence of additional lineages of Rh. sanguineus (s.l.) in Europe and Asia. Herein, we assessed the biological compatibility between two lineages of Rh. sanguineus (s.l.) found in southern Europe, namely Rhipicephalus sp. I (from Italy) and Rhipicephalus sp. II (from Portugal). Methods: Ticks morphologically identified as Rh. sanguineus (s.l.) were collected in southern Portugal and southern Italy. Tick colonies were established and crossbreeding experiments conducted. Morphological, biological and genetic analyses were conducted. Results: Crossbreeding experiments confirmed that ticks from the two studied lineages were able to mate and generate fertile hybrids. Hybrid adult ticks always presented the same genotype of the mother, confirming maternal inheritance of mtDNA. However, larvae and nymphs originated from Rhipicephalus sp. I females presented mtDNA genotype of either Rhipicephalus sp. I or Rhipicephalus sp. II, suggesting the occurrence of paternal inheritance or mitochondrial heteroplasmy. While biologically compatible, these lineages are distinct genetically and phenotypically. Conclusions: The temperate lineages of Rh. sanguineus (s.l.) studied herein are biologically compatible and genetic data obtained from both pure and hybrid lines indicate the occurrence of paternal inheritance or mitochondrial heteroplasmy. This study opens new research avenues and raises question regarding the usefulness of genetic data and crossbreeding experiments as criteria for the definition of cryptic species in ticks

Of these parasites, Aelurostrongylus abstrusus and Troglostrongylus brevior affect the respiratory tract of felids. These lungworms share both the ecological niche and the species of snail (Helix aspersa) acting as intermediate host. Recently, the ability of H. aspersa to shed infective third-stage larvae (L3s) of A. abstrusus and T. brevior in the environment has been demonstrated, matching previous knowledge of mode of transmission of zoonotic lungworms.     Native strains of Beauveria bassiana for the control of Rhipicephalus sanguineus sensu lato

    Background Rhipicephalus sanguineus sensu lato ticks are widespread worldwide due to their adaptability to survive under different environmental conditions. They may act as vectors of a wide range of pathogens to humans and animals and their control is based on the use of chemical products on dogs and in the environment. Alternative control strategies, such as the use of entomopathogenic fungi as bio-control agents have also been investigated. The ability of native strains of Beauveria bassiana sensu lato in causing mortality in different tick species (e.g., Amblyomma cajennense and Rhipicephalus microplus) has been demonstrated. However, limited studies have assessed the use of B. bassiana for the control of R. sanguineus s.l. and none of them have employed native strains of this fungus. Here we investigated the pathogenicity of a native strain of B. bassiana (CD1123) against all developmental stages of R. sanguineus s.l.. Methods Batches of eggs, larvae, nymphs and adult ticks were immersed in a suspension of 107 conidia/ml of B. bassiana s.l., isolated from a R. sanguineus s.l. engorged female. All treatment and control groups were observed for 20 days, and the biological parameters (i.e., mortality, hatching, moulting percentage, pre-oviposition period, oviposition period and rate, eggs production efficiency, reproductive efficiency and fitness indexes) were assessed. Results The effect of the B. bassiana strain tested herein on eggs, larvae, nymphs and adults showed a significantly higher mortality than those of the control groups (p < 0.05) at 5 days post-infection. No infected eggs hatched and no infected larvae moulted. Only 15% of infected nymphs moulted into adults. All biological parameters of treated groups differed significantly (p < 0.001) from those of control groups. Conclusions This study demonstrates that a suspension containing 107 conidia/ml of a native B. bassiana strain is highly virulent towards all life-cycle developmental stages of R. sanguineus s.l. and may be of potential interest as a biological control agent against these ticks

    Conhecimento dos profissionais de saúde acerca da toxoplasmose gestacional e congênita

    O objetivo do presente estudo foi avaliar o conhecimento dos profissionais de saúde acerca da toxoplasmose gestacional e congênita. Para tanto, foi realizado estudo transversal, quantitativo, de outubro a dezembro de 2021, através da aplicação de um questionário digital. Um total de 37 profissionais de nível médio e superior (03 médicos, 20 enfermeiros, 03 técnicos em enfermagem e 11 agentes comunitários de saúde) participaram da pesquisa. O estudo demonstrou que 89,2% (33) dos profissionais tinham conhecimento sobre o agente etiológico da toxoplasmose. O alimento contaminado foi considerado importante meio de transmissão por 91,3% (21) dos profissionais de nível superior, e por 35,7% (5) dos de nível técnico/médio. O aborto foi classificado como manifestação clínica da doença por 78,4% (18) dos profissionais entrevistados. Dentre as medidas profiláticas recomendadas pelos profissionais, 86,5% (32) afirmaram que orientavam a não ingestão de carnes cruas ou mal passadas. Apenas 2,7% (1) deles afirmaram ter realizado algum curso ou treinamento sobre o tema toxoplasmose. A educação em saúde é realizada com mais frequência por enfermeiros como foi relatado por 69,6% (16) do grupo do nível superior. Já entre o grupo de nível médio/técnico, 50,0% (7) relataram que os técnicos de enfermagem são os que realizam educação em saúde na unidade (p = 0,048). Apesar do conhecimento sobre alguns aspectos relacionados à toxoplasmose pelos profissionais de saúde, faz-se necessário ampliar o conhecimento destes profissionais através da oferta de capacitações e treinamentos para estarem aptos na realização de ações preventivas contra esta zoonose negligenciada

    Epidemiology and spatial distribution of Echinococcus granulosus in sheep and goats slaughtered in a hyperendemic European Mediterranean area

    Background: Cystic echinococcosis (CE) is a worldwide parasitic zoonosis caused by the larval stage of Echinococcus granulosus sensu lato affecting livestock, particularly sheep and goats. However, often this parasitosis is underestimated. For this reason, this study aimed to evaluate the epidemiological features and spatial distribution of CE in sheep and goats slaughtered in a hyperendemic Mediterranean area. Methods: A survey was conducted in the Basilicata region (southern Italy) from 2014 to 2019. A total of 1454 animals (1265 sheep and 189 goats) from 824 farms were examined for hydatid cyst detection by visual inspection, palpation and incision of target organs. All the CE cysts were counted and classified into five morphostructural types (unilocular, multiseptate, calcified, caseous and hyperlaminated). Molecular analysis was performed on 353 cysts. For spatial analysis, a kriging interpolation method was used to create risk maps, while clustering was assessed by Moran’s I test. Results: CE prevalence of 72.2% (595/824) and 58.4% (849/1454) was observed at the farm and animal levels, respectively, with higher values in sheep (62.9%) than goats (28.0%). The liver and lungs were the most frequently infected organs in both sheep and goats. Most of recovered cysts were of the calcified and multiseptate morphotypes. All the isolates were identified as E. granulosus sensu stricto (genotypes G1–G3). Spatial distribution showed a moderate clustering of positive animals. Conclusion: The findings of this study can be used to better understand the eco-epidemiology of echinococcosis and to improve CE surveillance and prevention programs in regions highly endemic for CE.Fil: Bosco, Antonio.     Parasites in vegetables used for human consumption: a risk for public health

    Vegetables are pivotal food source to the human beings, representing the source of key nutrients, which in turn the occurrence of chronic diseases. Nonetheless, the consumption of poorly washed or in nature vegetables may expose individuals to the risk of infection by parasites. The aim of this study was to assess the contamination by parasites of medical and veterinary concern in raw vegetables used for human consumption. Fresh vegetables [i.e., lettuces (n = 50), carrots (n = 50) and beet (n = 50)] were acquired in different market places across the municipality of Garanhuns (northeastern Brazil) and the presence of parasites assessed by a sedimentation technique. Parasites were detected in 44.7% (67/150) of the evaluated samples. This study provides important data on the occurrence of zoonotic agents in vegetables. Therefore, strict hygienic measures should be adopted to reduce the risk for vegetable contamination leading to a reduction of human infection by pathogens of medical importance

    Molecular diagnosis of American Cutaneous Leishmaniasis (ACL) in dogs from an endemic area in Pernambuco State, Brazil

    A survey was carried out to detect American cutaneous leishmaniasis (ACL) among dogs in an area where a human outbreak had occurred in the state of Pernambuco, in northeastern Brazil. Domiciled dogs living in the district of Três Ladeiras, Igarassu were used in the present study. The following procedures were performed: The Polymerase Chain Reaction (PCR) (n = 126); the Immunofluorescence Antibody Test (IFAT) (n = 80); and a parasitological examination to detect amastigote forms of Leishmania sp. in skin lesions (n = 43). Associations between the infection in animals and the clinical and epidemiological factors were analyzed using Fisher's exact test or the Pearson’s chi-squared test. In total, 46.8% (59/126) of the samples tested were PCRpositive. Although a higher frequency of positivity was detected among males (46.3 %) and animals aged between 3 and 4 years (50.0 %), no significant associations were recorded for these variables (p> 0.05). Similarly, the clinical signs and aspects related to the environment in which the animal lives did not differ significantly, but differences were recorded for the variable locality. In the IFAT, only 6.2% (5/80) of the dogs were positive and no amastigote forms of Leishmania sp. were detected

    Competência vetorial de Culex quinquefasciatus Say, 1823 exposto a diferentes densidades de microfilárias de Dirofilaria immitis (Leidy, 1856)

    The metropolitan region of Recife, Brazil is endemic for Dirofilaria immitis and has an environment favorable to the development of Culex quinquefasciatus. The goal of this study was to evaluate the vector competence of the Cx. quinquefasciatus RECIFE population for D. immitis transmission. A total of 2,104 females of Cx. quinquefasciatus RECIFE population were exposed to different densities of D. immitis microfilariae blood meals, ranging from 1,820 to 2,900 mf/ml of blood, in a natural membrane apparatus. The results showed a variation between 92.3% and 98.8% of females fed. The exposure of the Cx. quinquefasciatus RECIFE population to different densities of microfilariae did not influence the mortality of the mosquitoes. Infective larvae from D. immitis were observed in the Malpighian tubules beginning on the 12th day, whereas larvae were observed in the head and proboscis beginning on the 13th day following infection. The vector efficiency index (VEI) presented by the mosquitoes ranged from 7.8 to 56.5. The data demonstrates that the Cx. quinquefasciatus RECIFE population has great potential for the transmission of D. immitis, as it allowed the development of the filarid until the infectious stage at the different densities of microfilariae to which it was exposed.A Região Metropolitana do Recife é endêmica para Dirofilaria immitis e possui ambiente favorável para o desenvolvimento de Culex quinquefasciatus. Neste estudo avaliou-se a competência vetorial de Cx. quinquefasciatus população RECIFE para a transmissão de D. immitis. Para tanto, 2.104 fêmeas de Cx. quinquefasciatus população RECIFE foram expostas a diferentes densidades de microfilárias de D. immitis, variando de 1.820 a 2.900 mf/ml de sangue por meio de membrana natural. Os resultados obtidos demonstraram variação de 92,3% a 98,8% de fêmeas ingurgitadas após a alimentação. A exposição de Cx. quinquefasciatus população RECIFE a diferentes densidades de microfilárias não influenciou na mortalidade dos mosquitos. Larvas infectantes de D. immitis foram observadas nos túbulos de Malpighi a partir do 12º dia, enquanto na cabeça e na probóscide foram observadas a partir do 13º dia após a infecção. Os índices de eficiência vetorial (IEV) apresentados pelo culicídeo variaram de 7,8 a 56,5. Os dados obtidos demonstraram que Cx. quinquefasciatus população RECIFE tem grande potencial para a transmissão de D. immitis, pois permitiu o desenvolvimento do filarídeo até o estágio infectante nas diferentes densidades de microfilárias às quais foi exposto.65866

    Parasites with zoonotic potential in canine fecal samples from Garanhuns, Pernambuco, Brazil

    Among the gastrointestinal parasites of dogs that present public health importance, the species of the genus Ancylostoma and Toxocara are the most frequently detected, and have been important cause of soil contamination. The aim of this study was to detect gastrointestinal parasites in dog’s feces collected in households, streets and public squares of the city of Garanhuns, Pernambuco, Brazil. Fecal samples (n = 640) were collected in the environment and evaluated by parasitological methods of Willis-Mollay and Hoffman, Pons and Janer. Out of all samples, 51.09% (327/640) were positive for gastrointestinal parasites. In particular, in 70.94% (232/327) of the samples were detected eggs of Ancylostoma spp., and in 11.01% (36/327) coinfection by Ancylostoma spp. and Toxocara canis. In conclusion, the presence of eggs of zoonotic parasites in feces collected from the environment in the city of Garanhuns represent a risk for the transmission of Cutaneous Larva Migrans and Visceral Larva Migrans for the human population
