    Screening of von willebrand disease in iranian women with menorrhagia

    Results: Mean age of our patients was 32.5 ± 10.6 years. The level of von Willebrand factor in 22.5 and von Willebrand activity in 19.6 of patients was abnormal. The prevalence of vWD among patients with menorrhagia was 24. Conclusions: The high prevalence of vWD among our patients was the same as other previous reports, suggesting low awareness about this disease and under diagnosis of mild cases. Background: Menorrhagia is a common health problem in women, particularly those with bleeding disorders. Little is known about the course of menorrhagia or other bleeding symptoms in women with the most common congenital bleeding disorder, von Willebrand disease (vWD). Objectives: The aim of this study was to estimate the prevalence of vWD in women with diagnosed menorrhagia. Materials and Methods: In this cross-sectional study, a total of 460 consecutive patients, presenting menorrhagia, were analyzed. The initial screening and confirmation tests for the diagnosis of vWD included determination of prothrombin time (PT), partial thromboplastin time (PTT), bleeding time (BT), fibrinogen, factor VIII, vWF antigen, and vWF activity.     Identification of polymorphism in promoter region of growth hormone receptor (GHR) gene and its association with milk related traits in Holstein cows

    It is widely accepted that hormones, growth factors and other agents exert their biological effects on target tissues by binding to specific receptors on the plasma membrane. The variability in constituent sequences of growth hormone receptor (GHR) gene is very important because of its major role in mammary gland development. The aim of the present study is to detect polymorphism in promoter region of GHR gene and its association with milk related traits in Holstein cows. Blood samples were randomly collected from 93 Holstein cows, transported to the laboratory and stored at -20°C for further analysis. DNA was extracted using modified salting-out method and a fragment of 836 base pair from promoter region of GHR gene was amplified by a specific primer pairs using polymerase chain reaction (PCR). The PCR products were digested by AluI restriction enzyme and electrophoresed on 2% agarose gel. Result of enzyme digestion for GHR gene, showed allele AluI(-) with the fragment sizes of 14, 75 and 747, and allele AluI(+) with the fragment sizes of 14, 75, 145 and 602 base pairs. Frequency estimation of AluI(-) and AluI(+) alleles were 56 and 44%, respectively, and mean of heterozygosity was 0.49. Statistical analysis showed that cows with AluI(+) allele had significantly higher milk protein and fat percentage at first lactation compared with cows with AluI(-/-), while there was no significant relation between different genotypes and other traits.Keywords: Growth hormone receptor, polymorphism, milk, HolsteinAfrican Journal of Biotechnology Vol. 9(33), pp. 5460-5464, 16 August, 201

    Prevalence of Chlamydia trachomatis infections in symptomatic women by polymerase chain reaction (PCR) immunofluorescence and Giemsa stain

    Chlamydia trachomatis is a ubiquitous human pathogen that is responsible for the most prevalent bacterial sexually transmitted disease worldwide. Studies show that polymerase chain reaction (PCR) is more sensitive than cellular culture for detection of C. trachomatis infections. The aim of this study is to compare different laboratory methods, including Giemsa staining, direct immunofluorescence assay (DFA) and PCR for detection of C. trachomatis in women with urethral symptoms. In this study, 130 women with urethral symptoms admitted in the gynecology clinic, were used and specimens were obtained with endocervical swab for Giemsa staining, DFA and PCR. All the cases underwent these three techniques. Demographic data and the medical history of patients were obtained by direct interview; however, the mean age of cases was 33.8±9.06. Clinical symptoms included abnormal vaginal discharge in 101 cases (77.7%), spotting in 14 cases (10.8%), dysmenorrheal in 7 cases (5.4%), irritation in 6 cases (4.6%) and dysuria in 2 cases (1.5%). In DFA technique, 5 cases (3.8%) were positive and 3 (2.3%) were suspicious, while in the PCR technique, 6 cases (4.6%) were positive for C. trachomatis. However, 3 suspicious cases with DFA were negative in PCR. There was no positive case for C. trachomatis in Giemsa staining. In conclusion, C. trachomatis was not frequent in this study and it can be concluded that this infection was not a major hygienic problem in the same populations that were previously studied. Consequently, the causes that necessitate monogamy could be related to religious causes. Frequency of Chlamydia detection of DFA and PCR was same in the two groups. Nonetheless, Giemsa staining is not a reliable method for evaluating C. trachomatis.Key words: Chlamydia trachomatis, polymerase chain reaction (PCR), direct immunofluorescence assay (DFA)

    Effect of age on reproductive performance in female Caspian brown trout (Salmo trutta caspious, Kessler 1877)

    Caspian brown trout (Salmo trutta caspius) is one of the economically valuable species in the Caspian Sea.Artificial propagation and production of larvae are the main problems in the early culture of this species.The purpose of this paper is to study the effect of reproductive performance of female broods on opposition reproduction efficiency in Caspian brown trout in the breeding season of 2009. Three groups of female broods (4, 5 and 6 years old) were fertilized with 9 male fish individually. The results showed that,6 year old females have maximum body weight (2150.0 ± 86.6 g), total length (59±2 cm), eggs weight (255.0 ± 30.51 g), egg size (5.37 ± 0.058 mm) and absolute fecundity rate (3060 ± 366.15), while highest average number of ovules in each gram of body weight (16.33 ± 0.58) and relative fecundity (2.08 ± 0.12) belonged to 4 years old females. There were significant differences in mean fertilization rate and survival rate until absorption of yolk sack stage (p<0.05) among the treatments studied. The present study showed that the eggs produced from fertilization of 6 year old female eggs and male mixed milt showed maximum average fertilization percentage (97.5 %), survival rate until eyed stage (92%), hatching percentage (93%), and survival rate until absorption of yolk sack (94.5%)

    Blood pressure alterations in burn patients with septic shock under hydrocortisone treatment

    Background and Objectives: Hydrocortisone is widely used in septic shock cases resistant to fluid and vasopressor therapy. It may result in increased blood pressure and survival. However the efficacy is no established among patients with severe burn and septic shock. Accordingly it was assessed in this study. Materials and Methods: The patients older than 14 years of age with resistant septic shock were enrolled during one-year period. The hydrocortisone was prescribed 100 mg three times per day and the alterations in systolic and diastolic blood pressures were recorded. Results: Twenty-nine patients were enrolled including 19 men and 10 women. The mean age was 37 ± 19 years and the mean burn surface area was 60 ± 20. Fourteen patients had positive blood culture. The most common isolated microorganism were Pseudomonas aeuroginosa in 34.6(10 cases), and then Acinetobacter in 13.8(4 cases). The infection was from wound in 79 and the remaining 21 had pneumonia. Twenty-one patients had good response to hydrocortisone and the increase in systolic and diastolic blood pressures was significant; but the mortality rate was similar. Conclusion: Treatment with hydrocortisone would result in increase in systolic and diastolic blood pressure in burn patients with resistant septic shock. © 2015, Tehran University of Medical Science.     Optimizing psma radioligand therapy for patients with metastatic castration-resistant prostate cancer. A systematic review and meta-analysis

    The aim of the review was to evaluate patient and treatment characteristics for patients with metastatic castration-resistant prostate cancer (mCRPC) treated with PSMA radioligand therapy (PRLT) associated with above-average outcome. The systematic review and meta-analysis followed recommendations by the Preferred Reporting Items for Systematic reviews and Meta-Analysis (PRISMA). We searched for publications in PubMed, Embase, and ClinicalTrials.gov up to 31 September 2020. Thirty-six publications and four duplicates reported 2346 patients. Nearly two-thirds of the patients had bone metastases. Median overall survival (OS) was 16 months. Asymptomatic patients and patients with only lymph node metastases lived longer than symptomatic patients and patients with more extensive metastases. Patients treated with an intensified schedule of177Lu PRLT lived longer than those treated with a conventional schedule. Half of the patients obtained a PSA decline ≥ 50% and these patients lived longer than those with less PSA decline. Approximately 10% of the patients developed hematologic toxicity with anemia grade 3 as the most severe adverse effect. Characteristics for patients, cancer, restaging, and PRLT predict above average overall survival following treatment with PRLT
