Calibrating an ice sheet model using high-dimensional binary spatial data
Rapid retreat of ice in the Amundsen Sea sector of West Antarctica may cause
drastic sea level rise, posing significant risks to populations in low-lying
coastal regions. Calibration of computer models representing the behavior of
the West Antarctic Ice Sheet is key for informative projections of future sea
level rise. However, both the relevant observations and the model output are
high-dimensional binary spatial data; existing computer model calibration
methods are unable to handle such data. Here we present a novel calibration
method for computer models whose output is in the form of binary spatial data.
To mitigate the computational and inferential challenges posed by our approach,
we apply a generalized principal component based dimension reduction method. To
demonstrate the utility of our method, we calibrate the PSU3D-ICE model by
comparing the output from a 499-member perturbed-parameter ensemble with
observations from the Amundsen Sea sector of the ice sheet. Our methods help
rigorously characterize the parameter uncertainty even in the presence of
systematic data-model discrepancies and dependence in the errors. Our method
also helps inform environmental risk analyses by contributing to improved
projections of sea level rise from the ice sheets
‘An attempt to tip the scales’: music and embodied capital in an English secondary school
This article explores class, capital, and aspiration to social mobility in an ethnographic study of an English secondary school. In particular, the article considers the school’s musical instrument programme as a site for inculcating forms of capital, under the auspices of developing skills for upward social mobility. Bringing together Bourdieu’s conceptions of schooling with theories of materiality and situated learning, we contend that this school succeeds in cultivating new forms of embodied capital among students through the development of communities of musical practice. However, a tension remains between emancipatory aims of the programme and the sanctioning and championing of certain forms of dominant cultural practice. The learning environment of the instrument programme highlights the complex and conflicted impact that schooling can have on the development of cultural capital and on reproducing disadvantage and difference
Oncometabolites:linking altered metabolism with cancer
The discovery of cancer-associated mutations in genes encoding key metabolic enzymes has provided a direct link between altered metabolism and cancer. Advances in mass spectrometry and nuclear magnetic resonance technologies have facilitated high-resolution metabolite profiling of cells and tumors and identified the accumulation of metabolites associated with specific gene defects. Here we review the potential roles of such "oncometabolites" in tumor evolution and as clinical biomarkers for the detection of cancers characterized by metabolic dysregulation

Large ensemble modeling of the last deglacial retreat of the West Antarctic Ice Sheet: comparison of simple and advanced statistical techniques
A 3-D hybrid ice-sheet model is applied to the last deglacial retreat of the West Antarctic Ice Sheet over the last  ∼  20 000 yr. A large ensemble of 625 model runs is used to calibrate the model to modern and geologic data, including reconstructed grounding lines, relative sea-level records, elevation–age data and uplift rates, with an aggregate score computed for each run that measures overall model–data misfit. Two types of statistical methods are used to analyze the large-ensemble results: simple averaging weighted by the aggregate score, and more advanced Bayesian techniques involving Gaussian process-based emulation and calibration, and Markov chain Monte Carlo. The analyses provide sea-level-rise envelopes with well-defined parametric uncertainty bounds, but the simple averaging method only provides robust results with full-factorial parameter sampling in the large ensemble. Results for best-fit parameter ranges and envelopes of equivalent sea-level rise with the simple averaging method agree well with the more advanced techniques. Best-fit parameter ranges confirm earlier values expected from prior model tuning, including large basal sliding coefficients on modern ocean beds

Vaccine safety issues at the turn of the 21st century
Global gains in vaccination coverage during the early 21st century have been threatened by the emergence of antivaccination groups that have questioned the effectiveness of vaccines to generate public distrust of vaccines and immunisation programmes. This manuscript summarises six key topics that have been at the centre of global discussions on vaccine safety during the early 21st century: thiomersal in multi-dose non-live vaccines, aluminium adjuvants used with several non-live vaccines, autism and auto-immune conditions as possible consequences of vaccination, a risk of immune overload with increasing numbers of vaccinations, and detrimental non-specific effects (NSEs) of vaccination. For each topic, we describe the hypothesis behind the public concern, the evidence reviewed by the WHO's Global Advisory Committee for Vaccine Safety (GACVS) during 1999-2019, and any significant new data that has emerged since GACVS conclusions were made. Although the scientific evidence on these issues overwhelmingly supports the safety of vaccines, communication messages to caregivers and providers need to condense and convey scientific information in an appropriate way to address concerns contributing to vaccine distrust. In addition, there is need for further studies specifically designed to address both positive and negative NSE of vaccination. The role of GACVS will be increasingly important in evaluating the evidence and engaging the global community in promoting and assuring the safety of vaccines in the decades to come as we move into an era in which we use new vaccination platforms, antigens and formulations
