    Inflammatory and angiogenic protein detection in the human vitreous : cytometric bead assay

    Introduction. To evaluate clinical feasibility and reproducibility of cytometric bead assay (CBA) in nondiluted vitreous samples of patients with age-related macular degeneration (ARMD), diabetic macular edema (DME), and central retinal vein occlusion (CRVO). Methods. Twelve patients from a single clinics day qualified for intravitreal injections (ARMD n = 6, DME n = 3, CRVO n = 3) and underwent a combination treatment including a single-site 23 gauge core vitrectomy which yielded a volume of 0.6 mL undiluted vitreous per patient. Interleukin-6 (IL-6), vascular endothelial growth factor isoform A (VEGF-A), and monocyte chemo-attractant protein-1 (MCP-1) were assessed directly from 0.3 mL at the same day (fresh samples). To assess the reproducibility 0.3 ml were frozen for 60 days at -80°, on which the CBA was repeated (frozen samples). Results. In the fresh samples IL-6 was highest in CRVO (median IL-6 55.8 pg/mL) > DME (50.6) > ARMD (3.1). Highest VEGF was measured in CRVO (447.4) > DME (3.9) > ARMD (2.0). MCP-1 was highest in CRVO (595.7) > AMD (530.8) > DME (178). The CBA reproducibility after frozen storage was examined to be most accurate for MCP1 (P = 0.91) > VEGF (P = 0.68) > IL-6 (P = 0.49). Conclusions. CBA is an innovative, fast determining, and reliable technology to analyze proteins in fluids, like the undiluted vitreous, which is important to better understand ocular pathophysiology and pharmacology. There is no influence of intermittent storage at -80° for the reproducibility of the CBA

    Effect of deflector aerator on stepped spillway flow

    Systematic physical model tests are performed on a stepped spillway equipped with a bottom aerator at the beginning of the stepped part. A deflector is used to issue a jet in order to initiate air entrainment into the flow. A horizontal slot located in the vertical face of the first step allows for air supply underneath the flow. The cavity subpressure was measured to ensure optimal aerator performance, namely atmospheric pressure conditions. The air discharge entrained below the jet is measured to derive the aerator air entrainment coefficient. The local air concentrations are spatially measured downstream of the aerator at regularly spaced profiles, allowing the investigation of air transport and detrainment as well as the average and bottom air concentrations. The present paper focuses on the resulting spatial distribution of air concentration for five deflector geometries. The chute angle, step height, approach flow Froude number and approach flow depth were kept constant, so that the differences occur mostly on the jet length and air entrainment coefficient. The flow depth and the air concentration rapidly converge towards quasi-uniform flow values downstream of the aerator

    Chute aerators protect spillways from cavitation damage. Several parameters affect their efficiency, which were intensively discussed in literature. One implicit parameter is the cavity jet impact angle at the re-attachment point of the flow on the chute bottom, which may however not be defined a priori by the designer. The herein conducted experimental research derived this angle and a related general prediction of it, and shows its effect on the aerator performance. The experiments indicate that no distinctive effect occurs if considering the air entrainment coefficient, but that the relative air detrainment rate downstream of the re-attachment point significantly augments with increasing impact angle

    The influence of time headway on subjective driver states in adaptive cruise control

    There is no agreement on the relation between driving parameters and drivers’ subjective states. A linear as well as a threshold relationship for different subjective variables and driving parameters has been put forward. In this study we investigate the relationship between time headway and the ratings of risk, task difficulty, effort, and comfort. Knowledge about this interrelation may advance the development of adaptive cruise control and autonomous driving and can add to the discussion about driver behavior models. An earlier study (Lewis-Evans, De Waard, & Brookhuis, 2010) found a threshold effect for drivers’ ratings of subjective variables for time headways between 0.5 and 4.0 s at a speed of 50 km/h. This study aims to replicate the threshold effect and to expand the findings to time headways at different speeds. A new measure for criticality was added as a categorical variable, indicating the controllability of a driving situation to give indications for the appliance of time headway in adaptive cruise control systems. Participants drove 24 short routes in a driving simulator with predefined speed and time headway to a leading vehicle. Time headway was varied eightfold (0.5–4 s in 0.5 s increments) and speed was varied threefold (50, 100, 150 km/h). A threshold effect for the ratings of risk, task difficulty, effort, and comfort was found for all three different speeds. Criticality proved to be a useful variable in assessing the preferred time headway of drivers

    Free overfall weirs are hydraulically efficient and reliable in operation. With this, the overfall capacity depends on the hydraulically active developed length of the weir. Folded types of weir, such as the piano key weir (PKW), provide an over-proportionally developed length and are thus especially efficient, in particular for relatively small energy heads. The compact construction of the PKW in addition allows its economic and rapid creation on crest of gravity dams. Therefore, the PKW lends itself if and when the discharge capacity of existing flood spillways is to be increased or in case a subsidiary overflow is to be emplaced under cramped space conditions. The hydraulic functional capability of the PKW has been tested and optimised by means of several model studios. In addition, several PKW have been built and have confirmed their reliability under flood conditions
