    One aspect of the present invention relates to a method of preparing a fibrous protein smectic hydrogel by way of a solvent templating process, comprising the steps of pouring an aqueous fibrous protein solution into a container comprising a solvent that is not miscible with water; sealing the container and allowing it to age at about room temperature; and collecting the resulting fibrous protein smectic hydrogel and allowing it to dry.     Quantitative Analysis of Lens Nuclear Density Using Optical Coherence Tomography (OCT) with a Liquid Optics Interface: Correlation between OCT Images and LOCS III Grading

    Purpose. To quantify whole lens and nuclear lens densities using anterior-segment optical coherence tomography (OCT) with a liquid optics interface and evaluate their correlation with Lens Opacities Classification System III (LOCS III) lens grading and corrected distance visual acuity (BCVA). Methods. OCT images of the whole lens and lens nucleus of eyes with age-related nuclear cataract were analyzed using ImageJ software. The lens grade and nuclear density were represented in pixel intensity units (PIU) and correlations between PIU, BCVA, and LOCS III were assessed. Results. Forty-seven eyes were analyzed. The mean whole lens and lens nuclear densities were 26.99 ± 5.23 and 19.43 ± 6.15 PIU, respectively. A positive linear correlation was observed between lens opacities (R2 = 0.187, p<0.01) and nuclear density (R2 = 0.316, p<0.01) obtained from OCT images and LOCS III. Preoperative BCVA and LOCS III were also positively correlated (R2 = 0.454, p<0.01). Conclusions. Whole lens and lens nuclear densities obtained from OCT correlated with LOCS III. Nuclear density showed a higher positive correlation with LOCS III than whole lens density. OCT with a liquid optics interface is a potential quantitative method for lens grading and can aid in monitoring and managing age-related cataracts

    A Study on Virtual Reality Storytelling by Story Authoring Tool Algorithm

    The objective of this study was to examine the storytelling principles of virtual reality contents, which are recently grabbing much attention, and the patterns of their generation rules and, based on the results, to analyze the elements and structure of a storytelling method suitable for virtual reality contents. In virtual reality environment, a story is usually being generated between choices made by a user who behaves autonomously under simulated environmental factors and the environmental constraints. This corresponds to a mutually complementary role of representation and simulation, which has been hotly discussed in the field of interactive storytelling. This study was conducted based on the assumption that such a mutually complementary realization is ideal for virtual reality storytelling. A simulation-based story authoring tool is a good example that shows this mutual complementation, in that it develops a story through various algorithms which involves the interaction of agents which occur within the strata of a virtual environment. Therefore, it can be a methodology for virtual reality storytelling. The structures and elements of narratives used in virtual reality storytelling which achieve balance of representation and simulation are much similar to an algorithm strategy of a simulation-based story authoring tool. The virtual reality contents released up to now can be classified into four categories based on the two axes of representation and simulation. The study focused on contents which are layered in higher strata of both representation and simulation. In the perspective of representation strata, these contents are actively using such elements as goal, event, action, perception, internal element, outcome, and setting element, which are constituents of ‘Fabula model’, to generate time relations and cause-effect relations. And in the perspective of simulation strata, the use of the ‘Late commitment’ strategy allowed users to understand the meanings of their actions taken during the process of experimenting with various dynamic principles within the environment

    Reduction of metabolic waste products, ammonia and lactate, through the coupling of GS selection and LDH-A down-regulation in CHO cells

    The cultivation of Chinese hamster ovary (CHO) cells for the production of therapeutic proteins inevitably accompanies the production of metabolic wastes, mostly ammonia and lactate. Ammonia alters cell growth, productivity and the glycosylation patterns of proteins, and lactate acidifies culture media, having negative effects on cell culture. A stable CHO cell line should be established for the manufacturing process of therapeutic proteins, and the development of stable cell lines is usually based on two expression systems: the dihydrofolate reductase (DHFR) system and the glutamine synthetase (GS) system. Compared to the DHFR system, the GS system produces a reduced level of ammonia because the GS protein uses ammonia to produce glutamine. In order to overcome the lactate accumulation problem, down-regulation of the lactate-producing enzyme, lactate dehydrogenase-A (LDH-A), has been shown to be effective. Engineering of the LDH-A gene has been applied for several CHO cell lines with the DHFR system, but there has been no trial which couples the ammonia reduction from the GS system and lactate reduction through cell engineering. In the present study, the GS system was used for the expression of therapeutic antibody in CHO cells, thereby reducing ammonia in the culture media. In addition, the LDH-A gene was down-regulated with shRNA to reduce lactate production. The antibody-producing cell line produces a reduced level of ammonia compared to the host cell line due to the over-expression of the GS protein. The down–regulation of the LDH-A gene in the antibody-producing cell line not only reduces the level of lactate but also further reduces the level of ammonia, accomplishing complete waste reduction. LDH-A down-regulation was also applied to the host cell lines of the GS system – the CHO-K1 cell line and the GS deficient CHO-K1 cell line. However, LDH-A down-regulated host cells could not survive the pool-selection process. Given that the GS system uses a glutamine-depleted condition as a form of selection pressure, enhanced glycolysis is inevitable and the down-regulation of LDH-A appears to hinder metabolic changes. Taken together, the application of LDH-A down-regulation in the producing cell line of the GS system successfully reduced both ammonia and lactate levels. However, LDH-A engineering could not be applied to the host cell lines because it inhibits the selection process of the GS system

    How do Investors in the Korean Stock Market React to Nuclear Threats from North Korea?

    This article examines how investors react to nuclear threats from North Korea on the South Korean stock market. To investigate the differences in investor groups responses, we divide investors into three groups: domestic individual, domestic institution and foreign investors. In addition, we classify the nuclear threats into the one actual threat and the seven verbal threats and compare trading activities of investors against those in the peaceful period of 2004. The net-buying and LSV(1992) herding measures are applied to examine investors trading behavior. Our results show that it was only for the case of the actual threat that investors trades significantly differed with the peaceful period. In addition, our results show that the three investor groups reacted to the actual nuclear threat differently. When the actual nuclear threat occurred, individuals sold, institutes and foreigners bought stocks. Like in the peaceful period, institutes showed a positive slope for lagged returns whereas individuals displayed a negative slope. Foreigners, however, bought regardless of previous returns. Moreover, LSV herding measure increased in all investor groups

    Retrospective Analysis of Peripheral Blood Stem Cell Transplantation for the Treatment of High-Risk Neuroblastoma

    Disease relapse after autologous peripheral blood stem cell transplantation (APBSCT) is the main cause of treatment failure in high-risk neuroblastoma (NBL). To reduce relapse, various efforts have been made such as CD34+ selection and double APBSCT. Here the authors reviewed the clinical features and outcomes of high-risk NBL patients and analyzed their survival. The medical records of 36 patients with stage III or IV NBL who underwent APBSCT at Seoul National University Children's Hospital between May 1996 and May 2004 were reviewed. Total 46 APBSCTs were performed in 36 patients. Disease free survival (DFS) and overall survival of all patients were 47.7% and 68.8%, respectively. The patients were allocated to three groups according to the APBSCT type. The DFS of CD34+ non-selected single APBSCT patients (N=13), CD34+ selected single APBSCT patients (N=14), and CD34+ selected double APBSCT patients (N=9) were 55.6%, 40.6%, and 50.0%, respectively, which were not significantly different. Thus the survival was not found to be affected by CD34+ selection or transplantation number. To improve long-term survival, various efforts should be made such as chemotherapy dose intensification, more effective tumor purging, and control of minimal residual disease via the use of differentiating and immune-modulating agents

    Use of Microalgae for Advanced Wastewater Treatment and Sustainable Bioenergy Generation

    Given that sustainable energy production and advanced wastewater treatment for producing clean water are two major challenges faced by modern society, microalgae make a desirable treatment alternative by providing a renewable biomass feedstock for biofuel production, while treating wastewater as a growth medium. Microalgae have been known to be resilient to the toxic contaminants of highly concentrated organic wastewater (e.g., organic nitrogen, phosphorus, and salinity) and are excellent at sorbing heavy metals and emerging contaminants. Economic and environmental advantages associated with massive algae culturing in wastewater constitute a driving force to promote its utilization as a feedstock for biofuels. However, there are still many challenges to be resolved which have impeded the development of algal biofuel technology at a commercial scale. This review provides an overview of an integrated approach using microalgae for wastewater treatment, CO2 utilization, and biofuel production. The main goal of this article is to promote research in algae technologies by outlining critical needs along the integrated process train, including cultivation, harvesting, and biofuel production. Various aspects associated with design challenges of microalgae production are described and current developments in algae cultivation and pretreatment of algal biomass for biofuel production are also discussed.
