    Functional characterization of olfactory receptors in three Dacini fruit flies (Diptera: Tephritidae) that respond to 1-nonanol analogs as components in the rectal glands

    Dacini fruit flies (Tephritidae: Diptera), including destructive pest species, are strongly affected in their reproductive behaviors by semiochemicals. Notably, male lures have been developed for pest management e.g., aromatic compounds for the Oriental fruit fly Bactrocera dorsalis and the melon fruit fly Zeugodacus cucurbitae; terpenic α-ionone analogs for the solanaceous fruit fly, B. latifrons. Other than those specific male attractants, 1-nonanol analogs have been noticed as major aliphatic components in the male rectal gland, which is considered as a secretory organ of male sex pheromones. Although multiple semiochemicals associated with the life cycle of Dacini fruit flies have been identified, their behavioral role(s) and chemosensory mechanisms by which the perception occurs have not been fully elucidated. In this study, we conducted RNA sequencing analysis of the chemosensory organs of B. latifrons and Z. cucurbitae to identify the genes coding for chemosensory receptors. Because the skeletons of male attractants are different among Dacini fruit fly species, we analyzed phylogenetic relationships of candidate olfactory receptors (ORs) among the three species. We found that the OR phylogeny reflects the taxonomic relationships of the three species. We further characterized functional properties of OR74a in the three Dacini species to the 1-nonanol analogs related to components in the rectal glands. The three OR74a homologs responded to 1-nonanol, but their sensitivities differed from each other. The OR74a homologs identified from B. dorsalis and Z. cucurbitae responded significantly to 6-oxo-1-nonanol, but not to 1, 3-nonanediol and nonyl acetate, indicating similar binding properties of the homologous ORs

    New Paradigms of Radiotherapy for Bone Metastasis

    Proper care of patients with bone metastasis requires interdisciplinary treatments. Radiotherapy (RT) plays a central role in the management of painful bone metastasis. External beam RT can provide rapid successful palliation of painful bone metastasis in 50–80% of patients, is associated with very few adverse effects and leads to complete pain relief at the treated site in up to one‐third of patients. Intensity‐modulated RT (IMRT) or stereotactic body RT (SBRT) enables the delivery of higher doses to the target tumor while minimizing the dose to adjacent organs. Reirradiation using IMRT or SBRT is a valuable option for the management of bone metastases. A multidisciplinary team, especially one consisting of a spinal surgeon and rehabilitation physician, is particularly useful for treating patients with spinal bone metastases characterized by spinal instability. Rehabilitation intervention which increases the physical activity level and prevents deconditioning is important. Future developments in surgical procedures and RT will likely improve the management protocols for bone metastases and technology to reduce metal artifacts in radiation planning might improve the efficacy and safety of combination therapy

    A nationwide, multi-center, retrospective study of symptomatic small bowel stricture in patients with Crohn\u27s disease.

    BACKGROUND:Small bowel stricture is one of the most common complications in patients with Crohn\u27s disease (CD). Endoscopic balloon dilatation (EBD) is a minimally invasive treatment intended to avoid surgery; however, whether EBD prevents subsequent surgery remains unclear. We aimed to reveal the factors contributing to surgery in patients with small bowel stricture and the factors associated with subsequent surgery after initial EBD.METHODS:Data were retrospectively collected from surgically untreated CD patients who developed symptomatic small bowel stricture after 2008 when the use of balloon-assisted enteroscopy and maintenance therapy with anti-tumor necrosis factor (TNF) became available.RESULTS:A total of 305 cases from 32 tertiary referral centers were enrolled. Cumulative surgery-free survival was 74.0% at 1 year, 54.4% at 5 years, and 44.3% at 10 years. The factors associated with avoiding surgery were non-stricturing, non-penetrating disease at onset, mild severity of symptoms, successful EBD, stricture length < 2 cm, and immunomodulator or anti-TNF added after onset of obstructive symptoms. In 95 cases with successful initial EBD, longer EBD interval was associated with lower risk of surgery. Receiver operating characteristic analysis revealed that an EBD interval of ≤ 446 days predicted subsequent surgery, and the proportion of smokers was significantly high in patients who required frequent dilatation.CONCLUSIONS:In CD patients with symptomatic small bowel stricture, addition of immunomodulator or anti-TNF and smoking cessation may improve the outcome of symptomatic small bowel stricture, by avoiding frequent EBD and subsequent surgery after initial EBD

In particular, studies of gene expression dynamics and regulation landscape in COVID-19 infected individuals are limited. Here, we report on a thorough analysis of whole blood RNA-seq data from 465 genotyped samples from the Japan COVID-19 Task Force, including 359 severe and 106 non-severe COVID-19 cases. We discover 1169 putative causal expression quantitative trait loci (eQTLs) including 34 possible colocalizations with biobank fine-mapping results of hematopoietic traits in a Japanese population, 1549 putative causal splice QTLs (sQTLs; e.g. two independent sQTLs at TOR1AIP1), as well as biologically interpretable trans-eQTL examples (e.g., REST and STING1), all fine-mapped at single variant resolution. We perform differential gene expression analysis to elucidate 198 genes with increased expression in severe COVID-19 cases and enriched for innate immune-related functions. Finally, we evaluate the limited but non-zero effect of COVID-19 phenotype on eQTL discovery, and highlight the presence of COVID-19 severity-interaction eQTLs (ieQTLs; e.g., CLEC4C and MYBL2). Our study provides a comprehensive catalog of whole blood regulatory variants in Japanese, as well as a reference for transcriptional landscapes in response to COVID-19 infection
