Pesquisa com CĂ©lulas-tronco: Mitos e Realidade
RESUMENDurante os Ăşltimos anos, diversos tipos de cĂ©lulas-tronco (CT) tĂŞm sido descobertas a partir das cĂ©lulas embrionárias e de adultos. Cada uma delas surge como promessa revolucionária no futuro da regeneração celular para tratar uma grande variedade de doenças. A pesquisa com estas cĂ©lulas vem recebendo um grande apoio das polĂticas cientĂficas na maioria dos paĂses, provocando um intenso debate Ă©tico e religioso e despertando um grande interesse do pĂşblico em geral. Entretanto, muito ainda há para ser investigado, atravĂ©s de pesquisas básicas e prĂ©-clinicas, antes (irar o de) que possamos acreditar que este tipo de terapia esteja totalmente pronto para ser empregado nas pesquisas clĂnicas. O desenvolvimento de linhagens de CT in vitro, para o entendimento dos fatores envolvidos no crescimento e diferenciação celular, bem como o desenvolvimento de protocolos prĂ©-clĂnicos, mostrando como e por que estas cĂ©lulas regeneram os tecidos in vivo, sĂŁo pontos fundamentais a serem levados em consideração. ABSTRACTDuring the past few years, several types of stem cells have been described, both embryonic and adult ones. Each one of them represents a revolutionary promise in the cellular regeneration field, for future treatment of many diseases. Research using stem cells is being greatly supported in most countries, provoking an intense ethical and religious debate among scientists as well as the general public. There is, however, a long road to go in basic research before we can safely and effectively use this technology for treating diseases. The in vitro development of stem cell lineages and pre-clinical protocols will allow us to understand more about how these cells grow and differentiate, as well as how and why these cells regenerate tissues. Key words: Stem cells, Cell theraph

Terapia Celular para o Tratamento de Cardiopatias Isquêmicas: Desenvolvimento de um Protocolo Pré-clinico
RESUMEN Apesar dos avanços clĂnicos, cirĂşrgicos e farmacolĂłgicos, quando a doença cardĂaca isquĂŞmica Ă© muita avançada e difusa, os mĂ©todos atualmente disponĂveis nĂŁo sĂŁo totalmente efetivos para evitar a perda de cardiomiĂłcitos funcionais. Isso acarretará alterações na geometria ventricular (remodelamento), com perda progressiva da função ventricular e desenvolvimento de insuficiencia cardĂaca. Recentes avanços na compreensĂŁo da biologia das cĂ©lulas-tronco reinstalaram o conceito de transplante celular para substituir cĂ©lulas nĂŁo funcionais. A regeneração do miocárdio tem despertado um crescente interesse visando atender este propĂłsito. A proposta deste trabalho Ă© o de desenvolver um protocolo prĂ©-clĂnico seguro e eficiente que possa contribuir para uma melhor compreensĂŁo dos mecanismos envolvidos no processo de regeneração do mĂşsculo cardĂaco isquĂŞmico, atravĂ©s da terapia celular, associada ou nĂŁo a terapia gĂŞnica, com conseqĂĽente recuperação da função cardĂaca, em ratos infartados (modeloIAM).  ABSTRACT In spite of clinical, surgical and pharmacological advances, the currently available methods are not totally effective in treating advanced stage ischemic cardiopathy, permitting functional cardiomiocyte loss which leads to ventricle remodeling, progressive loss of ventricle function and the development of cardiac insufficiency. Recent advances in understanding stem cell biology have established the concept of cellular transplant to substitute non-functional cells. Myocardium regeneration is being greatly considered for such procedure. Our main goal is the development of a safe and effective pre-clinical protocol, which could contribute to clarify the mechanisms involved in myocardium regeneration using cellular therapy, combined or not with gene therapy, leading to cardiac function restoration in rats (IAM model).  Keywords: Stem Cell, myocardium regeneration, cell therapy

Genetic Structure and Inferences on Potential Source Areas for Bactrocera dorsalis (Hendel) Based on Mitochondrial and Microsatellite Markers
Bactrocera dorsalis (Diptera: Tephritidae) is mainly distributed in tropical and subtropical Asia and in the Pacific region. Despite its economic importance, very few studies have addressed the question of the wide genetic structure and potential source area of this species. This pilot study attempts to infer the native region of this pest and its colonization pathways in Asia. Combining mitochondrial and microsatellite markers, we evaluated the level of genetic diversity, genetic structure, and the gene flow among fly populations collected across Southeast Asia and China. A complex and significant genetic structure corresponding to the geographic pattern was found with both types of molecular markers. However, the genetic structure found was rather weak in both cases, and no pattern of isolation by distance was identified. Multiple long-distance dispersal events and miscellaneous host selection by this species may explain the results. These complex patterns may have been influenced by human-mediated transportation of the pest from one area to another and the complex topography of the study region. For both mitochondrial and microsatellite data, no signs of bottleneck or founder events could be identified. Nonetheless, maximal genetic diversity was observed in Myanmar, Vietnam and Guangdong (China) and asymmetric migration patterns were found. These results provide indirect evidence that the tropical regions of Southeast Asia and southern coast of China may be considered as the native range of the species and the population expansion is northward. Yunnan (China) is a contact zone that has been colonized from different sources. Regions along the southern coast of Vietnam and China probably served to colonize mainly the southern region of China. Southern coastal regions of China may also have colonized central parts of China and of central Yunnan
