Genetic dissection of the biochemical activities of human DNA repair protein, APE1

Human apurinic/apyrimidinic endonuclease 1 (APE1) is a key DNA repair enzyme involved in both base excision repair (BER) and nucleotide incision repair (NIR) pathways. In the BER pathway, APE1 cleaves DNA at AP sites and 3'-blocking moieties generated by DNA glycosylases. In the NIR pathway, APE1 incises DNA 5' to a number of oxidatively damaged bases. Here we propose to identify and characterize critical amino acids of APE1 involved in either BER and/or NIR functions by using the alignment of the known three-dimensional (or tertiary) structures of Xth family AP endonucleases including the Methanothermobacter thermautotrophicus Mth212, Bacillus subtilis ExoA (1), E. coli Xth and human APE1 proteins (2)

The recombinant fusion protein CFP10–ESAT6–dIFN has protective effect against tuberculosis in guinea pigs

Development of effective vaccine candidates against tuberculosis (TB) is currently the most important challenge in the prevention of this disease since the BCG vaccine fails to guarantee a lifelong protection, while any other approved vaccine with better efficiency is still absent. The protective effect of the recombinant fusion protein CFP10–ESAT6–dIFN produced in a prokaryotic expression system (Escherichia coli) has been assessed in a guinea pig model of acute TB. The tested antigen comprises the Mycobacterium tuberculosis (Mtb) proteins ESAT6 and CFP10 as well as modified human γ-interferon (dIFN) for boosting the immune response. Double intradermal immunization of guinea pigs with the tested fusion protein (2 × 0.5 µg) induces a protective effect against subsequent Mtb infection. The immunized guinea pigs do not develop the symptoms of acute TB and their body weight gain was five times more as compared with the non-immunized infected guinea pigs. The animal group immunized with this dose of antigen displays the minimum morphological changes in the internal organs and insignificant inflammatory lesions in the liver tissue, which complies with a decrease in the bacterial load in the spleen and average Mtb counts in macrophages

The phenomenon of «half-decay of competence» investigated in the work causes an objective need for the formation of the concept of a studying organization. A model for the inclusion of higher education institutions in the process of continuous education of workers in various spheres of professional activity is proposed.

Reproduction and use of labor resources in agricultural region

The reproduction and rational use of rural labor resources directly influences the economic growth in the region, therefore, objective analysis and assessment of human resources, as well as of the changes in the municipal areas of the region and in the agricultural industry as a guarantor of food security is an urgent task. The proposed method of study aims the perform comprehensive analysis of developments in this field as processes at the regional level, and across municipalities and agribusiness organizations. A survey of labor resources studies the dynamics of the distribution of organizational and legal forms, the level of vocational training as one of the main factors ensuring the development of the human potential of the territory, and looks at these aspects within the context of related industries (agriculture, processing of agricultural products, service agriculture, rural construction etc.) Based on the methodology a study of the reproduction and use of the agricultural labor resources of Orenburg region, which has over 40% of the rural population, was conducted. It was found that the structure of organizational and legal forms in 2008-2013 in the field of agribusiness changed significantly. The total number of companies increased by a third mainly due to the increase in the number of limited liability companies, the share of agricultural enterprises decreased, the level of vocational education improved and the number of employees with higher and secondary vocational degrees in agriculture increased. Wages in agricultural production is still substantially lower than in other sectors of the regional economy. A sharp reduction in the labor resources has been observed n organizations of the agricultural industry in rural districts, which will undoubtedly lead to their destruction. What is happening is not helping the policy of agriculture development in general and import substitution adopted in this country
