Effect of trace metal Ions on the conformational stability of the visual photoreceptor rhodopsin
Trace metals are essential elements that play key roles in a number of biochemical processes governing human visual physiology in health and disease. Several trace metals, such as zinc, have been shown to play important roles in the visual phototransduction process. In spite of this, there has been little research conducted on the direct effect of trace metal elements on the visual photoreceptor rhodopsin. In the current study, we have determined the effect of several metal ions, such as iron, copper, chromium, manganese, and nickel, on the conformational stability of rhodopsin. To this aim, we analyzed, by means of UV-visible and fluorescence spectroscopic methods, the effects of these trace elements on the thermal stability of dark rhodopsin, the stability of its active Metarhodopsin II conformation, and its chromophore regeneration. Our results show that copper prevented rhodopsin regeneration and slowed down the retinal release process after illumination. In turn, Fe3+, but not Fe2+, increased the thermal stability of the dark inactive conformation of rhodopsin, whereas copper ions markedly decreased it. These findings stress the important role of trace metals in retinal physiology at the photoreceptor level and may be useful for the development of novel therapeutic strategies to treat retinal disease.This research was funded by grants from Ministry of Science and Innovation, Spain (PID2019-104817GB-I00), and from the Government of Catalonia to Research Consolidated Groups (2021 SGR 00342) to P.G. Human blue cone opsin regeneration involves secondary retinal binding with analog specificity
Human color vision is mediated by the red, green, and blue cone visual pigments. Cone opsins are G-protein-coupled receptors consisting of an opsin apoprotein covalently linked to the 11-cis-retinal chromophore. All visual pigments share a common evolutionary origin, and red and green cone opsins exhibit a higher homology, whereas blue cone opsin shows more resemblance to the dim light receptor rhodopsin. Here we show that chromophore regeneration in photoactivated blue cone opsin exhibits intermediate transient conformations and a secondary retinoid binding event with slower binding kinetics. We also detected a fine-tuning of the conformational change in the photoactivated blue cone opsin binding site that alters the retinal isomer binding specificity. Furthermore, the molecular models of active and inactive blue cone opsins show specific molecular interactions in the retinal binding site that are not present in other opsins. These findings highlight the differential conformational versatility of human cone opsin pigments in the chromophore regeneration process, particularly compared to rhodopsin, and point to relevant functional, unexpected roles other than spectral tuning for the cone visual pigmentsPeer ReviewedPostprint (author's final draft
Material per a l'avaluació d'assignatures de química
El treball que es presenta forma part del Projecte Elaboració de material per a l'Avaluació d'assignatures de Química, de la convocatòria d'ajuts per a projectes de Millora de la docència 2009-2010 de la Universitat Politècnica de Catalunya. Els autors pertanyen als Grups GReDiQ i GRAPA integrats al projecte RIMA de l’Institut de Ciències de l’Educació de la UPC.
S’ha elaborat un banc de preguntes i problemes, accessible a tot el professorat de la UPC que imparteix assignatures de Química i que podrà ser utilitzat per posar en pràctica l’avaluació continuada de l’aprenentatge: autoavaluació formativa i avaluació sumativa. Hi ha qüestionaris Moodle referents a la utilització del material multimèdia generat en un projecte anterior (“Qüestionaris de laboratori”) i d’altres de problemes (“Qüestionaris de problemes”). Es pretén valorar la repercussió del material multimèdia en l’aprenentatge i l’assoliment dels objectius formatius.
Els qüestionaris de laboratori s’han utilitzat com part del pre-laboratori de l’alumnat de Experimentació en Química II (ETSEIB), Experimentació en Química (ETSEIB), Química (EET) i Anàlisi Química (EET). Els qüestionaris de problemes s’han utilitzat en les assignatures de Química i Anàlisi Química de l’EET i s’estan implementant aquest quadrimestre a la FOOT (a Química per a ciències de la visió) tant a la modalitat presencial com semipresencial.
L’avaluació dels primers resultats es va realitzar durant el primer quadrimestre del curs 2011-12 mitjançant una enquesta de satisfacció que es va passar a l’EET. L’alumnat de l’EET utilitza majoritàriament els qüestionaris i els troben útils per tal de valorar el seu progrés a l’assignatura.Peer Reviewe
GREDIQ-RIMA: The evolution of a teaching project of experimentation in chemistry
Authors of this project belong to the Resources Chemistry Teaching Group (GReDiQ) involved in RIMA (Research and innovation in learning methodologies). The audiovisual and multimedia material productions realized have been grouped into three themes: Basic Techniques of Experimentation in Chemistry, Safety in Chemistry Laboratories and Advanced Techniques of Experimentation in Chemistry. Material docent en format digital per a assignatures d'experimentació en química
En aquesta comunicació es presenta un projecte elaborat arran de l’interès mostrat per un grup de professorat en disposar de material audiovisual propi i comú per a assignatures experimentals de química de diferents titulacions impartides per diferents centres de la Universitat Politècnica de Catalunya (UPC). Els autors d’aquest treball pertanyen al Grup de Recursos per a la Didàctica de la Química (GReDiQ), integrat en el projecte RIMA (http://www.upc.edu/rima) de l’Institut de Ciències de l’Educació de la UPC. L’elaboració de material audiovisual digital constitueix un dels objectius principals plantejats pel GReDiQ i pretén generar tant un material de suport per a les activitats pre-laboratorio, com un element motivador i que faciliti l’aprenentatge de l’alumnat en les sessions de laboratori de les matèries de Química i Enginyeria química, que apareixen com assignatures de formació bàsica en gairebé tots els plans d’estudi dels nous Graus d’Enginyeria que s’ estan implantant a la UPC. La utilització del material audiovisual digital possibilita que l’alumnat prengui consciència de les activitats de laboratori, així com dels hàbits bàsics de manipulació i seguretat en el laboratori, del fonament d’operacions bàsiques o instrumentació a utilitzar.Peer Reviewe
