    Epizootiological and clinical significance of stephanurus dentatus nematode in swine

    Stephanuriosis is a disease of the urinary tract of both domestic pigs and wild boars occurring in tropical and subtropical regions, with a tendency to spread to other geographical areas. In endemic areas, the disease occurs more often in extensively bred herds, normally with nonspecific clinical picture. As a result of larval stage migrations and parasitism of adult forms of nematode Stephanurus dentatus, the damage occurs in the liver, kidneys, ureters, lungs, pancreas and perirenal adipose tissue. Damage to the liver is reminiscent of lesions caused by Ascaris suum, which is of particular clinical significance for differential diagnosis of parasitic swine diseases. Stephanurus dentatus is of great economic importance due to direct and indirect damage it causes. Infected animals delay in reaching market weight, their meat is declared unfit for consumption, and they are sent to forced slaughter. Examinations of potential presence of this parasite in Serbia should include populations of wild boars and domestic pigs from extensive traditional breeding outdoors. After intense climate changes that have occurred in recent decades, and also due to the possibility of importing infected animals from endemic areas, the proposed research would have both epizootiological and clinical significance. © 2022 Authors. U endemskim oblastima, oboljenje se češće javlja u zapatima koji se gaje ekstenzivno i protiče sa nespecifi čnom kliničkom slikom. Kao posledica migracije larvenih stadijuma i parazitiranja odraslih oblika nematode Stephanurus dentatus, nastaju oštećenja na jetri, bubrezima, ureterima, plućima, pankreasu i perirenalnom masnom tkivu. Oštećenja koja nastaju u jetri podsećaju na lezije uzrokovane larvama Ascaris suum, što je od posebnog kliničkog značaja za diferencijalnu dijagnozu parazitskih oboljenja svinja. Zbog direktnih i indirektnih šteta koje prouzrokuje, S. dentatus ima veliki ekonomski značaj. Infi cirane životinje kasne u postizanju tržišne težine, njihovo meso se proglašava neupotrebljivim i upućuju se na prinudno klanje. Ispitivanja eventualnog prisustva ovog parazita u Srbiji trebalo bi da obuhvate populaciju divljih svinja i domaće svinje iz ekstenzivnog tradicionalnog gajenja na otvorenim površinama.     Mystery called sarcoidosis: Forty-four years follow-up of chronic systemic disease

    Introduction. This is a presentation of a 61-year-old female patient. Since 44 years have passed from the onset of her first symptoms until the final diagnosis of sarcoidosis, this was the reason of our decision to publish the case. Case Outline. During the follow-up period of 44 years the patient had ocassional polymorphic complains, such as adynamia, nausea, abdominal pains, myalgia, arthralgia, body weight loss (8-10 kg) etc. The clinical course was predominated by splenomegaly, hepatitis and arthralgia, and later chronic renal failure also developed. Laboratory findings showed elevated markers of acute inflammation and autoantibodies. The patient was hospitalized in different university internal hospitals (gastroenterology, allergology, rheumatology, nephrology and pulmology). Liver biopsy was performed three times, rectum and kidney biopsy once each and finally bronchoscopy and pulmonary biopsy was done. At last, about 40 years from the onset of the first symptoms, in 2006 the diagnosis of lung sarcoidosis was established. Conclusion. The final diagnosis of spleen sarcoidosis was confirmed by pathologically verified sarcoidosis of the lungs. This case is particularly interesting because of the presence of familial sarcoidosis (the patient’s son also had recurrent pulmonary sarcoidosis).     Novel Patched 1 mutations in patients with nevoid basal cell carcinoma syndrome - case report

    Nevoid basal cell carcinoma syndrome (Gorlin syndrome) is a rare autosomal dominant disorder characterized by numerous basal cell carcinomas, keratocystic odontogenic tumors of the jaws, and diverse developmental defects. This disorder is associated with mutations in tumor suppressor gene Patched 1 (PTCH1). We present two patients with Gorlin syndrome, one sporadic and one familial. Clinical examination, radiological and CT imaging, and mutation screening of PTCH1 gene were performed. Family members, as well as eleven healthy controls were included in the study. Both patients fulfilled the specific criteria for diagnosis of Gorlin syndrome. Molecular analysis of the first patient showed a novel frameshift mutation in exon 6 of PTCH1gene (c.903delT). Additionally, a somatic frameshift mutation in exon 21 (c.3524delT) along with germline mutation in exon 6 was detected in tumor-derived tissue sample of this patient. Analysis of the second patient, as well as two affected family members, revealed a novel nonsense germline mutation in exon 8 (c.1148 C gt A)

    Relevance of TNF-alpha, IL-6 and IRAK1 gene expression for assessing disease severity and therapy effects in tuberculosis patients

    Introduction: Tuberculosis (TBC) is a contagious chronic respiratory disease which despite the known cause, Mycobacterium tuberculosis (Mtb), and many decades of successful therapy, remains one of the leading global health problems. Immune responses against Mtb infection involve both of types of immunity, but cellular immunity, in which certain cytokines and Th1 cells play a key role, is crucial. A better understanding of the functions of the cytokine network involved in the state and progression of TBC could identify specific molecular markers for monitoring of disease activity as well as therapy outcomes in TBC patients. Methodology: We investigated expression of TNF-alpha, IL-6 and IRAK1 genes using an RT-qPCR technique in peripheral blood mononuclear cells of 33 TBC patients and 10 healthy individuals. Results: Comparison between TBC patients and healthy individuals revealed statistically significant differences for all analyzed genes. The levels of expression of TNF-alpha and IL-6 mRNA were higher, while the level of IRAK1 mRNA was lower in the TBC group compared to controls. Moreover, a strong positive correlation was observed between TNF-alpha and IL-6 gene expression. When clinical parameters were analyzed, increased levels of TNF-alpha mRNA were detected in patients with a longer duration of therapy ( gt 2 months) compared to those with a shorter therapy duration ( lt 2 months), and in patients without anemia. Conclusions: Our results indicate that the inflammatory genes we examined play a crucial role in the pathogenesis of tuberculosis, and that the expression of the TNF-alpha gene could be a marker for monitoring the clinical effect of the ant-tuberculosis drugs during therapy

    Oslikavanje eritrocita bez obeležavanja pomoću konfokalne ili epifluorescentne mikroskopije je praktično nemoguće jer se fluorescencija od hemoglobina (Hb) ne može detektovati zbog brzih neradijativnih raspada koji su dominantni u odnosu na spontanu emisiju. Sa druge strane, demonstrirana je značajna dvofotonska apsorptivnost Hb u infracrvenoj oblasti1 kao i dvofotonski pobuđena fluorescencija (TPEF) od novoformiranog fotoprodukta2 nakon interakcije ultrakratkih laserskih impulsa sa Hb. TPEF mikroskopija je korišćena u brojnim studijama za oslikavanje Hb i eritrocita bez obeležavanja.3 U ovom radu prikazane su fotofizičke osobine i potencijalne primene novoformiranog fotoprodukta. Kreirane su fluorescentne mikrostrukture pomoću direktnog laserskog upisivanja na tankim Hb slojevima (micropatterning). Fotoprodukt je okarakterisan pomoću različitih (mikro)spektroskopskih tehnika: emisije nakon vofotonske apsorpcije, UV-VIS apsorpcije i spektralnog oslikavanja. Razmatrane su spektroskopske osobine protoporfirina IX (PpIX) pošto je on strukturni deo Hb i potencijalno može biti prekursor za formiranje fotoprodukta. Takođe, Hb je tretiran vodonik peroksidom da bi se uočila optička i spektroskopska sličnost tako dobijenog jedinjenja i fotoprodukta. Potvrđena je dugovremena fotostabilnost fotoprodukta u periodima od nekoliko meseci. Na osnovu toga što je fotoprodukt moguće dobiti na prostorno i vremenski kontrolisan način, izvršeno je fotoobeležavanje i praćenje kretanja pojedinačnih eritrocita u humanoj punoj krvi zdravog donora

    Puncture site bleeding complications in patients with Clopidogrel hyper-response: Three case reports

    Dual antiplatelet therapy (clopidogrel and acetylsalicylic acid) is a standard for the embolization of planned intracranial aneurysms with CNS stent due to the possibility of stent thrombus formation. All anti-aggregation drugs, including those listed, have bleeding as a side effect. Three patients with aneurysm had an elevated response to antiplatelet therapy with clopidogrel, which was confirmed by a multiplate test on the "VerifyNow" system. After reducing the dose of clopidogrel or after interrupting it, with the introduction of low molecular weight heparin for the duration of five days, aneurysms were successfully resolved by intracranial implantation of the stent. Perioperative angiograms and postoperative CT angiograms have verified hematomas at the place of punction of the femoral artery. Bleeding was resolved by the femoral artery suture by a vascular surgeon. All patients were discharged home without further complications and with dual antiplatelet therapy. By measuring the platelet function in vitro, the degree of inhibition of platelet activity achieved by the action of the drug can be assessed. A specific test can identify those patients who are highly responsive to the drug with increased platelet reactivity and the possibility of increased risk of bleeding. Our suggestion is to reduce the dosage of clopidogrel or to leave it out for 24 hours with preventive doses of low molecular weight heparin or to change the strategy of treatment of intracranial aneurysm, i.e. avoiding implantation of CNS stent

Потврђена је дуговремена фотостабилност фотопродукта у периодима од неколико месеци.     DISTRIBUTION OF SOME EUROPEAN LEPIDOPTERA BASED ON THE FINDINGS OF THEIR NON-ADULT STAGES PRESENTED THROUGH TROPHIC ASSOCIATIONS AND A QUANTITATIVE ANALYSIS OF THEIR PARASITOIDS

    We examined 638 Lepidoptera specimens on the territories of 13 European countries in our search for parasitoids. We collected eggs, larvae and pupae. In total, 251 Lepidoptera species were identified, belonging to 169 genera from 30 families. Of the total sample, approximately one-third (32.23%) were parasitized. In 168 samples (26.42%), we identified only one parasitoid species per host. In addition to these data, 224 plant species from 114 genera were identified, of which the vast majority were feeding plants

    Interactions of ultrashort laser pulses with hemoglobin: Photophysical aspects and potential applications

    Hemoglobin (Hb), a life-sustaining and highly abundant erythrocyte protein, is not readily fluorescent. A few studies have already reported Two-Photon Excited Fluorescence (TPEF) of Hb, however, the mechanisms through which Hb becomes fluorescent upon interaction with ultrashort laser pulses are not completely understood. Here, we characterized photophysically this interaction on Hb thin film and erythrocytes using fluorescence spectroscopy upon single-photon/two-photon absorption, and UV-VIS single-photon absorption spectroscopy. A gradual increase of the fluorescence intensity, ending up with saturation, is observed upon prolonged exposure of Hb thin layer and erythrocytes to ultrashort laser pulses at 730 nm. When compared to protoporphyrin IX (PpIX) and oxidized Hb by H2O2, TPEF spectra from a thin Hb film and erythrocytes showed good mutual agreement, broad peaking at 550 nm, supporting hemoglobin undergoes degradation and that same fluorescent specie(s) originating from the heme moiety are generated. The uniform square shaped patterns of the fluorescent photoproduct exhibited the same level of the fluorescence intensity even after 12 weeks from the formation, indicating high photoproduct stability. We finally demonstrated the full potential of the formed Hb photoproduct with TPEF scanning microscopy towards spatiotemporally controlled micropatterning in HTF and single human erythrocyte labelling and tracking in the whole blood
