    Cytokine-Induced Monocyte Characteristics in SLE

    Monocytes in SLE have been described as having aberrant behavior in a number of assays. We examined gene expression and used a genome-wide approach to study the posttranslational histone mark, H4 acetylation, to examine epigenetic changes in SLE monocytes. We compared SLE monocyte gene expression and H4 acetylation with three types of cytokine-treated monocytes to understand which cytokine effects predominated in SLE monocytes. We found that γ-interferon and α-interferon both replicated a broad range of the gene expression changes seen in SLE monocytes. H4 acetylation in SLE monocytes was overall higher than in controls and there was less correlation of H4ac with cytokine-treated cells than when gene expression was compared. A set of chemokine genes had downregulated expression and H4ac. Therefore, there are significant clusters of aberrantly expressed genes in SLE which are strongly associated with altered H4ac, suggesting that these cells have experienced durable changes to their epigenome

    Minimum Rank of Graphs with Loops

    A loop graph S is a finite undirected graph that allows loops but does not allow multiple edges. The set S(S) of real symmetric matrices associated with a loop graph of order n is the set of symmetric matrices A = [a(ij)] is an element of R-nxn such that a(ij) not equal 0 if and only if ij is an element of E(S). The minimum (maximum) rank of a loop graph is the minimum (maximum) of t he ranks of the matrices in S(S). Loop graphs having minimum rank at most two are characterized (by forbidden induced subgraphs and graph complements) and loop graphs having minimum rank equal to the order of the graph are characterized. A Schur complement reduction technique is used to determine the minimum ranks of cycles with various loop configurations; the minimum ranks of complete graphs and paths with various configurations of loops are also determined. Unlike simple graphs, loop graphs can have maximum rank less than the order of the graph. Some results are presented on maximum rank and which ranks between minimum and maximum can be realized.     Dendritic cell-bound IgE functions to restrain allergic inflammation at mucosal sites

    Antigen-mediated crosslinking of Immunoglobulin E (IgE) bound to mast cells/basophils via FcεRI, the high affinity IgE Fc-receptor, is a well-known trigger of allergy. In humans, but not mice, dendritic cells (DCs) also express FcεRI that is constitutively occupied with IgE. In contrast to mast cells/basophils, the consequences of IgE/FcεRI signals for DC function remain poorly understood. We show that humanized mice that express FcεRI on DCs carry IgE like non-allergic humans and do not develop spontaneous allergies. Antigen-specific IgE/FcεRI crosslinking fails to induce maturation or production of inflammatory mediators in human DCs and FcεRI-humanized DCs. Furthermore, conferring expression of FcεRI to DCs decreases the severity of food allergy and asthma in disease-relevant models suggesting anti-inflammatory IgE/FcεRI signals. Consistent with the improved clinical parameters in vivo, antigen-specific IgE/FcεRI crosslinking on papain or LPS-stimulated DCs inhibits the production of pro-inflammatory cytokines and chemokines. Migration assays confirm that the IgE-dependent decrease in cytokine production results in diminished recruitment of mast cell progenitors; providing a mechanistic explanation for the reduced mast cell-dependent allergic phenotype observed in FcεRI-humanized mice. Our study demonstrates a novel immune regulatory function of IgE and proposes that DC-intrinsic IgE signals serve as a feedback mechanism to restrain allergic tissue inflammation
