    Exogenous Amino Acids Regulate Trophectoderm Differentiation in the Mouse Blastocyst through an mTOR-Dependent Pathway

    AbstractAt the late blastocyst stage, the epithelial trophectoderm cells of the mammalian embryo undergo a phenotypic change that allows them to invade into the uterine stroma and make contact with the maternal circulation. This step can be regulated in vitro by the availability of amino acids. Embryos cultured in defined medium lacking amino acids cannot form trophoblast cell outgrowths on fibronectin, an in vitro model of implantation, but remain viable for up to 3 days in culture and will form outgrowths when transferred into complete medium. The amino acid requirement is a developmentally regulated permissive event that occurs during a 4- to 8-h period at the early blastocyst stage. Amino acids affect spreading competence specifically by regulating the onset of protrusive activity and not the onset of integrin activation. Rapamycin, a specific inhibitor of the kinase mTOR/FRAP/RAFT1, blocks amino acid stimulation of embryo outgrowth, demonstrating that mTOR is required for the initiation of trophectoderm protrusive activity. Inhibition of global protein translation with cycloheximide also inhibits amino acid-dependent signals, suggesting that mTOR regulates the translation of proteins required for trophoblast differentiation. Our data suggest that mTOR activity has a developmental regulatory function in trophectoderm differentiation that may serve to coordinate embryo and uterus at the time of implantation

    The activation of contextually predictable words in syntactically illegal positions

    We present an eye-tracking study testing a hypothesis emerging from several theories of prediction during language processing, whereby predictable words should be skipped more than unpredictable words even in syntactically illegal positions. Participants read sentences in which a target word became predictable by a certain point (e.g. "bone" is 92% predictable given "The dog buried his…"), with the next word actually being an intensifier (e.g. "really"), which a noun cannot follow. The target noun remained predictable to appear later in the sentence. We used the boundary paradigm (Rayner, 1975) to present the predictable noun or an alternative unpredictable noun (e.g. "food") directly after the intensifier, until participants moved beyond the intensifier, at which point the noun changed to a syntactically legal word. Participants also read sentences in which predictable or unpredictable nouns appeared in syntactically legal positions. A Bayesian linear mixed model suggested a 5.7% predictability effect on skipping of nouns in syntactically legal positions, and a 3.1% predictability effect on skipping of nouns in illegal positions.     REtools: A laboratory program for restriction enzyme work: enzyme selection and reaction condition assistance

    BACKGROUND: Restriction enzymes are one of the everyday tools used in molecular biology. The continuously expanding panel of known restriction enzymes (several thousands) renders their optimal use virtually impossible without computerized assistance. Several manufacturers propose on-line sites that assist scientists in their restriction enzyme work, however, none of these sites meet all the actual needs of laboratory workers, and they do not take into account the enzymes actually present in one's own laboratory. RESULTS: Using FileMaker Pro, we developed a stand-alone application which can run on both PCs and Macintoshes. We called it REtools, for Restriction Enzyme tools. This program, which references all currently known enzymes (>3500), permits the creation and update of a personalized list of restriction enzymes actually available in one's own laboratory. Upon opening the program, scientists will be presented with a user friendly interface that will direct them to different menus, each one corresponding to different situations that restriction enzyme users commonly encounter. We particularly emphasized the ease of use to make REtools a solution that laboratory members would actually want to use. CONCLUSION: REtools, a user friendly and easily customized program to organize any laboratory enzyme stock, brings a software solution that will make restriction enzyme use and reaction condition determination straightforward and efficient. The usually unexplored potential of isoschizomers also becomes accessible to all, since REtools proposes all possible enzymes similar to the one(s) chosen by the user. Finally, many of the commonly overlooked subtleties of restriction enzyme work, such as methylation requirement, unusual reaction conditions, or the number of flanking bases required for cleavage, are automatically provided by REtools

    Torque magnetometry study of the spin reorientation transition and temperature-dependent magnetocrystalline anisotropy in NdCo5

    We present the results of torque magnetometry and magnetic susceptibility measurements to study in detail the spin reorientation transition (SRT) and magnetic anisotropy in the permanent magnet NdCo5. We further show simulations of the measurements using first-principles calculations based on density-functional theory and the disordered local moment picture of magnetism at finite temperatures. The good agreement between theory and experimental data leads to a detailed description of the physics underpinning the SRT. In particular we are able to resolve the magnetization of, and to reveal a canting between, the Nd and Co sublattices. The torque measurements carried out in the ac and ab planes near the easy direction allow us to estimate the anisotropy constants, K 1, K 2 and K 4 and their temperature dependences. Torque curves, τ(γ) recorded by varying the direction of a constant magnetic field in the crystallographic ac plane show a reversal in the polarity as the temperature is changed across the SRT (240 < T < 285 K). Within this domain, τ(γ) exhibits unusual features different to those observed above and below the transition. The single crystals of NdCo5 were grown using the optical floating zone technique

    Measurement of CH3_3D on Titan at Submillimeter Wavelengths

    We present the first radio/submillimeter detection of monodeuterated methane (CH3_3D) in Titan's atmosphere, using archival data from of the Atacama Large Millimeter/submillimeter Array (ALMA). The JK=21−11J_K=2_1-1_1 and JK=20−10J_K=2_0-1_0 transitions at 465.235 and 465.250 GHz (∼0.644\sim0.644 mm) were measured at significance levels of 4.6σ4.6\sigma and 5.7σ5.7\sigma, respectively. These two lines were modeled using the Non-linear optimal Estimator for MultivariatE spectral analySIS (NEMESIS) radiative transfer code to determine the disk-averaged CH3_3D volume mixing ratio = 6.157×10−66.157\times10^{-6} in Titan's stratosphere (at altitudes >130\gt130 km). By comparison with the CH4_4 vertical abundance profile measured by Cassini-Huygens mass spectrometry, the resulting value for D/H in CH4_4 is (1.033±0.081)×10−4(1.033\pm0.081)\times10^{-4}. This is consistent with previous ground-based and in-situ measurements from the Cassini-Huygens mission, though slightly lower than the average of the previous values. Additional CH3_3D observations at higher spatial resolution will be required to determine a value truly comparable with the Cassini-Huygens CH4_4 measurements, by measuring CH3_3D with ALMA close to Titan's equator. In the post-Cassini era, spatially resolved observations of CH3_3D with ALMA will enable the latitudinal distribution of methane to be determined, making this an important molecule for further studies.Comment: 9 pages, 4 figure

    Tunability of the spin reorientation transitions with pressure in NdCo5

    We present pressure-dependent magnetization measurements carried out in the domain of the spin reorientation transitions (SRTs) of a NdCo5 single crystal. The application of a hydrostatic pressure leads to a shift in the SRTs to higher temperatures. This shift is found to be very sensitive to pressure, with the SRT temperatures increasing at a rate of ≈17 K/GPa. To explain the experimental results, we have also performed first-principles calculations of the SRT temperatures for different applied strains, which corroborate the experimental findings. The calculations attribute the pressure dependence of the SRTs to a faster weakening of the Co contribution to the magnetocrystalline anisotropy with pressure compared to the Nd contribution

    Capitalization interacts with syntactic complexity

    We investigated whether readers use the low-level cue of proper noun capitalization in the parafovea to infer syntactic category, and whether this results in an early update of the representation of a sentence’s syntactic structure. Participants read sentences containing either a subject relative or object relative clause, in which the relative clause’s overt argument was a proper noun (e.g., The tall lanky guard who alerted Charlie/Charlie alerted to the danger was young) across three experiments. In Experiment 1 these sentences were presented in normal sentence casing or entirely in upper case. In Experiment 2 participants received either valid or invalid parafoveal previews of the relative clause. In Experiment 3 participants viewed relative clauses in only normal conditions. We hypothesized that we would observe relative clause effects (i.e., inflated fixation times for object relative clauses) while readers were still fixated on the word who, if readers use capitalization to infer a parafoveal word’s syntactic class. This would constitute a syntactic parafoveal-on-foveal effect. Furthermore, we hypothesised that this effect should be influenced by sentence casing in Experiment 1 (with no cue for syntactic category being available in upper case sentences) but not by parafoveal preview validity of the target words. We observed syntactic parafoveal-on-foveal effects in Experiment 1 and 3, and a Bayesian analysis of the combined data from all three experiments. These effects seemed to be influenced more by noun capitalization than lexical processing. We discuss our findings in relation to models of eye movement control and sentence processing theories
