Effects of Myristoylation on the Structure and Stability of Hisactophilin
Hisactophilin is a histidine rich, actin binding protein from Dictyostelium discoideum. The structure of Hisactophilin, an all-beta protein, is typical of the beta-trefoil superfamily of proteins. In addition, hisactophilin is myristoylated, which is a relatively common co-translational modification involving the covalent attachment of a myristic acid group to the N-terminal glycine of a protein. For most modified proteins, a myristoyl group is involved in reversible protein-protein and protein-membrane interactions via a myristoyl switch. The majority of studies on the molecular basis of myristoyl switches have focused on characterizing calcium-myristoyl switches, whereas research has suggested that hisactophilin binds reversibly to membranes by means of a pH dependent myristoyl switch. A number of in vivo studies of the modified protein have been completed to study the function of hisactophilin; however, the structure and stability of the protein have only been characterized using the recombinant non-myristoylated protein. In contrast, this study focuses on the effects of myristoylation on hisactophilin structure and stability. Myristoylated hisactophilin has been prepared by a dual plasmid expression system in E. coli. Optimization of the efficiency of myristoylaion was completed for the preparation and purification of myristoylated hisactophilin. Characterization of the structural changes caused by myristoylation were undertaken using multidimensional homo- and heteronuclear NMR experiments. Circular dichroism (CD) was used to obtain low resolution structural data and characterize protein stability as a function of pH using chemical denaturation. The molecular consequences of myristoylation on protein stability and structure, as well as the molecular basis for pH dependent myristoyl switch will be discussed
TOWARDS BUREAUCRATIZATION: AN ANALYSIS OF COMMON LEGAL REPRESENTATION PRACTICES BEFORE THE INTERNATIONAL CRIMINAL COURT
The status of victims in the international criminal project since the establishment of the International Criminal Court (ICC) is largely dealt with in the literature. Article 68(3) innovated as it allows victims of mass crimes to present their views and concerns before the first permanent international criminal jurisdiction. Yet, the case law over the last two decades shows that victims will not have the opportunity to directly take part in the ICC proceedings. Those who will participate to the trials are rather their legal representatives. This article explores victim participation through this new actor of international criminal trials. Common legal representation – i.e. the representation of hundreds, or even thousands, of victims by a sole lawyer – was promptly presented as unavoidable. To ensure operational efficiency, the selection of the legal representative was institutionalized. The practice of the Court prompted some impersonal lawyer-clients relationship. The author demonstrates the similarities between the organization of common legal representation in the ICC and the ideal-type of bureaucracy imagined by Weber.La place des victimes dans le projet pĂ©nal international a, depuis l’établissement de la Cour pĂ©nale internationale (CPI), Ă©tĂ© largement traitĂ©e par la littĂ©rature tant technique que critique. C’est donc Ă travers cet acteur que le prĂ©sent article souhaite traiter de la problĂ©matique de la participation des victimes devant la CPI. La reprĂ©sentation lĂ©gale commune – soit la reprĂ©sentation de centaines, voire milliers de victimes par un seul avocat – s’est rapidement rĂ©vĂ©lĂ©e inĂ©vitable. Dans un souci d’efficacitĂ©, la pratique actuelle a fait de la sĂ©lection du reprĂ©sentant lĂ©gal, un choix institutionnalisĂ© et incitĂ© Ă la dĂ©personnalisation de la relation avocat-clients.
