Anticancer effect of a novel palladium-saccharinate complex of terpyridine by inducing apoptosis on ehrlich ascites carcinoma (EAC) in balb-C mice
Background/Aim: [Pd(sac)(terpy)](sac)•4H2O (sac=saccharinate and terpy=2,2′:6′,2-terpyridine) is newlysynthesized palladium(II) (Pd) complex. We investigated the antiproliferative and apoptotic effects of this complex on Ehrlich ascites carcinoma (EAC). Materials and Methods: EAC cells were administered to 33 Balb/c mice. Mice were divided randomly into four groups: control, cisplatin, Pd(II) complex and paclitaxel. Control group animals received 0.9% NaCl; other groups received treatments cisplatin, Pd(II) complex and paclitaxel on days 7 and 12. At day 14, animals were sacrificed. Expression of active caspase-3, p53 and proliferating cell nuclear antigen (PCNA) was investigated and apoptosis was evaluated by terminal deoxynucleotidyltransferase (TdT)-mediated nick-end labelling (TUNEL) technique. Results: Expression of p53 and PCNA were found to be decreased (p<0.0001), cells with active caspase-3 and TUNEL-positive cells were found to be increased (p<0.0001) in all treatment groups. Conclusion: Like cisplatin and paclitaxel, this Pd(II) complex has a strong anticancer activity against EAC by inducing apoptosis and suppressing proliferation in vivo.Istanbul University (UDP-47082
Crocin ameliorates oxidative stress and suppresses renal damage in streptozotocin induced diabetic male rats
Diabetes mellitus (DM) is a prevalent metabolic disorder with complications including nephropathy, neuropathy, retinopathy and peripheral vascular disease. Crocin is a water soluble carotenoid that exhibits strong antioxidant activity. I investigated the potential hypoglycemic and reno-protective effects of crocin for type 1 diabetic male rats using periodic acid-Schiff and hematoxylin staining; metalloproteinase-7 (MMP-7), phosphatase and tensin homolog (PTEN) and phospho-Akt (p-Akt) immunohistochemistry; measurement of blood glucose, malondialdehyde (MDA), superoxide dismutase (SOD), total antioxidant and total oxidant status; and oxidative stress index. I used four groups of rats: control (saline treated), diabetic (single dose of 65 mg/kg streptozotocin), crocin (treated with 50 mg/kg) and diabetic + crocin. Crocin decreased levels of MDA, SOD, oxidative stress index and glucose significantly in diabetic animals. Renal damage from DM also was decreased by crocin treatment. MMP-7 and p-Akt immunoreactivity were stronger in the diabetic group compared to other groups, but PTEN immunoreactivity was weaker. Crocin treatment returned MMP-7 and PTEN expression to near normal. The ameliorative effect of crocin is attributed to its stimulation of the antioxidant defense system and its ability to regulate the MMP-7/PTEN/Akt signaling cascade
Kortikosteron ve Glukokortikoid Reseptör İnhibitörü Mifepristonun Epileptik Sıçanların Nöbetler ve Anksiyete Düzeyleri Üzerine Etkileri

Glukokortikoid (GK) kortikoste- ron (CORT) olarak
adlandırılmaktadır. Mifepriston (RU 486)(MİF) bir gluko- kortikoid reseptör
(GR) antagonistidir ve CORT'un hücre içi reseptörüne bağ- lanmasını önleyerek, etkilerini ortaya koymasını
engeller. Epilepsi, kendiliğin- den tekrarlayan nöbetlerle karakterize edilen,
sinir sistemini etkileyen nörolojik bir bozukluktur.
GK'lerin nöronal uyarılabilirlik, epileptik aktivite ve nöbetler
üzerindeki etkileri tartışmalıdır. Diğer taraftan epileptik
vakaların üçte biri gibi
yüksek bir oranında depresyon eşlik eder ve depresyon, epilepsi hastalarının
yaşam kalitesini düşüren önemli nedenlerden biridir. Çalışmamızda sıçanlarda
bulunan doğal glukokortikoid olan Kortikosteronun ve glukokortikoid reseptör inhibitörü olan Mifepristonun epileptik nöbet parametreleri ile birlikte hayvan- ların anksiyete düzeylerine
etkilerini açıklamayı amaçladık.
Çalışmamızda 66 adet erkek Wistar albino sıçan
rastgele her grupta 11 hayvan olacak şekilde Kontrol, DMSO, PTZ, CORT+PTZ,
MİF+PTZ, MİF+CORT+PTZ olmak üzere 6 gruba ayrıldı. Kindling epilepsi modelinde
nöbetler 30 dakika gözlenerek Racine
skalasına göre skorlandı. Kindling süreci
sonrasında anksiyeteyi test etmek için Yükseltilmiş Artı Labirent ve Açık Alan Testleri uygulandı.
Kindling PTZ grubunda sıçanların nöbet
latansları azalırken, nöbet fre- kansının ve şiddetinin arttığı görüldü. PTZ ve
CORT+PTZ grupları arasında, nöbet latansı, frekansı ve nöbet şiddeti değerleri
açısından anlamlı fark görül- medi. MİF+PTZ grubu latans süresinin PTZ
grubundan anlamlı olarak daha uzun, nöbetlere ait frekans ve şiddet
değerlerinin ise daha düşük olduğu göz- lemlendi. Anksiyete testleri sonuçları
incelendiğinde MİF+PTZ ve MİF+CORT+PTZ gruplarının anksiyete düzeylerinin hem
kontrol grubundanhem de PTZ ve CORT+PTZ gruplarından daha düşük olduğu
görülmüştür. PTZ ile indüklenen kindling epilepsi modelinde, mifepristonun
sıçanların nö- bet davranışları ve anksiyete düzeyleri üzerine olumlu etkileri
söz konusudur
Valeriana officinalis extract and 7-Nitroindozole ameliorated seizure behaviours, and 7-Nitroindozole reduced blood pressure and ECG parameters in pentylenetetrazole-kindled rats
Nitric oxide (NO) is known to be a neuromodulator with dual proconvulsive and anticonvulsive action. Valeriana officinalis (VAL) was previously believed to be antiepileptic, but is today known as a sedative and sleep regulator. Seizures may be associated with abnormal electrocardiographic changes and cardiac dysfunction arising from epilepsy may be related with neuronal nitric oxide (nNO).
