    Changes in Protective Behavioral Strategies and Alcohol Use among College Students

    Background—Protective behavioral strategies (PBS) are specific cognitive-behavioral strategies designed to reduce alcohol consumption and resulting negative consequences. A host of studies have examined the cross-sectional relationship between such strategies and alcohol use in the high-risk population of United States college students, but prospective studies on the construct are lacking. The primary purposes of this study were to determine if PBS use prospectively predicted subsequent alcohol use/alcohol-related problems and if changes in PBS use were associated with less alcohol use and fewer problems. Methods—Data were examined from 521 heavy drinking college students (60% male, 84% White, mean age = 18.9 years). Participants completed questionnaires assessing alcohol use, alcohol-related problems, and PBS use at baseline, 6-month, and 12-month follow-ups. Results—Analysis of residualized change scores indicated that increases in some PBS across time were associated with less alcohol use and fewer alcohol-related problems at follow-up. Findings regarding the prospective relationship between PBS use and subsequent alcohol use/ problems were equivocal. Discussion—Results from the study suggest that PBS may have value in alcohol-related interventions among college students. Clinicians who help clients increase their use of PBS may help those clients increase the probability of drinking less and experiencing fewer alcohol-related problems in the future

    Sex differences and within-family associations in the broad autism phenotype

    While there is a strong sex bias in the presentation of autism, it is unknown whether this bias is also present in subclinical manifestations of autism among relatives, or the broad autism phenotype. This study examined this question and investigated patterns of co-occurrence of broad autism phenotype traits within families of individuals with autism. Pragmatic language and personality features of the broad autism phenotype were studied in 42 fathers and 50 mothers of individuals with autism using direct assessment tools used in prior family studies of the broad autism phenotype. Higher rates of aloof personality style were detected among fathers, while no sex differences were detected for other broad autism phenotype traits. Within individuals, pragmatic language features were associated with the social personality styles of the broad autism phenotype in mothers but not in fathers. A number of broad autism phenotype features were correlated within spousal pairs. Finally, the associations were detected between paternal broad autism phenotype characteristics and the severity of children’s autism symptoms in all three domains (social, communication, and repetitive behaviors). Mother–child correlations were detected for aspects of communication only. Together, the findings suggest that most features of the broad autism phenotype express comparably in males and females and raise some specific questions about how such features might inform studies of the genetic basis of autism. </jats:p

    Who plays with whom: farrowing environment influences isolation of foster piglets in play

    Cross fostering piglets is a common management practise in the pig industry to manage large and heterogeneous litters, whereby piglets are moved from their biological litter to be reared by another sow. At present research has focused on immediate survival consequences and time of cross fostering, with little attention given to positive aspects of welfare such as social affiliations and the potential for positive interactions for these piglets such as play behaviour. The focus of our study was purely observational to record behaviour of piglets reared in either impoverished (farrowing crates) or enriched neonatal environments (PigSAFE pens) where fostering was practised as part of normal husbandry routines to promote piglet survival. We employed social network analysis to understand more about the behaviour of foster piglets in these environments and their acceptance into their new litter. In line with previous work, piglets exposed to enriched neonatal farrowing pens demonstrated more play behaviour than piglets reared in farrowing crates. We showed that pen piglets received and initiated more play invitations (higher degree centrality) than piglets in crates. We also found effects of cross fostering irrespective of neonatal environment. Non-foster piglets received and initiated more play behaviours (higher degree centrality) 2–3 weeks post-farrowing compared to piglets fostered into the litter and as such, fostered piglets remained isolated from play for the first few weeks of life. However, our data suggests this may be mitigated by neonatal environment; foster piglets reared in pens were better connected (betweenness centrality) within their foster litter than those fostered in crates. Our findings highlight the importance of the neonatal environment and its potential influence on the isolation of cross-fostered piglets and suggest that rearing in enriched neonatal environments may help mitigate against social isolation in early life of cross-fostered piglets, having obvious immediate, and long-term consequences for piglet welfare and behaviour. We also highlight the importance and application of methodologies such as social network analysis, for gaining more insight and understanding about the sociality of animal behaviour and its potential for measuring indicators of positive welfare, thus highlighting its application for veterinary science and animal behaviour and welfare research

    KIF3A Tail Domain Phosphorylation Is Not Required for Ciliogenesis in Mouse Embryonic Fibroblasts

    Primary cilia are essential signaling organelles that protrude from most cells in the body. Heterodimeric kinesin-2 (KIF3A/KIF3B/KAP3) powers several intracellular transport processes, including intraflagellar transport (IFT), essential for ciliogenesis. A long-standing question is how a motor protein is differentially regulated for specific cargos. Since phosphorylation of the KIF3A tail domain was suggested to regulate the activity of kinesin-2 for ciliogenesis, similarly as for the cytosolic cargo N-Cadherin, we set out to map the phosphosites involved in this regulation. Using well-characterized Kif3a−/−; Kif3b−/− mouse embryonic fibroblasts, we performed ciliogenesis rescue assays with a library of phosphomimetic mutants comprising all predicted phosphosites in the KIF3A tail domain. In contrast to previous reports, we found that KIF3A tail domain phosphorylation is dispensable for ciliogenesis in mammals. Thus, mammalian kinesin-2 is differently regulated for IFT than currently thought, consistent with the idea of differential regulation for ciliary and cytosolic cargo

    Fecal Viral Community Responses to High-Fat Diet in Mice.

    Alterations in diet can have significant impact on the host, with high-fat diet (HFD) leading to obesity, diabetes, and inflammation of the gut. Although membership and abundances in gut bacterial communities are strongly influenced by diet, substantially less is known about how viral communities respond to dietary changes. Examining fecal contents of mice as the mice were transitioned from normal chow to HFD, we found significant changes in the relative abundances and the diversity in the gut of bacteria and their viruses. Alpha diversity of the bacterial community was significantly diminished in response to the diet change but did not change significantly in the viral community. However, the diet shift significantly impacted the beta diversity in both the bacterial and viral communities. There was a significant shift away from the relatively abundant Siphoviridae accompanied by increases in bacteriophages from the Microviridae family. The proportion of identified bacteriophage structural genes significantly decreased after the transition to HFD, with a conserved loss of integrase genes in all four experimental groups. In total, this study provides evidence for substantial changes in the intestinal virome disproportionate to bacterial changes, and with alterations in putative viral lifestyles related to chromosomal integration as a result of shift to HFD.IMPORTANCE Prior studies have shown that high-fat diet (HFD) can have profound effects on the gastrointestinal (GI) tract microbiome and also demonstrate that bacteria in the GI tract can affect metabolism and lean/obese phenotypes. We investigated whether the composition of viral communities that also inhabit the GI tract are affected by shifts from normal to HFD. We found significant and reproducible shifts in the content of GI tract viromes after the transition to HFD. The differences observed in virome community membership and their associated gene content suggest that these altered viral communities are populated by viruses that are more virulent toward their host bacteria. Because HFD also are associated with significant shifts in GI tract bacterial communities, we believe that the shifts in the viral community may serve to drive the changes that occur in associated bacterial communities

    Optical absorption of ion-beam sputtered amorphous silicon coatings

    Low mechanical loss at low temperatures and a high index of refraction should make silicon optimally suited for thermal noise reduction in highly reflective mirror coatings for gravitational wave detectors. However, due to high optical absorption, amorphous silicon (aSi) is unsuitable for being used as a direct high-index coating material to replace tantala. A possible solution is a multimaterial design, which enables exploitation of the excellent mechanical properties of aSi in the lower coating layers. The possible number of aSi layers increases with absorption reduction. In this work, the optimum heat treatment temperature of aSi deposited via ion-beam sputtering was investigated and found to be 450 °C. For this temperature, the absorption after deposition of a single layer of aSi at 1064 nm and 1550 nm was reduced by more than 80%

    A decade on where is the UK poultry industry for emergency on-farm killing?

    Millions of poultry are farmed intensively every year across the United Kingdom (UK) to produce both meat and eggs. There are inevitable situations that require birds to be emergency killed on farm to alleviate pain and suffering. In Europe and the UK, emergency methods are regulated by the European Council Regulation (EC) No. 1099/2009 and The Welfare of Animals at the Time of Killing Regulations (England 2015; Scotland 2012; Wales and Northern Ireland 2014). Cervical dislocation has been reported to be the most widely used method prior to these legislative changes which took place from 1 January 2013. Based on limited scientific evidence and concern for bird welfare, these legislative changes incorporated restrictions based on bird weight for both manual (≤3 kg) and mechanical (≤5 kg) cervical dislocation, and introduced an upper limit in the number of applications for manual cervical dislocation (up to 70 birds per person per day). Furthermore, it removed methods which showed evidence of crushing injury to the neck. However, since legal reform new scientific evidence surrounding the welfare consequences of cervical dislocation and the development of novel methods for killing poultry in small numbers on farm have become available. Whether the UK poultry industry have adopted these novel methods, and whether legislative reform resulted in a change in the use of cervical dislocation in the UK remains unknown. Responses from 215 respondents working across the UK poultry industry were obtained. Despite legal reform, manual cervical dislocation remains the most prevalent method used across the UK for killing poultry on farm (used by 100% of farms) and remains the preferred method amongst respondents (81.9%). The use of alternative methods such as Livetec Nex® and captive bolt guns were available to less than half of individuals and were not frequently employed for broilers and laying hens. Our data suggests there is a lack of a clear alternative to manual cervical dislocation for individuals working with larger species and a lack of gold standard methodology. This risks bird welfare at killing and contributes to inconsistency across the industry. We suggest providing stakeholders with practical alternatives prior to imposing legislative changes and effective knowledge transfer between the scientific community and stakeholders to promote positive change and protect bird welfare
