Expertise modulates hemispheric asymmetry in holistic processing: evidence from Chinese character processing
Conference Theme: Mind, Technology, and SocietyHolistic processing (HP) has been proposed to be a character-istic of right hemisphere (RH) processing. Here we test this claim using the divided visual field paradigm with Chinese character stimuli. HP is assessed through the composite para-digm, which is commonly used in perceptual expertise re-search. We found that in novice Chinese readers, a standard HP pattern emerged only in the left visual field/RH but not in the right visual field/left hemisphere, consistent with the ana-lytic/holistic hemispheric dichotomy in the literature. However, in expert Chinese readers, neither visual field showed the HP pattern, consistent with the finding that re-duced HP is an expertise maker for Chinese character recog-nition. Thus, the RH does not always employ holistic process-ing; it depends on the perceiversβ experience with the stimuli. Enhancing 2D Growth of Organic Semiconductor Thin Films with Macroporous Structures via a Small-Molecule Heterointerface
The physical structure of an organic solid is strongly affected by the surface of the underlying substrate. Controlling this interface is an important issue to improve device performance in the organic electronics community. Here we report an approach that utilizes an organic heterointerface to improve the crystallinity and control the morphology of an organic thin film. Pentacene is used as an active layer above, and m-bis(triphenylsilyl) benzene is used as the bottom layer. Sequential evaporations of these materials result in extraordinary morphology with far fewer grain boundaries and myriad nanometre-sized pores. These peculiar structures are formed by difference in molecular interactions between the organic layers and the substrate surface. The pentacene film exhibits high mobility up to 6.3 cm(2)V(-1)s(-1), and the pore-rich structure improves the sensitivity of organic-transistor-based chemical sensors. The Multifunctional Sorting Protein PACS-2 Regulates SIRT1-Mediated Deacetylation of p53 to Modulate p21-Dependent Cell-Cycle Arrest
SIRT1 regulates the DNA damage response by deacetylating p53, thereby repressing p53 transcriptional output. Here, we demonstrate that the sortingprotein PACS-2 regulates SIRT1-mediated deacetylation of p53 to modulate the DNA damage response. PACS-2 knockdown cells failed to efficiently undergo p53-induced cell-cycle arrest in response to DNA damage. Accordingly, p53 acetylation was reduced both in PACS-2 knockdown cells and thymocytes from Pacs-2-/- mice, thereby blunting induction of the cyclin-dependent kinase inhibitor p21 (CDKN1A). The SIRT1 inhibitor EX-527 or SIRT1 knockdown restored p53 acetylation and p21 induction as well as p21-dependent cell-cycle arrest in PACS-2 knockdown cells. Traffickingstudies revealed that cytoplasmic PACS-2 shuttled to the nucleus, where it interacted with SIRT1 andrepressed SIRT1-mediated p53 deacetylation. Correspondingly, invitro assays demonstrated that PACS-2 directly inhibited SIRT1-catalyzed p53 deacetylation. Together, these findings identify PACS-2 as an invivo mediator of the SIRT1-p53-p21 axis that modulates the DNA damage response
Do logarithmic proximity measures outperform plain ones in graph clustering?
We consider a number of graph kernels and proximity measures including
commute time kernel, regularized Laplacian kernel, heat kernel, exponential
diffusion kernel (also called "communicability"), etc., and the corresponding
distances as applied to clustering nodes in random graphs and several
well-known datasets. The model of generating random graphs involves edge
probabilities for the pairs of nodes that belong to the same class or different
predefined classes of nodes. It turns out that in most cases, logarithmic
measures (i.e., measures resulting after taking logarithm of the proximities)
perform better while distinguishing underlying classes than the "plain"
measures. A comparison in terms of reject curves of inter-class and intra-class
distances confirms this conclusion. A similar conclusion can be made for
several well-known datasets. A possible origin of this effect is that most
kernels have a multiplicative nature, while the nature of distances used in
cluster algorithms is an additive one (cf. the triangle inequality). The
logarithmic transformation is a tool to transform the first nature to the
second one. Moreover, some distances corresponding to the logarithmic measures
possess a meaningful cutpoint additivity property. In our experiments, the
leader is usually the logarithmic Communicability measure. However, we indicate
some more complicated cases in which other measures, typically, Communicability

VEZF1 elements mediate protection from DNA methylation
There is growing consensus that genome organization and long-range gene regulation involves partitioning of the genome into domains of distinct epigenetic chromatin states. Chromatin insulator or barrier elements are key components of these processes as they can establish boundaries between chromatin states. The ability of elements such as the paradigm β-globin HS4 insulator to block the range of enhancers or the spread of repressive histone modifications is well established. Here we have addressed the hypothesis that a barrier element in vertebrates should be capable of defending a gene from silencing by DNA methylation. Using an established stable reporter gene system, we find that HS4 acts specifically to protect a gene promoter from de novo DNA methylation. Notably, protection from methylation can occur in the absence of histone acetylation or transcription. There is a division of labor at HS4; the sequences that mediate protection from methylation are separable from those that mediate CTCF-dependent enhancer blocking and USF-dependent histone modification recruitment. The zinc finger protein VEZF1 was purified as the factor that specifically interacts with the methylation protection elements. VEZF1 is a candidate CpG island protection factor as the G-rich sequences bound by VEZF1 are frequently found at CpG island promoters. Indeed, we show that VEZF1 elements are sufficient to mediate demethylation and protection of the APRT CpG island promoter from DNA methylation. We propose that many barrier elements in vertebrates will prevent DNA methylation in addition to blocking the propagation of repressive histone modifications, as either process is sufficient to direct the establishment of an epigenetically stable silent chromatin stat
Is PTEN loss associated with clinical outcome measures in human prostate cancer?
Inactivating PTEN mutations are commonly found in prostate cancer, resulting in an increased activation of Akt. In this study, we investigate the role of PTEN deletion and protein expression in the development of hormone-refractory prostate cancer using matched hormone-sensitive and hormone-refractory tumours. Fluorescent in situ hybridisation and immunohistochemistry was carried out to investigate PTEN gene deletion and PTEN protein expression in the transition from hormone-sensitive to hormone-refractory prostate cancer utilising 68 matched hormone sensitive and hormone-refractory tumour pairs (one before and one after hormone relapse). Heterogeneous PTEN gene deletion was observed in 23% of hormone sensitive tumours. This increased significantly to 52% in hormone-refractory tumours (P=0.044). PTEN protein expression was observed in the membrane, cytoplasm and the nucleus. In hormone sensitive tumours, low levels of cytoplasmic PTEN was independently associated with shorter time to relapse compared to high levels of PTEN (P=0.028, hazard ratio 0.51 (95%CI 0.27β0.93). Loss of PTEN expression in the nucleus of hormone sensitive tumours was independently associated with disease-specific survival (P=0.031, hazard ratio 0.52, 95%CI 0.29β0.95). The results from this study demonstrate a role for both cytoplasmic and nuclear PTEN in progression of prostate cancer to the hormone-refractory state
