    Development of a Two-Photon Laser Scanning Microscope

    The objective of this project was to convert a Sarastro 2000 confocal laser scanning microscope into a system capable of imaging using two-photon excitation (TPE) fluorescence for the use of the PSU biology department. TPE microscopy operates on the ability of fluorophores to accept two photons each with half the energy of a desired transition in a single quantum event via a virtual energy state and then emit a higher energy photon upon relaxation. This is preferable to single-photon excitation (SPE) due to the lower energy photons causing less damage to delicate biological samples. The adaptation process included physically altering the system by replacing the on-board argon laser with a Ti:sapphire laser, replacing optical components for optimization with the new laser, and implementing a new scan mechanism. The new scan mechanism, a custom all-analog two-mirror galvo head, interfaces with an analog data acquisition board and a computer with software designed for analog motor control and data analysis

    Specification of the underspecified: A pragmatic analysis of the injunctive in the Rgveda

    This paper uses formal pragmatics to show that discourse context alone is inadequate to explain the function of “injunctive” verb forms (i.e., finite verbs unspecified for tense or mood) in Rgvedic Sanskrit. Prior treatments, which explain the temporal and modal specification of the injunctive as being picked up from other verb forms in the immediate discourse, do not fully account for the injunctive’s observed meanings. By applying a framework known in neo-Gricean pragmatics as a “Horn strategy” to tense and modality, I explain the various functions of the injunctive as arising from partial blocking relationships that hold between it and other verb forms with which it competes

    Evaluation of ride quality prediction methods for helicopter interior noise and vibration environments

    The results of a simulator study conducted to compare and validate various ride quality prediction methods for use in assessing passenger/crew ride comfort within helicopters are presented. Included are results quantifying 35 helicopter pilots discomfort responses to helicopter interior noise and vibration typical of routine flights, assessment of various ride quality metrics including the NASA ride comfort model, and examination of possible criteria approaches. Results of the study indicated that crew discomfort results from a complex interaction between vibration and interior noise. Overall measures such as weighted or unweighted root-mean-square acceleration level and A-weighted noise level were not good predictors of discomfort. Accurate prediction required a metric incorporating the interactive effects of both noise and vibration. The best metric for predicting crew comfort to the combined noise and vibration environment was the NASA discomfort index

    Predictive modeling was used to identify the degree that hypothesized moderators of dialectical behavioral therapy for adolescents (DBT-A) treatment outcomes predicted anxiety and depression symptoms over time. Participants were 66 adolescents (41 girls; 25 boys) with a mean age of 15.38 years (SD = 1.51) who completed a 7-week DBT-A intervention. Analyses revealed convergent models, wherein emotion regulation and interpersonal effectiveness were substantial predictors of change in the symptoms of anxiety, F(4, 65) = 23.21, p \u3c .01, R2 = .60, and depression, F(4, 65) = 29.76, p \u3c .01, R2 = .66

    Two methods of quantifying helicopter ride quality; absorbed power for vibration only and the NASA ride comfort model for both noise and vibration are discussed. Noise and vibration measurements were obtained on five operational US Army helicopters. The data were converted to both absorbed power and DISC's (discomfort units used in the NASA model) for specific helicopter flight conditions. Both models indicate considerable variation in ride quality between the five helicopters and between flight conditions within each helicopter
