    Data mining analysis of miR-638 and key genes interaction in cisplatin resistant triple-negative breast cancer

    Cisplatn is one of the chemotherapy for the treatment of triple‐negatve breast cancer (TNBC), but its effectveness is limited because of the phenomenon of chemoresistance. miR‐638 was shown to regulate chemoresistance; however, it has never been validated in the cisplatn‐resistant tumor from patents. This present study aimed to identfy the key gene regulatory networks of miR‐638 and evaluate the potental role of the miR‐638 and its targets as potental prognosis biomarkers for cisplatn‐resistance triple‐negatve breast cancer patents. The miR‐638 target was obtained from the miRecords database while the mRNA of chemoresistance biomarker candidate was obtained from the GSE18864 of GEO database, which is mRNA of cisplatn‐resistance TNBC patents. CCND1 and FZD7 are potental candidates for cisplatn chemoresistance biomarkers in patents with TNBC. Moreover, a Kaplan‐Meier survival plot showed that breast cancer patents with low mRNA levels of FZD7 had signifcantly worse overall survival than those in higher mRNA expression group. Taken together, miR‐638 plays a role in cisplatn resistance mechanism through a mechanism involving its target gene CCND1 and FZD7. Overall, miR‐638, CCND1, and FZD7 are candidates for cisplatn biomarker resistance in TNBC

    The Effect of Store Atmosphere, Promotion, and Retail Service on Customer Satisfaction and Repurchase Intention

    The purpose of this study was to examine the effect of store atmosphere, promotion, and retail service on customer satisfaction and repurchase intention, we also investigate the effect of customer satisfaction on Indomaret customer repurchase intention in Denpasar City. The population in this study were all Indomaret customers in Denpasar City, whose numbers cannot be stated or counted with certainty (infinite). Determination of the sample in this study using purposive sampling technique. The sample set in this study were 105 respondents. Data analysis in this study used path analysis with the Partial Least Square (PLS) approach. The results showed that store atmosphere, promotion, and retail service had a positive and significant effect on customer satisfaction and repurchase intention, and customer satisfaction also had a positive and significant effect on repurchase intention.     Pengaruh Retail Mix terhadap Kepuasan Pelanggan pada Foodmart Primo Lippo Mall Kuta

    A strategy that retail businesspeople can use to win the competition is by applying the right retail mix strategy. Elements of the retail mix can be combined and combined in such a way as to satisfy and retain existing customers. The purpose of this research is to explain the influence of retail mix simultaneously and partially to customer satisfaction at Foodmart Primo Lippo Mall Kuta. The research subjects were all Foodmart Primo Lippo Mall customers in Kuta, with a sample of 115 people. Determination of sample using purposive sampling method, that is sample determination technique with certain consideration. Data were analyzed using multiple linear regression analysis technique. The results showed that the retail mix (price, merchandise, promotion, service, store location, and store atmosphere) simultaneously and partially significant effect on customer satisfaction at Foodmart Primo Lippo Mall Kuta. This has the meaning that the better the application of the retail mix at Foodmart Primo Lippo Mall Kuta then the increase also the level of customer satisfaction

    Caesalpinia sappan L. Ethanolic Extract Decrease Intracellular ROS Level and Senescence of 4T1 Breast Cancer Cells

    Highly and uncontrolled cell proliferation on cancer cells may boost ROS level intracellular accumulation up significantly. Sappan heartwood (Caesalpinia sappan L.) have been known to have cytotoxicity effect toward several cancer cells. This research was conducted to develop Caesalpinia sappan L. heartwood ethanolic extract (CSE) as chemopreventive agent which can be used as cancer therapy, looked from antioxidant and anti-senescence activity toward 4T1 breast cancer cell. CSE was obtained through maceration using ethanol 70% as solvent. Cytotoxicity activity of CSE was done by using MTT assay with IC50 as parameter. This IC50 was used as basic to the next assay, ROS assay by using DCFDA staining flowcytometry to look ROS level intracellular of CSE toward 4T1 cell and senescence assay by using senescence associated β-galactosidase (SA β-gal) assay with % cell senescence as their parameter. CSE toxic to 4T1 cell that proved from IC50 value of 25 µg/mL. Single treatment of CSE on concentration 12,5; 25; and 37.5 µg/mL able to suppress ROS level intracellular. If compared with untreated cell, single treatment of CSE on concentration 6 and 12 µg/mL showed % cell senescence not significantly different. Based on this result, CSE is cytotoxic, has antioxidant and antisenescence activity, so it has great potential to developed as chemoprevention agent on cancer therapy.Keywords : Caesalpinia sappan L., 4T1 breast cancer line, Reactive Oxygen Species (ROS), anti-senescenc

    Hesperidin, senyawa flavonoid menunjukkan efek toksik pada beberapa sel kanker. Tujuan penelitian Inl adalah untuk mengetahui aktlvltas sitotoksik hesperidin tunggal dan kombinasinya dengan doxorubicin pada sel MCF-7. Vilabilitas sel perlakuan hesperidin, doxorubicin dan kombinasinya ditetapkan dengan uji MTT. pengamatan apoptosis dilakukan dengan metode double staining menggunakan Ethidium Bromide-Acridine Orange. Hesperidin belum menunjukkan efek sitotoksik namun doxorubicin memberikan efek sitotoksik dengan IC&#8325&#8320467 nM. Perlakuan hesperidin (5, SO dan 100 µM) meningkatkan efek sitotoksik doxorubicin 200 nM dibandingkan perlakuan doxorubicin tunggal. Efek sitotoksik terbesar diperoleh pada kombinasi doxorubicin 200 nM dan hesperidin 100 µM. Kombinasi doxorubicin 200 nM dan hesperidin 100µM juga menunjukkan pemacuan apoptosis sel MCF-7. Hesperidin potensial untuk dikembangkan sebagai agen ko-kemoterapi pada kanker payudara, namun mekanisme molekulernya masih perlu diteliti lebih lanjut

    Keberadaan Usaha Kecil Menengah (UMKM) memiliki peran yang sangat penting dan strategis di dalam pembangunan ekonomi. Untuk mampu bertahan di tengah persaingan yang semakin ketat, UMKM perlu memiliki keunggulan kompetitif dengan menciptakan inovasi. Tujuan penelitian ini yaitu untuk menganalisis variabel penentu daya saing dalam meningkatkan inovasi yang berperan dalam keunggulan bersaing UMKM batik Kota Tasikmalaya. Metode penelitian yang digunakan yaitu metode deskriptif kuantitatif. Data didapatkan melalui proses wawancara dengan menggunakan kuesioner tertutup. Hasil menunjukkan bahwa daya saing UMKM batik Kota Tasikmalaya secara langsung dipengaruhi oleh orientasi produk dan orientasi pasar. Kedua faktor tersebut secara tidak langsung mempengaruhi daya saing melalui strategi diferensiasi. Strategi diferensiasi menjadi penting agar UMKM dapat bertahan di tengah persaingan. Diferensiasi menjadi faktor kunci bagi terciptanya inovasi produk. UMKM yang memiliki karakteristik unik dinilai dapat mempertahankan keberlanjutan usahanya

    Hesperidin increase cytotoxic effect of doxorubicin in MCF-7 cells

    Hesperidin,  a  flavonoid,  shows  strong  cytotoxic  effect  in  several  cancer cell  lines.  The  aim  of  this  research  was  to  investigate  cytotoxic  activities  of hesperidin  alone  and  in  combination  with  doxorubicin.  Cell  viability  assay  of hesperidin,  doxorubicin,  and  combination  treatments  were  carried  out  by  using MTT  assay.  Apoptosis  assay  was  done  using  double  staining  method  using Ethidium  Bromide-Acridine  Orange.  Hesperidin  did  not  show  cytotoxic  effect but doxorubicin showed cytotoxic effect with IC50467 nM. Hesperidin (5, 50 and 100  µM)  increased  cytotoxic  effect  of  doxorubicin  compared  with  doxorubicin alone.  The  strongest  cytotoxic  activity  was  showed  by  the  combination  of  200 nM  doxorubicin  and  100  µM  hesperidin.  Combination  treatment  of  doxorubicin 200  nM  and  hesperidin  100  µM  induced  apoptosis  in  MCF-7 cells.  Hesperidin  is potentially  to  be  developed  as  co-chemotherapeutic  agent  for  breast  cancer, while molecular mechanism need to be explored.Key words: Hesperidin, doxorubicin, synergism, MCF-7, apoptosis

    Combination of Tangeretin and 5-Fluorouracil Modulates Cell Cycle and Induce Apoptosis on WiDr Cells

    Co-chemotherapeutics approaches are increasing in cancer treatment in order mainly to suppress the resistence phenomenon of cancer treatment and to enhance the cytotoxic effect of the main chemotherapeutics agent. Tangeretin has been known to have cytotoxic effect to some cancer cells through some pathways in the cells. To explore the potential effect of tangeretin as co-chemotherapeutics agent this research was subjected to study the cytotoxic effect of tangeretin in combination with 5-Fluoro Uracil (5-FU) on WiDr colon cancer cells covering the modulation of cell cycle and apoptosis induction. Cytotoxic effect was examined by using MTT assay while apoptotis induction was determined by annexin-V flowcytometry. Under MTT assay, tangeretin showed weak cytotoxic activity on the cells. However, tangeretin significantly enhanced the cytotoxic effect of 5-FU on the cells. This co-chemotherapeutics effect likely correlated with cell cycle modulation effect, especially in inducing polyploidy phenomenon as expressed in the flowcytometric graph of the DNA content. This combination also increased apoptosis induction. These result suggest that tangeretin is potential to be developed as co-chemotherapeutic agent for 5-Fu on colon cancer and further molecular mechanism need to be explored.Keywords: Tangeretin, 5-Fluorourasil, WiDr, cell cycle, apoptosis
