IgE plays a pivotal role in allergic asthma especially in the acute response to antigen and in the propagation of airw,ay inflammation. Propolis which has been used widely in folk medicine, has been shown to exhibit various biological activilies. Investigation of these activities ofpropolis using an ovalbumin-induced asthma animcl model has been conducted- Mousewere immunized and sensitized by exposure to ovalbumin (OVA) antigen and administered propolis ethanolic extrocts with dose I (6 mg/kg body weightlday) and dose II (7.5 mg/kg body weight/day) by tube feeding. The investigation demonstrated that propolis ethanalic extracts could suppressed the serum levels of IgE in OVA-sensitized mouse. Efek Ekstrak Etanol Propolis Isolat Gunung Lawu terhadap Hitung Netrofil, Eosinofil Dan IL-17 pada Mencit Model Asma Kronik

Asma merupakan penyakit inflamasi kronik saluran nafas, sebagai hasil interaksi faktor-faktor imunitas seluler (netrofil, eosinofil dan makrofag) maupun humoral. IL-17 menginduksi perekrutan neutrofil ke dalam saluran nafas, dan berperan dalam patogenesis asma serta penentuan keparahan penyakit. Tujuan khusus penelitian ini adalah menganalisis efek ekstrak etanol propolis (EEP) isolat Gunung Lawu terhadap hitung eosinofil dan netrofil serta kadar IL-17 pada mencit Balb/C model asma kronik. Penelitian ini menggunakan mencit Balb/C jantan, dibagi empat kelompok masing-masing kelompok tujuh ekor. Kelompok (i) kontrol, (ii) asma, (iii) asma+EEP dosis 6 mg/kgBB/hari/oral, dan (iv) asma+antihistamin. Untuk membuat model asma kronik, semua hewan coba kecuali kelompok control disensitisasi pada hari ke-0 dan 5 dengan injeksi intraperitoneal (i.p.) ovalbumin (OVA, 100 μg/mencit) dan aluminium hidroksida (10%, 50 mg/mencit) dalam phosphate-buffered saline (PBS). Hewan coba disensitisasi selama 6 minggu dengan inhalasi 2% (b/v) OVA aerosol dalam NaCl 0,9% selama 30 menit/hari setiap 3 hari/minggu. Deteksi kadar IL-17 serum menggunakan ELISA. Hitung eosinofil dan netrofil di laboratorium swasta. Data yang diperoleh di analisis Uji ANOVA atau alternatifnya menggunakan program SPSS for Windows Release 15 dan p <0,05 dipilih sebagai tingkat minimal signifikansinya. Kadar IL-17 kelompok kontrol (17,15±8,25 pg/mL), asma (75,62±17,64 pg/mL), propolis (45,59±10,69 pg/mL), dan antihistamin (38,98±7,57 pg/mL). Hitung eosinofil kelompok kontrol (149,29±42,38 sel/mm3), asma (156,43±48,18 sel/mm3), propolis (100,57±35,58 sel/mm3), dan antihistamin (105,43±33,20 sel/mm3). Hitung netrofil kelompok kontrol (155,83±80,31 sel/mm3), asma (277,10±115.67 sel/mm3), propolis (261,39±238,75 sel/mm3), dan antihistamin (141,89±86,09 sel/mm3).
