    Antidiabetic activity of isoquercetin in diabetic KK -Ay mice

    <p>Abstract</p> <p>Background</p> <p>Tartary buckwheat bran is an important natural source of quercetin and isoquercetin. Quercetin and isoquercetin are both powerful α-glucosidase inhibitors. Although the IC<sub>50 </sub>of isoquercetin as α-glucosidase inhibitor was much higher than that of quercetin, the bioavailability of isoquercetin was higher than that of quercetin. Hence, we are interested in the antidiabetic effect of isoquercetin in diabetic KK -A<sup>y </sup>mice.</p> <p>Methods</p> <p>The hypoglycemic effect of isoquercetin in a type 2 diabetic animal model (KK-A<sup>y </sup>mice) was studied. Isoquercetin was administrated at doses of 50, 100 and 200 mg/kg for 35 days.</p> <p>Results</p> <p>It was found that fasting blood glucose concentration was decreased with the 200 mg/kg group (<it>p </it>< 0.01) the most efficient compared with the diabetic control group. In addition, there was significant decrease in plasma C-peptide, triglyceride, total cholesterol and blood urea nitrogen levels after 35 days. Meanwhile, glucose tolerance was improved, and the immunoreactive of pancreatic islets β-cells was promoted.</p> <p>Conclusions</p> <p>These results suggest that isoquercetin had a regulative role in blood glucose level and lipids, and improved the function of pancreatic islets. Isoquercetin may be useful in the treatment of type 2 diabetes mellitus.</p

Protein is considered a key nutritional advantage of quinoa grain, and many studies have highlighted the nutritional and physicochemical properties of quinoa protein. In addition, quinoa protein is a good precursor of bioactive peptides. This review focused on the biological properties of quinoa protein hydrolysate and peptides, and gave a summary of the preparation and functional test of quinoa protein hydrolysate and peptides. A combination of milling fractionation and solvent extraction is recommended for the efficient production of quinoa protein. The biological functionalities of quinoa protein hydrolysate, including antidiabetic, antihypertensive, anti-inflammatory, antioxidant activities, and so on, have been extensively investigated based on in vitro studies and limited animal models. Additionally, bioinformatics analysis, including proteolysis simulation, virtual screening, and molecular docking, provides an alternative or assistive approach for exploring the potential bioactivity of quinoa protein and peptides. Nevertheless, further research is required for industrial production of bioactive quinoa peptides, verification of health benefits in humans, and mechanism interpretation of observed effects

    Individual or mixing extrusion of Tartary buckwheat and adzuki bean: Effect on quality properties and starch digestibility of instant powder

    IntroductionTartary buckwheat and adzuki bean, which are classified as coarse grain, has attracted increasing attention as potential functional ingredient or food source because of their high levels of bioactive components and various health benefits.MethodsThis work investigated the effect of two different extrusion modes including individual extrusion and mixing extrusion on the phytochemical compositions, physicochemical properties and in vitro starch digestibility of instant powder which consists mainly of Tartary buckwheat and adzuki bean flour.ResultsCompared to mixing extrusion, instant powder obtained with individual extrusion retained higher levels of protein, resistant starch, polyphenols, flavonoids and lower gelatinization degree and estimated glycemic index. The α-glucosidase inhibitory activity (35.45%) of the instant powder obtained with individual extrusion was stronger than that obtained with mixing extrusion (26.58%). Lower levels of digestibility (39.65%) and slower digestion rate coefficient (0.25 min−1) were observed in the instant powder obtained with individual extrusion than in mixing extrusion (50.40%, 0.40 min−1) by logarithm-of-slope analysis. Moreover, two extrusion modes had no significant impact on the sensory quality of instant powder. Correlation analysis showed that the flavonoids were significantly correlated with physicochemical properties and starch digestibility of the instant powder.DiscussionThese findings suggest that the instant powder obtained with individual extrusion could be used as an ideal functional food resource with anti-diabetic potential

    A Determination of Potential α-Glucosidase Inhibitors from Azuki Beans (Vigna angularis)

    A 70% ethanol extract from azuki beans (Vigna angularis) was extracted further with CH2Cl2, EtOAc and n-BuOH to afford four fractions: CH2Cl2-soluble, EtOAc-soluble, n-BuOH-soluble and residual extract fractions. The EtOAc-soluble fractions showed the highest α-glucosidase inhibitory activity. Two pure flavonoid compounds, vitexin and isovitexin, were isolated (using the enzyme assay-guide fractionation method) from the EtOAc-soluble fractions. We further evaluated the interaction between the flavonoid compounds and α-glucosidase by fluorescence spectroscopy. Vitexin and isovitexin showed high inhibitory activities, with IC50 values of 0.4 mg·mL−1 and 4.8 mg·mL−1, respectively. This is the first study of the active compositions of azuki beans against α-glucosidase

    Major Phenolic Compounds, Antioxidant Capacity and Antidiabetic Potential of Rice Bean (Vigna umbellata L.) in China

    Interest in edible beans as nutraceuticals is increasing. In the present study, the individual phenolic acids, the total phenolic content (TPC), the total flavonoid content (TFC), and the antioxidant and antidiabetic potential of 13 varieties of rice beans from China were investigated. Eight phenolic compounds (catechin, epicatechin, p-coumaric acid, ferulic acid, vitexin, isovitexin, sinapic acid, quercetin) were analyzed on an ultra-performance liquid chromatography (UPLC) mass spectrometry (MS) system. The rice bean varieties had significant differences in total phenolic compounds (ranging from 123.09 ± 10.35 to 843.75 ± 30.15 μg/g), in TPC (ranging from 3.27 ± 0.04 to 6.43 ± 0.25 mg gallic acid equivalents (GAE)/g), in TFC (ranging from 55.95 ± 11.16 to 320.39 ± 31.77 mg catechin (CE)/g), in antioxidant activity (ranging from 39.87 ± 1.37 to 46.40 ± 2.18 μM·TE/g), in α-glucosidase inhibition activity (ranging from 44.32 ± 2.12 to 68.71 ± 2.19) and in advanced glycation end products formation inhibition activity (ranging from 34.11 ± 0.59 to 75.75 ± 0.33).     Biological Potential of Sixteen Legumes in China

    Phenolic acids have been identified in a variety of legumes including lima bean, broad bean, common bean, pea, jack bean, goa bean, adzuki bean, hyacinth bean, chicking vetch, garbanzo bean, dral, cow bean, rice bean, mung bean and soybean. The present study was carried out with the following aims: (1) to identify and quantify the individual phenolic acid and determine the total phenolic content (TPC); (2) to assess their antioxidant activity, inhibition activities of α-glucosidase, tyrosinase, and formation of advanced glycation endproducts; and (3) to investigate correlations among the phytochemicals and biological activity. Common bean possesses the highest antioxidant activity and advanced glycation endproducts formation inhibition activity. Adzuki bean has the highest α-glucosidase inhibition activity, and mung bean has the highest tyrosinase inhibition activity. There are significant differences in phytochemical content and functional activities among the bean species investigated. Selecting beans can help treat diseases such as dermatological hyperpigmentation illness, type 2 diabetes and associated cardiovascular diseases

    Determination of Multi-Class Mycotoxins in Tartary Buckwheat by Ultra-Fast Liquid Chromatography Coupled with Triple Quadrupole Mass Spectrometry

    Considering crops are susceptible to toxicogenic fungi during plantation, pre-processing and storage, an ultra-fast liquid chromatography coupled with triple quadrupole mass spectrometry (UFLC-QTrap-MS/MS) method was developed and validated for simultaneous determination of the 12 most frequent mycotoxins, including aflatoxin B1, B2, G1, G2, HT-2, T-2 toxin, ochratoxin A, fumonisin B1, B2, zearalanone, zearalenone, and deoxynivalenol, in 14 batches of Tartary buckwheat cultivar, collected from different origins in Sichuan Province, China. Differing from those complicated approaches, a simple and cost-efficient pretreatment method based on dilute-and-shoot was employed. Based on optimized chromatographic and mass spectrometry conditions, these 12 mycotoxins could be analyzed with high correlation coefficients (all over 0.995), high precision (RSD 0.47–9.26%), stability (RSD 0.72–11.36%), and recovery (79.52% to 108.92%, RSD 4.35–14.27%). Furthermore, this analysis method exhibited good determination performance with little disturbance of the matrix effect. Finally, this proposed method was applied for 14 batches of Tartary buckwheat seeds, in which aflatoxin B1 (AFB1) was detected in one moldy cultivar, Meigu No. 2, with its concentration exceeding the maximum residue limits set by EU regulations. The method thus established, which has significant advantages, could provide a preferred determination approach candidate for measurement of multiple mycotoxins measurement in Tartary buckwheat, even other kinds of foodstuffs
