    Seroprevalence of and associated risk factors for Leptospira interrogans serovar Hardjo infection of cattle in Setif, Algeria

    Background: Leptospirosis is a cosmopolitan zoonosis caused by Leptospira interrogans responsible for heavy loss both economically and in health, in humans and animals. This study was conducted to determine the seroprevalence and risk factors associated with Leptospira interrogans serovar Hardjo infection in cattle in the state of Setif, northeastern Algeria.Methodology: Between the period 2015 and 2019, a total of 48 randomly selected herds of cattle were investigated, and 406 sera from apparently healthy cattle were analyzed using an indirect enzyme-linked immuno-sorbent assay (ELISA). In order to determine possible risk factors related to leptospirosis, a pre-validated questionnaire was administered to herd owners.Results: The herd prevalence of Leptospira interrogans serovar Hardjo was 31.25% (15/48, 95% CI 19.95–45.33) while the cattle prevalence was 5.42% (22/406, 95% CI 3.61–8.07). Multivariable logistic regression analysis showed that the age of cattle between 3 and 6 years (OR = 9.25; p< 0.03), breeding herd size > 20 cows (OR = 13.65; p< 0.01), and semi-intensive management system (OR = 0.21; p< 0,02) were significantly associated with seropositivity to Leptospira interrogans serovar Hardjo.Conclusion: We concluded from this study that Leptospira interrogans serovar Hardjo is circulating among cattle farms in the state of Setif, Algeria. Furthermore, we recommend more studies to be carried out to prove the infectivity and implementation good hygienic practices among cattle farms and people at risk.     Salmonella Dublin associated with abortion in dairy cattle in Algiers and comparison of different diagnostic methods

    Background: In cattle, many serotypes of Salmonella enterica are responsible for a wide variety of clinical manifestations, which can cause considerable economic loss. Some serotypes can cause cows to abort sporadically, such as the Dublin serotype. This study was carried out on different cattle farms in the Algiers region to determine the prevalence of Salmonella Dublin using bacteriological and immunological methods.Methodology: The prevalence of Salmonella was determined by bacteriological analysis in accordance with the reference method AFNOR NF U 47-100 on faecal samples collected from 184 cattle belonging to 19 different farms, and serotyping for S. Dublin. Immunological analysis by enzyme-linked immunosorbent assay (ELISA) for S. Dublin was carried out on milk samples collected from 91 cattle. A survey of case (n=5) and control (n=14) farms for comparative analysis was performed to demonstrate a link between abortion in cows and prevalence of S. Dublin with both bacteriological and immunological methods. Sensitivity, specificity, Cohen Kappa coefficient, McNemar test odds ratios, and confidence intervals were calculated using Winepiscope 2.0 and StatA 9.1 software,and p<0.05 was considered as statistically significant.Results: The bacteriological results showed a prevalence of 7.6% (95%CI: 3-10), for Salmonella and serotyping revealed a prevalence for S. Dublin of 2.7%. The immunological analysis of milk by the ELISA technique revealed a prevalence of 13.2% (95%CI: 5-20) for S. Dublin. The comparative study between immunological results from milk and bacteriological results from faeces for detecting S. Dublin  showed poor agreement between the two tests (k=0.25), with enzyme immunoassay being significantly more sensitive than the bacteriological test (p<0.05). The results of the survey did not demonstrate a clear association between bacteriological detection of S. Dublin in faeces and abortion in cows (OR=8.66, 95%CI: 0.58-130.12). However, with the immunological analysis of milk for S. Dublin, there was a significant positive association (OR=62.33, 95%CI: 2.13-18.22) between a positive antibody response to S. Dublin in milk and the presence of abortions on the farm.Conclusion: In view of these results, we can conclude that Salmonella infections should systematically feature in the differential diagnosis of abortions in dairy cattle in Algeria.     Infection with Babesia canis in dogs in the Algiers region: Parasitological and serological study

    Background and Aim: Canine babesiosis is a vector-borne disease transmitted by ticks of the Ixodidae family. The effects of infection in dogs can range from the subclinical to the severe lethal form. This study aimed to make an original contribution to the knowledge of circulating species of Babesia spp. in dogs in the region of Algiers as well as mechanisms and risk factors for their transmission. Materials and Methods: An epidemiological study was carried out on 189 blood samples taken from dogs from April 2015 to January 2016. The samples taken underwent parasitological by Giemsa-stained blood smear and serological analyzes by indirect fluorescent antibody test (IFAT). The ticks were looked on all the dogs taken. Results: Giemsa-stained blood smears revealed the presence of two groups of parasites of the genus Babesia: Large Babesia (3/25, 12%) and small Babesia (22/25, 88%). The IFAT at a dilution of 1/32 showed an overall seroprevalence with Babesia canis of 17.98% (95% confidence interval 11.53-22.46). The distribution of the antibody titers for the positive samples showed that of the 34 positive sera with a titer ≥1/32, 28 sera remained positive at a dilution of 1/64 (14.81%), 22 at a dilution of 1/128 (11.64%) and 15 sera remained positive at a dilution of 1/256 (7.93%). Although seroprevalence varied according to canine population (20% and 19.49% in pet dogs and canine pound dogs, respectively, and 6.66-0% in farm dogs and hunting dogs, respectively), statistical analysis showed no significant differences between populations. The antibody titers obtained after several dilutions showed that 22 canine pound dog sera remained positive at a dilution of 1/128 compared to pet dogs and farm dogs which ceased to be positive at the dilution of 1/64. The comparison between the two diagnostic methods showed a strong agreement between the parasitological examination by blood smear and the serological method by IFAT. However, IFAT was much more sensitive. The analysis of risk factors, which may influence B. canis seroprevalence, has shown the influence of age, tick presence, and season. Finally, of the 242 ticks collected from a total of 59 dogs, only one tick species was identified, Rhipicephalus sanguineus. Conclusion: This study indicates a frequent circulation of species of Babesia in the dog in the Algiers region and R. sanguineus was the only tick identified

    Anaplasmosis is an infectious disease affecting goats and other ruminants. Our goal was to estimate the prevalence of Anaplasma spp. infections, and to identify different tick species in goa ts along with their associated risk factors. The epidemiology of Anaplasma spp. (A. ovis/A. marginale) was investigated from April to September 2016 in dairy goats from three different agro-ecological areas in the northeast region of Algeria (Guelma and El-Taref). We tested 182 goat sera using a MSP5 competitive ELISA (cELISA) test for IgG antibody screening, and by microscopic examination using the Giemsa (May-Greunwald-Giemsa) stain of 128 blood smears to detect intraerythrocytic inclusions bodies. The results demonstrated a total infection rate of 78.02 % and 42.18 % using cELISA and MGG, respectively. Only two species of ticks collected were identified, i. e. Rhipicephalus sanguineus (85.39 %) and Rhipicephalus bursa (14.60 %). Our study revealed that factors, such as the season, the type of farming, the hygienic status and the climatic conditions of the studied areas were found to be significantly associated with the tick infestation of goats (P < 0.05). The Anaplasma spp. infection prevalence was found to be highly dependent on the climatic conditions, the season (P < 0.05) and most closely related to the type of breeding and herd management (P = 0.000)

    Comparison of different serological methods to detect antibodies specific to Neospora caninum in bovine and canine sera

    Neospora caninum is an apicomplexan parasite responsible for paresis in dogs and abortion in cattle worldwide. Dogs serve as a definitive host, while cattle serve as intermediate host. Many different methods have been developed to detect specific antibodies present in cattle and dog serum. In the present study, the dense granule protein NcGRA6 was incorporated in a latex beads agglutination test (LAT), and compared to other serological methods, including enzyme-linked immunosorbent assay, the direct agglutination test, the immunoblot, and the indirect fluorescent antibody test (IFAT). Using the IFAT as the reference method, 100 sera isolated from Algerian cattle and 100 sera isolated from Algerian dogs, both possibly infected with N. caninum, were used to evaluate the LAT. The sensitivity, specificity, and kappa index were calculated for each host species and assay. For dog sera, the sensitivity and the specificity of the LAT was 76% and 100%, respectively. The McNemar test showed that the LAT was not significantly different from IFAT (P &gt; 0.05). For cattle sera, the sensitivity and the specificity of the LAT were 60% and 100%, respectively. The McNemar test indicated that the LAT was significantly different from IFAT (P &lt; 0.01) and that the LAT was only positive for cattle sera with titers of 1:800 or greater, indicating that LAT can be used for cattle in a clinical context. As well, the LAT has the advantage of being easy and rapid to perform compared to the other assays.</p
