    Monitoring PD-L1 positive circulating tumor cells in non-small cell lung cancer patients treated with the PD-1 inhibitor Nivolumab

    Controversial results on the predictive value of programmed death ligand 1 (PD-L1) status in lung tumor tissue for response to immune checkpoint inhibitors do not allow for any conclusive consideration. Liquid biopsy might allow real-time sampling of patients for PD-L1 through the course of the disease. Twenty-four stage IV NSCLC patients included in the Expanded Access Program with Nivolumab were enrolled. Circulating tumor cells (CTCs) were analyzed by CellSearch with anti-human B7-H1/PD-L1 PE-conjugated antibody. PD-L1 expressing CTCs were assessed at baseline, at 3 and 6 months after starting therapy, and correlated with outcome. At baseline and at 3 months of treatment, the presence of CTCs and the expression of PD-L1 on their surface were found associated to poor patients outcome. Nevertheless, the high frequency of PD-L1 expressing CTCs hampered to discriminate the role of PD-L1 in defining prognosis. Conversely although CTCs were found in all patients 6 months after treatment, at this time patients could be dichotomized into two groups based PD-L1 expression on CTCs. Patients with PD-L1 negative CTCs all obtained a clinical benefit, while patients with PD-L1 (+) CTCs all experienced progressive disease. This suggests that the persistence of PD-L1(+) CTCs might mirror a mechanism of therapy escape

    A pectin-honey hydrogel prevents postoperative intraperitoneal adhesions in a rat model.

    BACKGROUND: Adhesions are a common postoperative surgical complication. Liquid honey has been used intraperitoneally to reduce the incidence of these adhesions. However, solid barriers are considered more effective than liquids in decreasing postoperative intra-abdominal adhesion formation; therefore, a new pectin-honey hydrogel (PHH) was produced and its effectiveness was evaluated in a rat cecal abrasion model. Standardized cecal/peritoneal abrasion was performed through laparotomy in 48 adult Sprague-Dawley rats to induce peritoneal adhesion formation. Rats were randomly assigned to a control (C) and treatment (T) group. In group T, PHHs were placed between the injured peritoneum and cecum. Animals were euthanized on day 15 after surgery. Adhesions were evaluated macroscopically and adhesion scores were recorded and compared between the two groups. Inflammation, fibrosis, and neovascularization were histologically graded and compared between the groups. RESULTS: In group C, 17 of 24 (70.8%) animals developed adhesions between the cecum and peritoneum, while in group T only 5 of 24 (20.8%) did (p = 0.0012). In group C, one rat had an adhesion score of 3, sixteen had scores of 2, and seven rats had scores of 0. In group T, four rats had adhesion scores of 2, one rat had an adhesion score of 1 and nineteen have score 0 (p = 0.0003). Significantly lower grades of inflammation, fibrosis, and neovascularization were seen in group T (p = 0.006, p = 0.001, p = 0.002, respectively). CONCLUSION: PHH is a novel absorbable barrier that is effective in preventing intra-abdominal adhesions in a cecal abrasion model in rats.     Anti-nociceptive activity and toxicity evaluation of Cu(II)-fenoprofenate complexes in mice

    The proposed curative properties of copper(II)-non-steroidal anti-inflammatory drugs (NSAIDs) have led to the development of numerous copper(II)-NSAID complexes with enhanced anti-inflammatory activity. In this work, the antinociceptive and toxic effects of two new coordination complexes: Cu₂(fen)₄(caf)₂ [fen: fenoprofenate anion; caf: caffeine] and Cu₂(fen)₄(dmf)₂ [dmf: N-N'-dimethylformamide] were evaluated in mice. The antinociceptive effect was evaluated with two models: acetic acid-induced writhing response and formalin test. For the sub-acute exposure, the complexes were added to the diet at different doses for 28days. Behavioral and functional nervous system parameters in a functional observational battery were assessed. Also, hematological, biochemical and histopathological studies were performed. Cu₂(fen)₄(caf)₂ and Cu₂(fen)₄(dmf)₂ significantly decreased the acetic acid-induced writhing response and the licking time on the late phase in the formalin test with respect to the control and fenoprofen salt groups. The sub-acute exposure to Cu₂(fen)₄(caf)₂ complex increased the motor activity, the number of rearings and the arousal with respect to the control and fenoprofen salt groups. These impaired parameters in mice exposed to Cu₂(fen)₄(caf)₂ can be attributable to the presence of caffeine as stimulating agent. On the other hand, all exposed groups decreased the urine pools in the functional observational battery and increased the plasmatic urea. These effects could be due to the decrease in the glomerular filtration caused by NSAIDs. In conclusion, both complexes Cu₂(fen)₄(dmf)₂ and Cu₂(fen)₄(caf)₂ were more potent antinociceptive agents than fenoprofen salt. Sub-acute exposure to different doses of these complexes did not produce significant changes in the parameters that evaluate toxicity.Fil: Gumilar, Fernanda Andrea.     REFORMA UNIVERSITÁRIA NO BRASIL: UMA ANÁLISE DOS DOCUMENTOS OFICIAIS E DA PRODUÇÃO CIENTÍFICA SOBRE O REUNI - PROGRAMA DE APOIO A PLANOS DE REESTRUTURAÇÃO E EXPANSÃO DAS UNIVERSIDADES FEDERAIS

    Este estudo tem como objetivo analisar os aspectos relevantes e pertinentes ao Programa de Apoio a Planos de Reestruturação e Expansão das Universidades Federais – REUNI, que vêm sendo discutidos em documentos oficiais e artigos científicos durante o período de 2007 a 2010 Para tanto, pesquisou-se documentos disponíveis em sítios governamentais, tais como do Planalto, da Presidência, do Ministério da Educação e artigos científicos encontrados em diferentes sítios de busca, tais como: Scielo, Google Acadêmico e Domínio Público. Ademais, também foram pesquisadas revistas da área de Administração e de Educação, como: Revista Lua Nova, RAE, RAC, entre outras. Os documentos oficiais proporcionaram a contextualização das reformas da educação superior, principalmente no que se refere ao REUNI. Em relação às produções científicas, o estudo mostra certa desconfiança por parte dos pesquisadores em relação ao REUNI, em virtude da dificuldade relacionada ao financiamento da estrutura operacional e administrativa nas universidades em detrimento da manutenção da qualidade de ensino e da pesquisa e dos serviços de extensão; bem como a insuficiência de trabalhos científicos sobre o tema devido à sua recente implantação
