    DNA damage at the base-sequence, epigenome and chromosome level is a fundamental cause of developmental and degenerative diseases. Multiple micronutrients and their interactions with the inherited and/or acquired genome determine DNA damage and genomic instability rates. The challenge is to identify for each individual the combination of micronutrients and their doses (i.e. the nutriome) that optimises genome stability and DNA repair. In this paper I describe and propose the use of high-throughput nutrient array systems with high content analysis diagnostics of DNA damage, cell death and cell growth for defining, on an individual basis, the optimal nutriome for DNA damage prevention and cancer growth control

    A method for non-invasive genotyping of APCmin/+ mice using fecal samples

    The APCmin/+ mouse is commonly used in cancer research and is just one of many genetically altered models that is currently being developed. With high numbers of breeding programs, it is important to have a simple method that can be used to genotype the mice non-invasively. Here we report a reproducible method for genotyping mice with DNA extracted from fecal samples. Comparison of fecal results with those obtained from intestinal tissue DNA and clinical outcome (presence/absence of tumors) showed this technique to have 100% accuracy. This non-invasive method of genotyping may be applied to other transgenic mouse models

    A quantitative PCR method for measuring absolute telomere length

    We describe a simple and reproducible method to measure absolute telomere length (aTL) using quantitative real-time polymerase chain reaction (qPCR). This method is based on the Cawthon method for relative measurement of telomere length (TL) but modified by introducing an oligomer standard to measure aTL. The method describes the oligomer standards, the generation of the standard curve and the calculations required to calculate aTL from the qPCR data. The necessary controls and performance characteristics of the assay are described in detail and compared relative to other methods for measuring TL. Typical results for this assay for a variety of human tissue samples are provided as well as a troubleshooting schedule. This method allows high throughput measurement of aTL using small amounts of DNA making it amenable for molecular epidemiological studies. Compared to the traditional relative TL qPCR assays, the aTL method described in this protocol enables a more direct comparison of results between experiments within and between laboratories

    The area known as Tas-Silg is situated in the south-eastern part of the island of Malta, close to Marsaxlokk harbour. In reality the place name refers to the small church dedicated to Our Lady of the Snows (hence Tas-Silg) situated at the point where the narrow · road from Zejtun forks out in two directions: to Delimara and Xrobb il-Ghagin due south-east and to Marsaxlokk village due south-west. A British-period fort occupying the highest point of the elongated hill further south along· the first road also carries the same place name. The lower and more compact hill on which the excavations have been conducted is called 'Ta' Berikka' , but since it is so close to the above-mentioned church (only 50 m to the north) the tradition of calling it Tas-Silg is now well established and there is no sense in changing it. The site has a commanding view of the Marsaxlokk harbour to the south and overlooks two other bays, Marsascala and St Thomas's bay, to the north-east. On all sides the slope is broken up by man-made terraced fields There is no doubt that the topography of the site must have been a determining factor in its choice for the establishment of a religious centre in the Temple period of Maltese pehistory (3000--2500 BC), though one must keep in mind that close to Tas-Silg there are three other prehistoric temple sites. each one with a completely different topography. The Temple people were quite introverted in their cultural isolation and do not seem to have been much interested in seafaring and in the outside world. The situation changed radically in the following age, the Bronze Age. when the island was occupied by people who set up villages on naturally defensible hilltops, occasionally fortifying them with artificial ramparts. The Tas-Silg hill with its temple ruins was occupied by these people, but it is not as yet clear for what purpose. The scenario changed again in historical times when the central and western Mediterranean started to be parcelled out among the commercial powers originating in the eastern Mediterranean. The Greeks do not seem to have even tried 10 colonize Malta as they did in neighbouring Sicily. The Phoenicians, however, did occupy the island, apparently through a slow process of peaceful penetration and eventual political and cultural assimi lation. It was in this period that the ruins of the megalithic temple were transformed into a Phoenician extraurban shrine dedicated to Astarte, which in time expanded into a full y-fledged sanctuary with an international reputation. The last chapter in the millennia- long history of the site was written when the colonnaded courtyard in front of the old temple was transformed into an early Christian church. Any use made of the site in the following Arab period is, once again, poorly understood.peer-reviewe

    Biomarkers of Alzheimer\u27s disease risk in peripheral tissues; focus on buccal cells

    Alzheimer\u27s disease (AD) is a progressive degenerative disorder of the brain and is the most common form of dementia. To-date no simple, inexpensive and minimally invasive procedure is available to confirm with certainty the early diagnosis of AD prior to the manifestations of symptoms characteristic of the disease. Therefore, if population screening of individuals is to be performed, more suitable, easily accessible tissues would need to be used for a diagnostic test that would identify those who exhibit cellular pathology indicative of mild cognitive impairment (MCI) and AD risk so that they can be prioritized for primary prevention. This need for minimally invasive tests could be achieved by targeting surrogate tissues, since it is now well recognized that AD is not only a disorder restricted to pathology and biomarkers within the brain. Human buccal cells for instance are accessible in a minimally invasive manner, and exhibit cytological and nuclear morphologies that may be indicative of accelerated ageing or neurodegenerative disorders such as AD. However, to our knowledge there is no review available in the literature covering the biology of buccal cells and their applications in AD biomarker research. Therefore, the aim of this review is to summarize some of the main findings of biomarkers reported for AD in peripheral tissues, with a further focus on the rationale for the use of the buccal mucosa (BM) for biomarkers of AD and the evidence to date of changes exhibited in buccal cells with AD

    Aleurone flour increases red-cell folate and lowers plasma homocyst(e)ine substantially in man

    Aleurone flour (ALF) is a rich source of natural folate (>500 μg/100 g wet weight). Our objective was to establish whether intake of ALF in man can significantly improve folate status and reduce plasma homocyst(e)ine. We performed a randomised, controlled intervention, of 16 weeks duration, in free-living healthy individuals (mean age 46–52 years). Participants were assigned to one of three groups: ALF, 175 g bread made with ALF and placebo tablet each day; PCS, 175 g bread made with pericarp seed coat (PCS) flour and placebo tablet each day (low-folate control); or FA, 175 g bread made with PCS flour and tablet containing 640 μg folic acid each day (high-folate control). The daily folate intake contributed by the bread and tablet was 233 μg in the PCS group, 615 μg in the ALF group and 819 μg in the FA group. The number of participants completing all phases of the PCS, ALF and FA interventions was twenty-five, twenty-five and eighteen, respectively. Plasma and red-cell folate increased significantly (P<0·0001) and plasma homocyst(e)ine decreased significantly (P<0·0001) in the ALF and FA groups only. Plasma folate and red-cell folate in the ALF group (mean, 95 % CI) increased from baseline values of 12·9 (9·9, 15·7) nmol/l and 509 (434, 584) nmol/l to 27·1 (22·5, 31·7) nmol/l and 768 (676, 860) nmol/l, respectively. Plasma homocyst(e)ine in the ALF group decreased from 9·1 (8·2, 10·0) μmol/l at baseline to 6·8 (6·2, 7·5) μmol/l after 16 weeks. In conclusion, moderate dietary intake of ALF can increase red-cell folate and decrease plasma homocyst(e)ine substantially.Michael Fenech, Manny Noakes, Peter Clifton and David Toppin
