    Hepatocyte Lysosomal Membrane Stabilization by Olive Leaves against Chemically Induced Hepatocellular Neoplasia in Rats

    Extensive efforts are exerted looking for safe and effective chemotherapy for hepatocellular carcinoma (HCC). Specific and sensitive early biomarkers for HCC still in query. Present work to study proteolytic activity and lysosomal membrane integrity by hepatocarcinogen, trichloroacetic acid (TCA), in Wistar rats against aqueous olive leaf extract (AOLE).TCA showed neoplastic changes as oval- or irregular-shaped hepatocytes and transformed, vesiculated, and binucleated liver cells. The nuclei were pleomorphic and hyperchromatic. These changes were considerably reduced by AOLE. The results added, probably for the first time, that TCA-induced HCC through disruption of hepatocellular proteolytic enzymes as upregulation of papain, free cathepsin-D and nonsignificant destabilization of lysosomal membrane integrity, a prerequisite for cancer invasion and metastasis. AOLE introduced a promising therapeutic value in liver cancer, mostly through elevating lysosomal membrane integrity. The study substantiated four main points: (1) the usefulness of proteolysis and lysosomalmembrane integrity in early prediction of HCC. (2) TCA carcinogenesis is possibly mediated by lysosomal membrane destabilization, through cathepsin-D disruption, which could be reversed by AOLE administration. (3) A new strategy for management of HCC, using dietary olive leaf system may be a helpful phytotherapeutic trend. (4) A prospective study on serum proteolytic enzyme activity may introduce novel diagnostic tools

    Risk Factors of Epistaxis in Primary School Children in Dakahlia Governorate, Egypt

    Background: Epistaxis is a common pediatric complaint, so we study its risk factors trying to control and decrease it among primary school children. Objectives: To determine frequency of epistaxis and to assess the risk factors of epistaxis in primary school children (8-12 years old). Patients and Methods: A cross-sectional study that was conducted on 312 children. Sample was collected by systematic random technique from the children attending to Family Health Center, Sanafa, Mit-Ghamr city, Dakahlia Governorate, Egypt. Participants’ data regarding socio-demographic characteristics, and risk factors were collected via predesignedquestionnaire. Results: The study revealed that prevalence of epistaxis among the studied group was 32.4% and the most common risk factors among the studied group were smoking, head trauma, factory gases, chronic cough and upper respiratory tract infection (URTI) (56.7%, 45.8%, 35.6%, 32.7% & 30.1% respectively).Conclusion: Recurrent epistaxis can be troublesome and alarming for parents and children. so special attention must be given for it

    Characterization of anomalous Zeeman patterns in complex atomic spectra

    The modeling of complex atomic spectra is a difficult task, due to the huge number of levels and lines involved. In the presence of a magnetic field, the computation becomes even more difficult. The anomalous Zeeman pattern is a superposition of many absorption or emission profiles with different Zeeman relative strengths, shifts, widths, asymmetries and sharpnesses. We propose a statistical approach to study the effect of a magnetic field on the broadening of spectral lines and transition arrays in atomic spectra. In this model, the sigma and pi profiles are described using the moments of the Zeeman components, which depend on quantum numbers and Land\'{e} factors. A graphical calculation of these moments, together with a statistical modeling of Zeeman profiles as expansions in terms of Hermite polynomials are presented. It is shown that the procedure is more efficient, in terms of convergence and validity range, than the Taylor-series expansion in powers of the magnetic field which was suggested in the past. Finally, a simple approximate method to estimate the contribution of a magnetic field to the width of transition arrays is proposed. It relies on our recently published recursive technique for the numbering of LS-terms of an arbitrary configuration.Comment: submitted to Physical Review

    Relationship between Sap prevalence and biofilm formation among resistant clinical isolates of Candida albicans

    Background: Fungal infections represent a serious health problem especially in immunocompromised individuals. Candida albicans is the most common fungi that cause superficial and systemic infections with high mortality rates. Anti-fungal resistance of C. albicans may be attributed to its virulence. Biofilm formation and proteolytic activity are major virulence determents that may influence both pathogenicity and anti-fungal resistance of Candida albicans. Objective: This work studied the relation between biofilm formation, proteolytic activity and prevalence of some Sap genes with reduced susceptibility of C. albicans to different anti-fungal agents. Methods: Fifty three C. albicans strains isolated from patients with systemic infections, identified by germ tube, chromogenic agar and confirmed by PCR, were subjected to evaluate their proteolytic activity, the degree of biofilm production and the prevalence of Sap9 and Sap10 genes. The susceptibility of the isolates was determined by disk diffusion method against five antifungal drugs. Results and conclusion: Four of the C. albicans isolates were resistant to 3 anti-fungal drugs, strong biofilm producer, have proteolytic activity and contain either Sap9 or Sap10 or both. Conclusively, although anti-fungal resistance among the isolates was rare, a relation between the anti-fungal resistance and some major virulence factors was evidently proved in this study

    HEALING POTENCY OF HAEMATOCOCCUS PLUVIALIS EXTRACT FOR TREATING TYPE 2 DIABETES IN RATS

    Objective: The present study aims to evaluate the antidiabetic effect of ethanolic extract of Haematococcus pluvialis (H. pluvialis) in streptozotocin (STZ)-induced diabetic rats.Methods: The antidiabetic activity of H. pluvialis was investigated by the determination of glucose and insulin levels, aspartate (AST), alanine transaminases (ALT), lipid profile including total cholesterol (TC), triglycerides (TG), low-density lipoprotein-cholesterol (LDL-C) and high-density-lipoprotein-cholesterol (HDL-C). Histopathological examination of pancreas and liver were also carried out.Results: The results revealed that the levels of glucose, TC, TG, LDL-C as well as AST and ALT enzyme activities were increased significantly in diabetic rats. While, insulin and HDL-C levels decreased significantly in STZ-induced diabetic rats. The remediation of diabetic rats with H. pluvialis attenuated the elevated levels of glucose, TC, TG, LDL-C as well as AST and ALT activities in diabetic rats. Besides, it improved insulin, HDL-C levels, pancreas and hepatic architectures.Conclusion: H. pluvialis extract has a promising antidiabetic potency through attenuation of several metabolic disorders associated diabetes

    ASSOCIATION OF OBESITY WITH RS1421085 AND RS9939609 POLYMORPHISMS OF FTO GENE WITH T2DM IN EGYPTIAN FEMALES

    Objective: Obesity has been described as a worldwide increasing health problem and risk factor of various disorders including type 2 diabetes mellitus (T2DM). So, our study aim to determine of common variants of fat mass and obesity associated gene polymorphisms rs1421085 and rs 9939609; confers risk of obesity and type 2 diabetic mellitus in Egyptian females.Methods: In this population rs1421085 and rs9939609 polymorphisms of fat mass and obesity (FTO) gene were genotyped in 105 obese patients and 100 healthy controls with ages 14-60 y were collected from Medicine Specialized Hospital, Mansoura University, Egypt during the period between Jul.-Oct. 2016, genotyping of SNPs was performed by restriction fragment length polymorphism (RFLP) assay, fasting blood glucose, homeostasis model assessment of insulin resistance (HOMA IR), body mass index (BMI), waist-to-hip ratio (WHR) lipid profile was determined.Results: There was the significantly higher frequency of the AA compared to controls p=0.0001) of genotypers9939609. Also, cases have shown a significantly higher frequency of the C allele, p<0.00001) of rs1421085 genotype polymorphisms increased the risks of obesity. On the other hand, there were no significant correlations between genotypes and obesity-related (anthropometric body composition) parameters. Only the fasting blood glucose was significantly higher in the TA p=0.004).Conclusion: The FTO rs9939609 and rs1421085 single nucleotide polymorphisms (SNPs) was associated with increased risk of obesity in type 2 diabetic populations on Egyptian females

    Study of obesity associated proopiomelanocortin gene polymorphism: Relation to metabolic profile and eating habits in a sample of obese Egyptian children and adolescents

    AbstractBackgroundMelanocortinergic system represents a known system involved in the central regulation of body weight with the central proopiomelanocortin (POMC) neurons forming a potent anorexigenic network. Polymorphisms in the POMC gene locus are associated with obesity phenotypes.AimTo assess the contribution of the POMC gene 9-bp insertional polymorphism in the susceptibility to obesity and its relation to body mass index (BMI) and adiposity-related co-morbidities in obese children and adolescents; as well as binge eating behavior.Patients and methodsFifty obese children and adolescents with simple obesity were screened for Binge Eating Disorder (BED) by The Diagnostic and Statistical Manual of Mental Disorders, Fifth Edition (DSM-5), they were compared to 50 age, sex and pubertal stage-matched non obese controls. Anthropometric measurements, blood pressure, abdominal ultrasound for fatty liver, measurement of fasting lipid profile, fasting insulin, fasting blood glucose and assessment of POMC gene 9-bp insertional polymorphism were done.ResultsObese patients had significantly higher anthropometric measurements, blood pressure percentiles, fasting glucose, fasting insulin, homeostasis model assessment for insulin resistance (HOMA-IR) and fasting lipid profiles, and higher frequency of occurrence of non alcoholic fatty liver disease and BED. Allelic frequencies of POMC gene 9bp insertional polymorphism were comparable in patients and controls (p=0.956). Fasting insulin levels were significantly higher in the heterozygous cases having the polymorphism than in wild homozygous cases; whereas no difference was observed among the controls.ConclusionThis polymorphism was associated with higher fasting insulin levels in the obese patients only. These findings support the hypothesis that the melanocortin pathway may modulate glucose metabolism in obese subjects indicating a possible gene-environment interaction. POMC variant may be involved in the natural history of polygenic obesity, contributing to the link between type 2 diabetes and obesity
