    The Freshwater Biological Association at Wray Castle: Recollections of its first director

    In 1937 the Development Commission provided an annual grant to the Freshwater Biological Association to pay for a director and secretary. The author moved to the Lake District in the same year, and at that time T.T. Macan was working on invertebrates; K.R. Allen on fish; C.H. Mortimer on chemistry and physics of the aquatic environment, and Marie Rosenberg on phytoplankton. They were backed by George Thompson as laboratory assistant and Rosa Bullen as secretary. The work of the Association continued and expanded throughout the Second World War with some far-reached discoveries made. For example, the recovery of lake sediment cores and the examination of diatom remains, so starting the discipline of archaeo-limnology. Also, a hydrological survey of the Windermere catchment area found significant traces of sulphuric acid in rain gauges. This was more than 30 years before "acid rain" became fashionable

    A Recombinant Avian Infectious Bronchitis Virus Expressing a Heterologous Spike Gene Belonging to the 4/91 Serotype

    We have shown previously that replacement of the spike (S) gene of the apathogenic IBV strain Beau-R with that from the pathogenic strain of the same serotype, M41, resulted in an apathogenic virus, BeauR-M41(S), that conferred protection against challenge with M41 [1]. We have constructed a recombinant IBV, BeauR-4/91(S), with the genetic backbone of Beau-R but expressing the spike protein of the pathogenic IBV strain 4/91(UK), which belongs to a different serogroup as Beaudette or M41. Similar to our previous findings with BeauR-M41(S), clinical signs observations showed that the S gene of the pathogenic 4/91 virus did not confer pathogenicity to the rIBV BeauR-4/91(S). Furthermore, protection studies showed there was homologous protection; BeauR-4/91(S) conferred protection against challenge with wild type 4/91 virus as shown by the absence of clinical signs, IBV RNA assessed by qRT-PCR and the fact that no virus was isolated from tracheas removed from birds primarily infected with BeauR-4/91(S) and challenged with IBV 4/91(UK).     Characterisation of Clostridium difficile biofilm formation, a role for Spo0A.

    Clostridium difficile is a Gram-positive anaerobic, spore-forming bacillus that is the leading cause of nosocomial diarrhoea worldwide. We demonstrate that C. difficile aggregates and forms biofilms in vitro on abiotic surfaces. These polymicrobial aggregates are attached to each other and to an abiotic surface by an extracellular polymeric substance (EPS). The EPS matrix provides the scaffold bonding together vegetative cells and spores, as well as forming a protective barrier for vegetative cells against oxygen stress. The master regulator of sporulation, Spo0A, may play a key role in biofilm formation, as genetic inactivation of spo0A in strain R20291 exhibits decreased biofilm formation. Our findings highlight an important attribute of C. difficile pathogenesis, which may have significant implications for infection, treatment and relapse
