    Making the implicit explicit: raising pragmatic awareness in trainee interpreters, using semi-authentic spontaneous discourse samples

    Following the recent interest in the teaching of pragmatics and the recognition of its importance for both cross-cultural communication and new speakers of an additional language, the authors carried out an action research project to evaluate the effectiveness of a new approach to the teaching of pragmatics. This involved the use of semiauthentic discourse samples of New Zealand English to raise the pragmatic awareness of trainee interpreters in an undergraduate course taught by the first author. The researchers analysed qualitative data from learner blogs, written during instruction as part of the course requirements, for evidence that the learners noticed paralinguistic features used for pragmatic effect in conversations they overheard or took part in outside the classroom. The class teacher also kept a reflective journal recording her observations on learner progress and the usefulness of the materials. The researchers found there was a growth in the number of learners noticing pragmatic features during tuition, suggesting that the semiauthentic samples were effective as learning tools. Use of these samples also facilitated the noticing and discussion of cross-cultural differences and enabled more learner-centered methodology to be used

    Reflection and dialogue on postgraduate professional development for experienced language teachers

    Recent discussion has focused on the benefits and constraints of using and teaching reflection for professional self-development (Farrell, 2007; Volk, 2010). Alongside this is an interest in the value of dialogue in teacher development (for example Edge, 2007). This paper describes the experience of advanced language teachers participating in a reflective practice project undertaken as a paper in a professional master’s qualification in a New Zealand tertiary institution. Using data from teacher participant reflective essays and an end of course evaluation, the paper describes teachers’ growth in reflectivity and notes the role of dialogue in promoting professional development. The paper also explores the extent to which Stanley’s framework (1998) was useful in measuring levels of reflectivity. The researchers found that teachers believed the course promoted their professional development in several ways, and that the activities participants found most helpful were ones that contained an element of dialogic interaction. Participants’ level of reflectivity at the end of the course was high on Stanley’s (1998) framework, but it was necessary to modify parts of the framework for use in this context

    Teaching pragmatics to lower level learners

    The teaching and learning of the pragmatics (socio-culturally determined norms) of a second language is vital if migrants and refugees are to live and work in the country of resettlement without miscommunication (Yates, 2008). One way of teaching these is by having learners listen to and work with naturalistic samples of native speaker interaction (Burns & Joyce, 1997). Following a series of action research investigations into the teaching of pragmatic norms using elicited recorded samples of native speaker role-play at intermediate level (Denny, 2008), the authors have turned their attention to learners at lower levels of proficiency. There are indications that it may be difficult to use authentic or semi-authentic samples to teach at this lower level (Denny & Basturkmen, 2011). This project sought to discover if lower-level learners can learn pragmatics by being helped to notice pragmalinguistic features in recorded discourse samples created from native speaker role-play. The research showed that this is possible, but that the teaching methodology needed to be adapted to the needs of this group of learners, using more teacher-facilitated activities and scaffolding, and focussing on the teaching of formulaic expressions

    On the behaviour of lung tissue under tension and compression

    Lung injuries are common among those who suffer an impact or trauma. The relative severity of injuries up to physical tearing of tissue have been documented in clinical studies. However, the specific details of energy required to cause visible damage to the lung parenchyma are lacking. Furthermore, the limitations of lung tissue under simple mechanical loading are also not well documented. This study aimed to collect mechanical test data from freshly excised lung, obtained from both Sprague-Dawley rats and New Zealand White rabbits. Compression and tension tests were conducted at three different strain rates: 0.25, 2.5 and 25 min−1. This study aimed to characterise the quasi-static behaviour of the bulk tissue prior to extending to higher rates. A nonlinear viscoelastic analytical model was applied to the data to describe their behaviour. Results exhibited asymmetry in terms of differences between tension and compression. The rabbit tissue also appeared to exhibit stronger viscous behaviour than the rat tissue. As a narrow strain rate band is explored here, no conclusions are being drawn currently regarding the rate sensitivity of rat tissue. However, this study does highlight both the clear differences between the two tissue types and the important role that composition and microstructure can play in mechanical response

    Should Research Ethics Encourage the Production of Cost-Effective Interventions?

    This project considers whether and how research ethics can contribute to the provision of cost-effective medical interventions. Clinical research ethics represents an underexplored context for the promotion of cost-effectiveness. In particular, although scholars have recently argued that research on less-expensive, less-effective interventions can be ethical, there has been little or no discussion of whether ethical considerations justify curtailing research on more expensive, more effective interventions. Yet considering cost-effectiveness at the research stage can help ensure that scarce resources such as tissue samples or limited subject popula- tions are employed where they do the most good; can support parallel efforts by providers and insurers to promote cost-effectiveness; and can ensure that research has social value and benefits subjects. I discuss and rebut potential objections to the consideration of cost-effectiveness in research, including the difficulty of predicting effectiveness and cost at the research stage, concerns about limitations in cost-effectiveness analysis, and worries about overly limiting researchers’ freedom. I then consider the advantages and disadvantages of having certain participants in the research enterprise, including IRBs, advisory committees, sponsors, investigators, and subjects, consider cost-effectiveness. The project concludes by qualifiedly endorsing the consideration of cost-effectiveness at the research stage. While incorporating cost-effectiveness considerations into the ethical evaluation of human subjects research will not on its own ensure that the health care system realizes cost-effectiveness goals, doing so nonetheless represents an important part of a broader effort to control rising medical costs

    AID-Targeting and Hypermutation of Non-Immunoglobulin Genes Does Not Correlate with Proximity to Immunoglobulin Genes in Germinal Center B Cells

    Upon activation, B cells divide, form a germinal center, and express the activation induced deaminase (AID), an enzyme that triggers somatic hypermutation of the variable regions of immunoglobulin (Ig) loci. Recent evidence indicates that at least 25% of expressed genes in germinal center B cells are mutated or deaminated by AID. One of the most deaminated genes, c-Myc, frequently appears as a translocation partner with the Ig heavy chain gene (Igh) in mouse plasmacytomas and human Burkitt's lymphomas. This indicates that the two genes or their double-strand break ends come into close proximity at a biologically relevant frequency. However, the proximity of c-Myc and Igh has never been measured in germinal center B cells, where many such translocations are thought to occur. We hypothesized that in germinal center B cells, not only is c-Myc near Igh, but other mutating non-Ig genes are deaminated by AID because they are near Ig genes, the primary targets of AID. We tested this “collateral damage” model using 3D-fluorescence in situ hybridization (3D-FISH) to measure the distance from non-Ig genes to Ig genes in germinal center B cells. We also made mice transgenic for human MYC and measured expression and mutation of the transgenes. We found that there is no correlation between proximity to Ig genes and levels of AID targeting or gene mutation, and that c-Myc was not closer to Igh than were other non-Ig genes. In addition, the human MYC transgenes did not accumulate mutations and were not deaminated by AID. We conclude that proximity to Ig loci is unlikely to be a major determinant of AID targeting or mutation of non-Ig genes, and that the MYC transgenes are either missing important regulatory elements that allow mutation or are unable to mutate because their new nuclear position is not conducive to AID deamination

    Mouse models of neurodegenerative disease: preclinical imaging and neurovascular component.

    Neurodegenerative diseases represent great challenges for basic science and clinical medicine because of their prevalence, pathologies, lack of mechanism-based treatments, and impacts on individuals. Translational research might contribute to the study of neurodegenerative diseases. The mouse has become a key model for studying disease mechanisms that might recapitulate in part some aspects of the corresponding human diseases. Neurode- generative disorders are very complicated and multifacto- rial. This has to be taken in account when testing drugs. Most of the drugs screening in mice are very di cult to be interpretated and often useless. Mouse models could be condiderated a ‘pathway models’, rather than as models for the whole complicated construct that makes a human disease. Non-invasive in vivo imaging in mice has gained increasing interest in preclinical research in the last years thanks to the availability of high-resolution single-photon emission computed tomography (SPECT), positron emission tomography (PET), high eld Magnetic resonance, Optical Imaging scanners and of highly speci c contrast agents. Behavioral test are useful tool to characterize di erent ani- mal models of neurodegenerative pathology. Furthermore, many authors have observed vascular pathological features associated to the di erent neurodegenerative disorders. Aim of this review is to focus on the di erent existing animal models of neurodegenerative disorders, describe behavioral tests and preclinical imaging techniques used for diagnose and describe the vascular pathological features associated to these diseases

    Genetic drivers of heterogeneity in type 2 diabetes pathophysiology.

    Type 2 diabetes (T2D) is a heterogeneous disease that develops through diverse pathophysiological processes1,2 and molecular mechanisms that are often specific to cell type3,4. Here, to characterize the genetic contribution to these processes across ancestry groups, we aggregate genome-wide association study data from 2,535,601 individuals (39.7% not of European ancestry), including 428,452 cases of T2D. We identify 1,289 independent association signals at genome-wide significance (P < 5 × 10-8) that map to 611 loci, of which 145 loci are, to our knowledge, previously unreported. We define eight non-overlapping clusters of T2D signals that are characterized by distinct profiles of cardiometabolic trait associations. These clusters are differentially enriched for cell-type-specific regions of open chromatin, including pancreatic islets, adipocytes, endothelial cells and enteroendocrine cells. We build cluster-specific partitioned polygenic scores5 in a further 279,552 individuals of diverse ancestry, including 30,288 cases of T2D, and test their association with T2D-related vascular outcomes. Cluster-specific partitioned polygenic scores are associated with coronary artery disease, peripheral artery disease and end-stage diabetic nephropathy across ancestry groups, highlighting the importance of obesity-related processes in the development of vascular outcomes. Our findings show the value of integrating multi-ancestry genome-wide association study data with single-cell epigenomics to disentangle the aetiological heterogeneity that drives the development and progression of T2D. This might offer a route to optimize global access to genetically informed diabetes care
