    Incidence and severity of cassava mosaic disease in farmer’s fields in Ghana

    A survey of cassava mosaic disease (CMD) was carried out in Ghana from 2007-2008 to assess CMD incidence, infection type, severity and adult whitefly population. A total of 136 farmers’ fields across major cassava producing areas in the Brong Ahafo, Western, Northern, Ashanti and Volta regions were assessed. CMD was prevalent in most of the 136 fields surveyed. Frequently encountered local landraces were susceptible to the disease. CMD incidence reached 100% in farmers’ fields. Mean disease incidence ranged from 46% in the Ashanti region to 90% in the Western region of the country. CMD incidence averaged 66.0%, with cutting-borne infection and Whitefly-borne infections being 54.0% and 12.0%, respectively. CMD shoot symptom severity ranged from 2.0 to 3.7 in the farmers’ fields, with a mean of 2.9. Mean adult whitefly population was 0.47. The high prevalence of CMD requires a concerted effort in the management of CMD in the country

    Marker Assisted Foreground Selection for Identification of Striga Resistant Backcross Lines in Sorghum bicolor

    Striga is a major constraint to sorghum production causing high yield loss due to increasing infestation. Locally-adapted cultivars with resistant genes/QTLs could be an effective control strategy for Striga. Marker-Assisted Foreground Selection was used to select backcross lines possessing Striga resistance QTLs from N13. Marker polymorphism was conducted for the donor parent N13 and 10 recurrent parents using 10 Simple Sequence Repeat (SSR) markers. Recurrent parents with SSR alleles, polymorphic to the donor parent allele were selected. F1 lines were developed by making a cross between the selected recurrent parent and the donor. The F1 were confirmed for heterozygosity using SSR markers. Selected heterozygote F1s were backcrossed to their recurrent parent to develop backcross populations (BC1F1 and BC2F1). BC1F1 and BC2F1 populations were genotyped using SSR markers flanking the Striga resistant QTLs in N13. Forty two DANYANA-N13 BC2F1 lines (with 4 QTLs in 3 lines, 3 QTLs in 10 lines and other 28 lines having 1 to 2 QTLs) were selected for the presence of N13 QTLs. Forty three SAMSORG39-N13 BC2F1 lines (with 3 QTLs in 2 lines while 41 lines had 1 to 2 QTLs) were also selected for the presence of N13 QTLs. Although, selected lines will be genotyped for the recovery of recurrent parent background and evaluated to identify elite genotypes for possible release as varieties, the successful introgression of Striga resistance QTLs using Marker Assisted Selection suggests that in developing superior sorghum varieties, breeders could make use of molecular marker technologies to speed up breeding programmes

    Genetic Variation for Striga hermonthica Resistance and Yield Among Sorghum Accessions in Nigeria

    Striga hermonthica (Delile) Benth., commonly referred to as witch weed, is a major constraint to sorghum (Sorghum bicolor (L.) Moench) production in the Northern region of Nigeria because of high yield losses due to infestation. To identify parental lines useful in breeding for S. hermonthica resistant sorghum genotypes adapted to Nigeria, twenty-five sorghum accessions were evaluated in Nigeria across three test environments. Both phenotypic and genetic components influenced the variation observed in the sorghum accessions. The estimates for the genetic coefficient of variation, heritability and genetic advance for the area under Striga number progress curve (ASUNPC), Striga emergence counts, yield and other agronomic traits, obtained in this study revealed that genetic gain for resistance to S. hermonthica could be realized through selection. Based on the performance of the 25 sorghum accessions SRN39, Danyana, Sepon82, and SAMSORG40 were the top four accessions found to be most resistant to S. hermonthica. Assessment of resistance was based on the low Striga emergence counts and the ASUNPC values. These accessions can be used as donor sources of S. hermonthica resistant genes for introgression into cultivars adapted to Nigeria, followed by recombination breeding for pyramiding the different resistance mechanisms

    Genetic diversity assessment of sorghum (Sorghum bicolor (L.) Moench) accessions using single nucleotide polymorphism markers

    Sorghum (Sorghum bicolor (L.) Moench) is an important resource to the national economy and it is essential to assess the genetic diversity in existing sorghum germplasm for better conservation, utilization and crop improvement. The aim of this study was to evaluate the level of genetic diversity within and among sorghum germplasms collected from diverse institutes in Nigeria and Mali using Single Nucleotide Polymorphic markers. Genetic diversity among the germplasm was low with an average polymorphism information content value of 0.24. Analysis of Molecular Variation revealed 6%variation amonggermplasmand 94%withingermplasms.Dendrogramrevealed threegroupsof clusteringwhich indicatevariationswithinthegermplasms. Privatealleles identified inthesorghum accessions from National Center for Genetic Resources and Biotechnology, Ibadan, Nigeria and International Crop Research Institute for the Semi-Arid Tropics, Kano, Nigeria shows their prospect for sorghum improvement and discovery of new agronomic traits. The presence of private alleles and genetic variation within the germplasms indicates that the accessions are valuable resources for future breeding programs

    Genome-wide association study of resistance to cassava green mite pest and related traits in cassava

    Published: 26 July 2018Cassava green mite [CGM, Mononychellus tanajoa (Bondar)] is a dry-season pest that usually feeds on the underside of young leaves causing leaf chlorosis, stunted growth, and root yield reduction by 80%. Since cassava (Manihot esculenta Crantz) leaves and roots serve as a primary staple food source, a decline in cassava yield can lead to household food, nutrition, and income insecurity. To evaluate the existence of CGM resistance alleles in the available germplasm, a diversity panel of 845 advanced breeding lines obtained from IITA, CIAT, and the National Root Crops Research Institute (NRCRI) were evaluated for CGM severity (CGMS), leaf pubescence (LP), leaf retention (LR), stay green, shoot tip compactness, and shoot tip size. A genome-wide association mapping detected 35 single-nucleotide polymorphisms (SNPs) markers significantly associated with CGMS, LP, and LR on chromosome 8. Colocalization of the most significant SNP associated with CGMS, LP, and LR on chromosome 8 is possibly an indication of pleiotropy or the presence of closely linked genes that regulate these traits. Seventeen candidate genes were found to be associated to CGM resistance. These candidate genes were subdivided into seven categories according to their protein structure namely, Zn finger, pentatricopeptide, MYB, MADS, homeodomain, trichome birefringence-related protein, and ethylene-responsive transcription factor genes. This study revealed significant loci associated with CGM, not previously reported, which together represent potential sources for the ongoing effort to develop multiple pest- and disease-resistant cassava cultivars

    Genetic characterization of cassava (Manihot esculenta Crantz) genotypes using agro-morphological and single nucleotide polymorphism markers

    Open Access Article; Published online: 23 Dec 2019Dearth of information on extent of genetic variability in cassava limits the genetic improvement of cassava genotypes in Sierra Leone. The aim of this study was to assess the genetic diversity and relationships within 102 cassava genotypes using agro-morphological and single nucleotide polymorphism markers. Morphological classification based on qualitative traits categorized the germplasm into five different groups, whereas the quantitative trait set had four groups. The SNP markers classified the germplasm into three main cluster groups. A total of seven principal components (PCs) in the qualitative and four PCs in the quantitative trait sets accounted for 79.03% and 72.30% of the total genetic variation, respectively. Significant and positive correlations were observed between average yield per plant and harvest index (r = 0.76***), number of storage roots per plant and harvest index (r = 0.33*), height at first branching and harvest index (0.26*), number of storage roots per plant and average yield per plant (r = 0.58*), height at first branching and average yield per plant (r = 0.24*), length of leaf lobe and petiole length (r = 0.38*), number of leaf lobe and petiole length (r = 0.31*), width of leaf lobe and length of leaf lobe (r = 0.36*), number of leaf lobe and length of leaf lobe (r = 0.43*), starch content and dry matter content (r = 0.99***), number of leaf lobe and root dry matter (r = 0.30*), number of leaf lobe and starch content (r = 0.28*), and height at first branching and plant height (r = 0.45**). Findings are useful for conservation, management, short term recommendation for release and genetic improvement of the crop

    Genetic Diversification and Selection Strategies for Improving Sorghum Grain Yield Under Phosphorous-Deficient Conditions in West Africa

    Sorghum, a major crop for income generation and food security in West and Central Africa, is predominantly grown in low-input farming systems with serious soil phosphorus (P) deficiencies. This study (a) estimates genetic parameters needed to design selection protocols that optimize genetic gains for yield under low-phosphorus conditions and (b) examines the utility of introgressed backcross nested association mapping (BCNAM) populations for diversifying Malian breeding materials. A total of 1083 BC1F5 progenies derived from an elite hybrid restorer “Lata-3” and 13 diverse donor accessions were evaluated for yield and agronomic traits under contrasting soil P conditions in Mali in 2013. A subset of 298 progenies were further tested under low-P (LP) and high-P (HP) conditions in 2014 and 2015. Significant genetic variation for grain yield was observed under LP and HP conditions. Selection for grain yield under LP conditions was feasible and more efficient than the indirect selection under HP in all three years of testing. Several of the BCNAM populations exhibited yields under LP conditions that were superior to the elite restorer line used as a recurrent parent. The BCNAM approach appears promising for diversifying the male parent pool with introgression of diverse materials using both adapted Malian breed and unadapted landrace material from distant geographic origins as donors

    Genetic diversity of provitamin-A cassava (Manihot esculenta Crantz) in Sierra Leone

    Open Access Article; Published online: 04 Mar 2020Understanding the genetic diversity among accessions and germplasm is an important requirement for crop development as it allows for the selection of diverse parental combinations for enhancing genetic gain in varietal selection, advancement and release. The study aimed to characterize 183 provitamin A cassava (Manihot esculenta Crantz) accessions and five Sierra Leonean varieties using morphological traits, total carotenoid content and SNP markers to develop a collection for conservation and further use in the cassava breeding program. Both morphological parameters and 5634 SNP markers were used to assess the diversity among the provitamin-A cassava accessions and varieties. Significant differences were observed among the accessions for most of the traits measured. The first five PCs together accounted for 70.44% of the total phenotypic variation based on yield and yield components among the 183 provitamin-A cassava accessions and five Sierra Leonean varieties. The present study showed that provitamin-A cassava accessions in Sierra Leone have moderate to high diversity based on morphological and molecular assessment studies. The similarity index among the 187 and 185 cassava accessions grouped them into 6 and 9 distinct clusters based on morphological and molecular analyses, respectively. A significant positive, but low correlation (r = 0.104; p\0.034), was observed between the two dendrograms. The results obtained will serve as a guide and basis of germplasm management and improvement for total carotenoid content, yield and African cassava mosaic disease resistance in Sierra Leone

    Identification of stable genotypes and genotype by environment interaction for grain yield in sorghum (Sorghum bicolor L. Moench)

    Sorghum is a staple food crop in Niger and its production is constrained by sorghum midge and the use of low yielding, local sorghum varieties. To improve sorghum productivity, it is crucial to provide farmers with high yielding sorghum cultivars that are resistant to midge. We evaluated 282 genotypes in four environments of Niger Republic. Alpha (0.1) lattice with two replications was the experimental design. Genotype and genotype by environment (GGE) biplot analysis was used to study grain yield (GY) stability and G Ă— E interactions. The results revealed that two distinct mega environments were present. Genotype L232 was the best genotype for GY in the first planting date at Konni and the first and second planting dates (PDs) at Maradi. Genotype L17 was the best for GY in the second PD at Konni. The second PD at Konni was the most discriminating environment while the first PD at Konni is suitable for selecting widely adapted genotypes for GY

    Genetic Variability and Its Implications on Early Generation Sorghum Lines Selection for Yield, Yield Contributing Traits, and Resistance to Sorghum Midge

    Sorghum is the second most important cereal crop in Niger. The crop is grown in a wide range of ecological environments in the country. However, sorghum grain yield in Niger is limited by both abiotic and biotic constraints. Recombinant inbred lines derived from the cross of a local variety with a midge resistant variety and two local checks were evaluated during the 2015 rainy season across two planting dates in two environments in Niger. The objective was to investigate genetic variability for yield, yield related traits, and resistance to sorghum midge. High phenotypic coefficient of variation (PCV) versus genotypic coefficient of variation (GCV) was observed in both sites and planting dates. Across planting dates at both Konni and Maradi, grain yield, plant height, panicle weight, and midge damage had high heritability coupled with high estimates of genetic advance. At Konni, high genetic advance coupled with high heritability was detected for grain yield, plant height, panicle weight, and resistance to midge. There were similar results at Maradi for grain yield, plant height, and panicle weight. Therefore, selection might be successful for the above characters in their respective environments
