    Unveiling novel genes in fern genomes for the design of stress tolerant crops

    Ferns are the second largest vascular plant lineage after angiosperms, shaping the plant biodiversity on our planet for more than 360 million years. Understanding the biological function of fern genes and their potential uses for agriculture, environment and health has been hampered by the lack of high-quality genome sequence of fern. The last five years have seen the assembly of reference fern genomes of Salvinia cucullata (0.26 ​Gb, n ​= ​9), Azolla filiculoides (0.75 ​Gb, n ​= ​22), Adiantum capillus-veneris (4.83 ​Gb, n ​= ​30), Alsophila spinulosa (6.27 ​Gb, n ​= ​69) and Ceratopteris richardii (7.46 ​Gb, n ​= ​39). These high-quality genome sequences will make significant contribution to the better understanding of evolution, ecology, functional genomics, and the diverse applications of ferns. Here, I review the physiological and some aspects of limited molecular mechanisms of abiotic (mainly heavy metal, drought, and shade) and biotic (pests and pathogens) tolerance in different fern species. The potential use of the fern genome toward crop abiotic and biotic stress tolerance is suggested. Thus, deciphering the genetic diversity and novel functional genes of ferns will provide tremendous boost to the knowledge advancement and the biotechnological applications of unique fern genes and proteins for sustainable agricultural production and environmental protection

    Does molecular and structural evolution shape the speedy grass stomata?

    It has been increasingly important for breeding programs to be aimed at crops that are capable of coping with a changing climate, especially with regards to higher frequency and intensity of drought events. Grass stomatal complex has been proposed as an important factor that may enable grasses to adapt to water stress and variable climate conditions. There are many studies focusing on the stomatal morphology and development in the eudicot model plant Arabidopsis and monocot model plant Brachypodium. However, the comprehensive understanding of the distinction of stomatal structure and development between monocots and eudicots, especially between grasses and eudicots, are still less known at evolutionary and comparative genetic levels. Therefore, we employed the newly released version of the One Thousand Plant Transcriptome (OneKP) database and existing databases of green plant genome assemblies to explore the evolution of gene families that contributed to the formation of the unique structure and development of grass stomata. This review emphasizes the differential stomatal morphology, developmental mechanisms, and guard cell signaling in monocots and eudicots. We provide a summary of useful molecular evidences for the high water use efficiency of grass stomata that may offer new horizons for future success in breeding climate resilient crops

    Editorial: Regulation and manipulation of nutrient-controlling genes in crops

    Agriculture, food, and health sectors are largely disconnected because little has been done to consider the complex relationship between agricultural production, food consumption, human nutrition, health, and the food supply chain. As society develops, a healthy diet has becoming increasingly important for humans. Unhealthy diets and nutritional imbalance are highly correlated with several common chronic diseases, such as obesity, cardiovascular disease, and some cancers (Lee et al., 2011). The staples of a healthy diet usually come from crops, and the nutritional qualities of these crops, including the chemical composition of food crops, are essential considerations for a healthy diet. For example, the ratio between amylose and amylopectin in rice and the ratio between saturated and unsaturated fatty acids—particularly the percentage of oleic acid—are both related to human health (Goddard et al., 1984; Mozaffarian et al., 2006). Another major issue is the presence, or absence, of some nutritional components that are essential for human health, such as lysine in rice and tocopherol in the oils (Wang and Galili, 2016; Martin and Li, 2017). All of these require a better understanding of genetic regulation and the interaction between those genes encoding these traits (Martin and Li, 2017). This Research Topic focuses on the genetic control of important nutrients in crops, the identification of important QTLs combining large effective modern genetic tools such as GWAS analysis, multiple omics analysis on nutrients, gene-controlling nutrients function assays, and the manipulation of genes, such as gene transferring and genome editing. The aim of this Research Topic is to identify key genes and understand the genetic control of nutrients in order to provide healthy foods for humans. The Research Topic includes three original research articles, one systematic review, and one mini review on the subject of carbohydrate-containing and oil-bearing crops

    Stomata at the crossroad of molecular interaction between biotic and abiotic stress responses in plants

    Increasing global food production is threatened by harsh environmental conditions along with biotic stresses, requiring massive new research into integrated stress resistance in plants. Stomata play a pivotal role in response to many biotic and abiotic stresses, but their orchestrated interactions at the molecular, physiological, and biochemical levels were less investigated. Here, we reviewed the influence of drought, pathogen, and insect herbivory on stomata to provide a comprehensive overview in the context of stomatal regulation. We also summarized the molecular mechanisms of stomatal response triggered by these stresses. To further investigate the effect of stomata–herbivore interaction at a transcriptional level, integrated transcriptome studies from different plant species attacked by different pests revealed evidence of the crosstalk between abiotic and biotic stress. Comprehensive understanding of the involvement of stomata in some plant– herbivore interactions may be an essential step towards herbivores’ manipulation of plants, which provides insights for the development of integrated pest management strategies. Moreover, we proposed that stomata can function as important modulators of plant response to stress combination, representing an exciting frontier of plant science with a broad and precise view of plant biotic interactions

    Field application of silicon alleviates drought stress and improves water use efficiency in wheat

    Detrimental impacts of drought on crop yield have tripled in the last 50 years with climate models predicting that the frequency of such droughts will intensify in the future. Silicon (Si) accumulation, especially in Poaceae crops such as wheat (Triticum aestivum L.), may alleviate the adverse impacts of drought. We have very limited information, however, about whether Si supplementation could alleviate the impacts of drought under field conditions and no studies have specifically manipulated rainfall. Using field–based rain exclusion shelters, we determined whether Si supplementation (equivalent to 39, 78 and 117 kg ha-1) affected T. aestivum growth, elemental chemistry [Si, carbon (C) and nitrogen (N)], physiology (rates of photosynthesis, transpiration, stomatal conductance, and water use efficiency) and yield (grain production) under ambient and drought (50% of ambient) rainfall scenarios. Averaged across Si treatments, drought reduced shoot mass by 21% and grain production by 18%. Si supplementation increased shoot mass by up to 43% and 73% in ambient and drought water treatments, respectively, and restored grain production in droughted plants to levels comparable with plants supplied with ambient rainfall. Si supplementation increased leaf-level water use efficiency by 32–74%, depending on Si supplementation rates. Water supply and Si supplementation did not alter concentrations of C and N, but Si supplementation increased shoot C content by 39% and 83% under ambient and drought conditions, respectively. This equates to an increase from 6.4 to 8.9 tonnes C ha-1 and from 4.03 to 7.35 tonnes C ha-1 under ambient and drought conditions, respectively. We conclude that Si supplementation ameliorated the negative impacts of drought on T. aestivum growth and grain yield, potentially through its beneficial impacts on water use efficiency. Moreover, the beneficial impacts of Si on plant growth and C storage may render Si supplementation a useful tool for both drought mitigation and C sequestration

    Sustainable protected cropping : a case study of seasonal impacts on greenhouse energy consumption during capsicum production

    Sustainable food production in protected cropping is increasing rapidly in response to global climate change and population growth. However, there are significant knowledge gaps regarding energy consumption while achieving optimum environmental conditions for greenhouse crop production. A capsicum crop cultivated in a high-tech greenhouse facility in Australia was analysed in terms of relationships between key environmental variables and the comparative analysis of energy consumption during different seasons. We showed that daily energy consumption varied due to the seasonal nature of the external environment and maintenance of optimal growing temperatures. Total power consumption reported throughout the entire crop cycle for heating (gas hot water system) and cooling (pad and fan) was 12,503 and 5183 kWh, respectively; hence, heating consumed ca. 70% of the total energy requirement over the 8-month growing period (early spring to late autumn) in the greenhouse facility. Regressions of daily energy consumption within each season, designated either predominantly for heating or cooling, indicated that energy consumption was 14.62 kWh per 1 °C heating and 2.23 kWh per 1 °C cooling. Therefore, changing the planting date to late spring is likely to significantly reduce heating energy costs for greenhouse capsicum growers in Australia. The findings will provide useful guidelines to maximise the greenhouse production of capsicum with better economic return by taking into consideration the potential optimal energy saving strategy during different external environment conditions and seasons

    Molecular evolution of calcium signaling and transport in plant adaptation to abiotic stress

    Adaptation to unfavorable abiotic stresses is one of the key processes in the evolution of plants. Calcium (Ca2+) signaling is characterized by the spatiotemporal pattern of Ca2+ distribution and the activities of multi-domain proteins in integrating environmental stimuli and cellular responses, which are crucial early events in abiotic stress responses in plants. However, a comprehensive summary and explanation for evolutionary and functional synergies in Ca2+ signaling remains elusive in green plants. We review mechanisms of Ca2+ membrane transporters and intracellular Ca2+ sensors with evolutionary imprinting and structural clues. These may provide molecular and bioinformatics insights for the functional analysis of some non-model species in the evolutionarily important green plant lineages. We summarize the chronological order, spatial location, and characteristics of Ca2+ functional proteins. Furthermore, we highlight the integral functions of calcium-signaling components in various nodes of the Ca2+ signaling pathway through conserved or variant evolutionary processes. These ultimately bridge the Ca2+ cascade reactions into regulatory networks, particularly in the hormonal signaling pathways. In summary, this review provides new perspectives towards a better understanding of the evolution, interaction and integration of Ca2+ signaling components in green plants, which is likely to benefit future research in agriculture, evolutionary biology, ecology and the environment

    Stress signaling convergence and nutrient crosstalk determine zinc-mediated amelioration against cadmium toxicity in rice

    Consumption of rice (Oryza sativa L.) is one of the major pathways for heavy metal bioaccumulation in humans over time. Understanding the molecular responses of rice to heavy metal contamination in agriculture is useful for eco-toxicological assessment of cadmium (Cd) and its interaction with zinc (Zn). In certain crops, the impacts of Cd stress or Zn nutrition on the biophysical chemistry and gene expression have been widely investigated, but their molecular interactions at transcriptomic level, particularly in rice roots, are still elusive. Here, hydroponic investigations were carried out with two rice genotypes (Yinni-801 and Heizhan-43), varying in Cd contents in plant tissues to determine their transcriptomic responses upon Cd15 (15 µM) and Cd15+Zn50 (50 µM) treatments. High throughput RNA-sequencing analysis confirmed that 496 and 2407 DEGs were significantly affected by Cd15 and Cd15+Zn50, respectively, among which 1016 DEGs were commonly induced in both genotypes. Multitude of DEGs fell under the category of protein kinases, such as calmodulin (CaM) and calcineurin B-like protein-interacting protein kinases (CBL), indicating a dynamic shift in hormonal signal transduction and Ca2+ involvement with the onset of treatments. Both genotypes expressed a mutual regulation of transcription factors (TFs) such as WRKY, MYB, NAM, AP2, bHLH and ZFP families under both treatments, whereas genes econding ABC transporters (ABCs), high affinity K+ transporters (HAKs) and Glutathione-S-transferases (GSTs), were highly up-regulated under Cd15+Zn50 in both genotypes. Zinc addition triggered more signaling cascades and detoxification related genes in regulation of immunity along with the suppression of Cd-induced DEGs and restriction of Cd uptake. Conclusively, the effective integration of breeding techniques with candidate genes identified in this study as well as economically and technologically viable methods, such as Zn nutrient management, could pave the way for selecting cultivars with promising agronomic qualities and reduced Cd for sustainable rice production

    Capsicum waste as a sustainable source of capsaicinoids for metabolic diseases

    Capsaicinoids are pungent alkaloid compounds enriched with antioxidants, anti-microbial, anti-inflammatory, analgesics, anti-carcinogenic, anti-obesity and anti-diabetic properties. These compounds are primarily synthesised in the placenta of the fruit and then transported to other vegetative parts. Different varieties of capsicum and chillies contain different capsaicinoid concentrations. As capsicums and chillies are grown extensively throughout the world, their agricultural and horticultural production leads to significant amount of waste generation, in the form of fruits and plant biomass. Fruit wastes (placenta, seeds and unused fruits) and plant biowaste (stems and leaves) can serve as sources of capsaicinoids which can provide opportunities to extract these compounds for development of nutraceutical products using conventional or advanced extraction techniques. Capsaicin and dihydrocapsaicin are two most abundantly found pungent compounds. Considering the health benefits of capsaicinoids, these compounds can help in reducing metabolic disease complications. The development of an advanced encapsulation therapy of safe and clinically effective oral capsaicinoid/capsaicin formulation seem to require evaluation of strategies to address challenges related to the dosage, limited half-life and bioavailability, adverse effects and pungency, and the impacts of other ligands antagonising the major capsaicinoid receptor

    Molecular evolution and interaction of membrane transport and photoreception in plants

    Light is a vital regulator that controls physiological and cellular responses to regulate plant growth, development, yield, and quality. Light is the driving force for electron and ion transport in the thylakoid membrane and other membranes of plant cells. In different plant species and cell types, light activates photoreceptors, thereby modulating plasma membrane transport. Plants maximize their growth and photosynthesis by facilitating the coordinated regulation of ion channels, pumps, and co-transporters across membranes to fine-tune nutrient uptake. The signal-transducing functions associated with membrane transporters, pumps, and channels impart a complex array of mechanisms to regulate plant responses to light. The identification of light responsive membrane transport components and understanding of their potential interaction with photoreceptors will elucidate how light-activated signaling pathways optimize plant growth, production, and nutrition to the prevailing environmental changes. This review summarizes the mechanisms underlying the physiological and molecular regulations of light-induced membrane transport and their potential interaction with photoreceptors in a plant evolutionary and nutrition context. It will shed new light on plant ecological conservation as well as agricultural production and crop quality, bringing potential nutrition and health benefits to humans and animals
