    Study of SREBP1 and SERBP2 in peripheral blood mononuclear cells fron hypercholesterolemic rabbits

    Peripheral blood mononuclear cells are a possible biomarker that could reveal molecular alterations before the development of the disease. Therefore, the objective of the present investigation is to study molecular and genetic changes that indicate metabolic modifications even with normal biochemical values. SERBP1 and SERBP2 are proteins involved in lipid metabolic. These molecules can be expressed in peripheral blood mononuclear cells. This allows study tissue changes without resorting to biopsies. In this study, one control group of New Zealand rabbits was fed with a balanced feed (C) and another group received the same balanced feed supplemented with 17% fat (F). These animals did not receive fructose overload, maintaining constant concentrations of carbohydrates and protein in both groups. In biochemical tests from both groups were observed similar levels of glucose (C group: 140.7 + 28.4 mg/dl / F group: 118.3 +12.0 mg/dl) and triglyceride (C group: 144.1 +15.5 mg/dl / F group: 135.6 +8.3 mg/dl), while F group showed increased levels of cholesterol (42.8 +21.6mg/dl) compared with C group (27.1 +4.5 mg/dl). However, there is variability in the cholesterol values because some animals of the F group do not experience significant increment despite the intake of fat. This interesting finding leads to the hypothesis that changes in lipid metabolism can be examined by the expression of different genes early. As preliminary results, we observed by immunohistochemistry the presence nuclear of SREBP1 and SREBP2 in lymphocytes of the F group, while in C group was not observed immunoreaction or cytoplasmic signal. This result is indicating an activation of the lipid metabolism before being able to observe changes at a biochemical level.Fil: Saez, Aldana.     La asociación epididimaria de espermatozoides es un fenómenoobservado en numerosas especies de mamíferos. Cuando los espermatozoides madurosalcanzan la cola del epidídimo pueden asociarse entre sí mediante sus cabezasformando estructuras características, cuya morfología depende de cada especie. Entrelas cabezas asociadas se describió la presencia de un material electron denso denaturaleza proteica. En la rata estas asociaciones se denominan ”rosetas“. De estemodo, los espermatozoides permanecen almacenados hasta la eyaculación. Una vezdepositados en el tracto reproductor femenino o bien liberados a un medio de cultivo seactiva la movilidad espermática. Esto produce que uno a uno los espermatozoides sedesprendan originando una suspensión de células móviles y aisladas. En las especiesde mamíferos estudiadas, este proceso de asociación reversible se relaciona con elgrado de maduración alcanzado por los espermatozoides. En cuanto a la funciónbiológica se ha propuesto tanto la protección de las gametas frente a una reacciónacrosomal anticipada como la adquisición de una una mayor velocidad dedesplazamiento de las asociaciones lo que supondría ventaja en caso de competenciaespermática. En trabajos previos nuestro grupo aisló e identifico en el fluido epididimariode la región caudal de rata la proteína Serpina 1 F que pertenece a la familia de lasSerpinas (inhibidores de serin proteasas). Luego demostramos que Serpina 1Fparticipaba en la asociación epididimaria de espermatozoides en esta especie medianteensayos de re asociación in vitro

    Efecto del aceite de oliva sobre el metabolismo del colesterol en conejos hipercolesterolémicos.

    El aceite de oliva, componente esencial de la Dieta Mediterránea, posee efectosbeneficiosos sobre la hipercolesterolemia, principal factor de riesgo de enfermedadescardiovasculares. La administración de una dieta grasa en conejos provoca laacumulación de colesterol en la membrana celular del espermatozoide y se correlacionacon defectos en la biogénesis del acrosoma y de la cabeza en general. Este incrementode colesterol interfiere con el normal funcionamiento de la gameta masculina y podría sercausa de infertilidad masculina. Interesantemente, la administración de aceite de oliva aestos animales mejoró los parámetros alterados por la ingesta grasa.Nuestro interés reside en estudiar la vía intracelular de colesterol en testículo deconejos bajo diferentes dietas experimentales.     Hypercholesterolemia Impaired Sperm Functionality in Rabbits

    Hypercholesterolemia represents a high risk factor for frequent diseases and it has also been associated with poor semen quality that may lead to male infertility. The aim of this study was to analyze semen and sperm function in diet-induced hypercholesterolemic rabbits. Twelve adult White New Zealand male rabbits were fed ad libitum a control diet or a diet supplemented with 0.05% cholesterol. Rabbits under cholesterol-enriched diet significantly increased total cholesterol level in the serum. Semen examination revealed a significant reduction in semen volume and sperm motility in hypercholesterolemic rabbits (HCR). Sperm cell morphology was seriously affected, displaying primarily a “folded head”-head fold along the major axe-, and the presence of cytoplasmic droplet on sperm flagellum. Cholesterol was particularly increased in acrosomal region when detected by filipin probe. The rise in cholesterol concentration in sperm cells was determined quantitatively by Gas chromatographic-mass spectrometric analyses. We also found a reduction of protein tyrosine phosphorylation in sperm incubated under capacitating conditions from HCR. Interestingly, the addition of Protein Kinase A pathway activators -dibutyryl-cyclic AMP and iso-butylmethylxanthine- to the medium restored sperm capacitation. Finally, it was also reported a significant decrease in the percentage of reacted sperm in the presence of progesterone. In conclusion, our data showed that diet-induced hypercholesterolemia adversely affects semen quality and sperm motility, capacitation and acrosomal reaction in rabbits; probably due to an increase in cellular cholesterol content that alters membrane related events

    Chemical, sensory and biological characteristics of selected monovarietal extra virgin olive oils

    The present study deals with the chemical, biological and sensory characterization of monovarietal extra virgin olive oils (EVOOs) obtained from cultivar grown in Marche region (Italy). EVOO is a food product having interesting beneficial properties, as evidenced by scientific literature [1]. One of the key features contributing to these properties are phenolic substances, which are related to the healthy, sensory and stability properties of EVOOs. They have been also related to the prebiotic activity of the oil [2]. Also volatile substances greatly contribute to the overall quality of EVOO and they are produced in chemical and biochemical pathways whose outcome depends on several factors, one of the most important of which, is the olives health state, and thus the microorganisms present. This study aims to investigate the relations between the chemical characteristics of monovarietal EVOOs mainly related to the overall quality and to correlate them with sensorial properties, prebiotic activity and yeasts characterizing the oils. Because of the influences of yeasts on the final EVOO quality, the high-throughput sequencing of internal transcribed spacers amplicons was employed to study the yeast biodiversity associated to the monovarietal EVOOs. Significant differences in taxa abundance among samples were obtained. Regarding prebiotic activity, it has been investigated by a semi-continuous fermentation system inoculated with a pool of human faeces under anaerobic conditions, to mime a colonic human fermentation [3]. Bifidobacteria, lactobacilli and clostridia and short chain fatty acids have been monitored using RT-PCR and gas-chromatography respectively, during 24h fermentation of each previously digested EVOO.     Chemical and sensory profiling of monovarietal extra virgin olive oils from the italian marche region

    Chemical and sensory peculiarities of monovarietal extra virgin olive oils (MEVOOs) from the cultivars (cvs.) Ascolana tenera (ASC), Coroncina (COR), Mignola (MIG), Piantone di Mogliano (MOG), and Raggia (RAG) from Marche region (Italy) are investigated. Their polar phenolic substances and α-tocopherol are analysed through high performance liquid chromatography with different detectors. Volatile substances, fatty acid composition, and squalene are analysed by gas chromatography coupled to mass spectrometry (MS) and to the flame ionization detector, respectively. Total antioxidant activity and sensory analysis were also performed. MOG showed high squalene content (on average 0.88 ± 0.16 g/100 g), high relative amount of α-copaene among volatiles, and the highest oleic acid percentage. MIG had high α-tocopherol content (on average 350.0 ± 57.6 mg kg−1) and high α-farnesene in the volatile fraction. ASC showed the highest sensory quality and the lignan pinoresinol with higher concentration as compared to the other MEVOOs (p < 0.05), which resulted in a possible chemical marker for this cv. RAG was characterized by the sensory note of almond, which corresponds to its highest (E)-2-hexenal percentage. Sensory analysis and an antioxidant activity assay performed on a set of industrial extra virgin olive oils purchased in supermarkets, highlighted MEVOOs’ superiority from these points of view.
