    We have designed and synthesized a novel zinc trisporphyrinate with a benzene tricarboxamide as the linker. In the presence of a large excess of 1-phenylethylamine, single crystals of the corresponding 1:3 host–guest complex were obtained, which provide the crystallographic structure of a host–guest complex consisting of an achiral porphyrin and a chiral monoamine. The structure reveals the 1-phenylethylamines adopt the “inside” binding mode that is stabilized by intramolecular hydrogen bonds. The NH<sub>2</sub> of the 1-phenylethylamine is involved in both coordination and hydrogen bonding interactions. Circular dichroism (CD) and ultraviolet–visible spectra revealed that the 1:3 host–guest complex is dominant in the presence of a large excess of 1-phenylethylamine. The crystal structure shows there are two diastereomers of the 1:3 host–guest complexes. Density functional theory and TDDFT calculations suggest that one of the diastereomers is more energetically favorable, which dominates the CD signals

    ObjectiveDiabetic nephropathy (DN) is a major microvascular complication of diabetes and the leading cause of end-stage renal disease. Early detection and prevention of DN are important. Retinol-binding protein 4 (RBP4) has been considered as a single diagnostic marker for the detection of renal impairment. However, the results have been inconsistent. The present meta-analysis aimed to determine the diagnostic potential of RBP4 in patients in type 2 diabetes mellitus (T2DM) with DN.MethodsWe searched PubMed, Web of Science, Embase, Wanfang and CNKI databases from inception until January 2024. The meta-analysis was performed by Stata version 15.0, and sensitivity, specificity, positive and negative likelihood ratios (PLR and NLR), diagnostic odds ratio (DOR) and area under the curve (AUC) were pooled. The Quality Assessment of Diagnostic Accuracy Studies-2 tool was utilized to assess the quality of each included study. In addition, heterogeneity and publication bias were evaluated.ResultsTwenty-nine studies were included in the meta-analysis. The pooled sensitivity and specificity were 0.76 [95% confidence interval (CI), 0.71–0.80] and 0.81 (95% CI, 0.76–0.85), respectively. The results showed a pooled PLR of 4.06 (95% CI, 3.16–5.21), NLR of 0.29 (95% CI, 0.24–0.36) and DOR of 13.76 (95% CI, 9.29–20.37). The area under the summarized receiver operating characteristic curve was given a value of 0.85 (95% CI, 0.82–0.88). No obvious publication bias existed in the Deeks’ funnel plot asymmetry test.ConclusionOur findings suggest that RBP4 has a promising diagnostic value with good sensitivity and specificity for patients with T2DM with DN.</p

    Overexpression of <i>HvHGGT</i> Enhances Tocotrienol Levels and Antioxidant Activity in Barley

    Vitamin E is a potent lipid-soluble antioxidant and essential nutrient for human health. Tocotrienols are the major form of vitamin E in seeds of most monocots. It has been known that homogentisate geranylgeranyl transferase (HGGT) catalyzes the committed step of tocotrienol biosynthesis. In the present study, we generated transgenic barley overexpressing <i>HvHGGT</i> under endogenous D-Hordein promoter (<i>proHor</i>). Overexpression of <i>HvHGGT</i> increased seed size and seed weight in transgenic barley. Notably, total tocotrienol content increased by 10–15% in seeds of transgenic lines, due to the increased levels of δ-, β-, and γ-tocotrienol, but not α-tocotrienol. Total tocopherol content decreased by 14–18% in transgenic lines, compared to wild type. The antioxidant activity of seeds was determined by using 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2′-azinobis­(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS), and lipid peroxidation assays. Compared to wild type, radical scavenging activity of seed extracts was enhanced by 17–18% in transgenic lines. Meanwhile, the lipid peroxidation level was decreased by about 20% in transgenic barley seeds.
