    Dado el carácter de "primitivismo" del Bovino Criollo, motivo de esta investigación, se realiza un somero estudio filogenético a los efectos de ubicar este tipo de ganado en estudios inmunogénéticos futuros vinculados a poblaciones de hábitats regionales, en correspondencia a distintas zonas de la República Argentina y países limítrofes. En base al origen Ibérico común del Longhorn Americano u Bovino Criollo se hace una primera investigación tentativa con animales de la Estación Experimental Agropecuaria Famaillá (INTA) de Leales, Tucumán, orientada a verificar la existencia de algunos "marcadores genéticos" que fueran coincidentes con los descubiertos por MILLER en Longhorn (Miller, 1966). Esta primera etapa permitió comprobar 15 fenogrupos del Sistema B, involucrados en el total de 27 detectados por MILLER. En general hay acuerdo respecto a los otros sistemas, con pequeñas diferencias de las frecuencias génicas en algunos casos.Given the primitivism of the character of the Creole Bovine, we made a superficial phylogenic study to place this type of cattle in future imnunnogenetics studies which will involve regional habitat populations of the Argentine Republic and bounding countries. Because of the common Iberian origin of the American Longhorn and "Bovino Criollo", a first tentative investigation has been performed with animals of the Estación Experimental Agropecnaria Famaillá (Leales, Tucumán) to verify the existence of some "genetic markers" that would be coincident with MILLER's disclosure in Longhorns (Miller, 1966).     Identification of SNPs and INDELS in swine transcribed sequences using short oligonucleotide microarrays

    <p>Abstract</p> <p>Background</p> <p>Genome-wide detection of single feature polymorphisms (SFP) in swine using transcriptome profiling of day 25 placental RNA by contrasting probe intensities from either Meishan or an occidental composite breed with Affymetrix porcine microarrays is presented. A linear mixed model analysis was used to identify significant breed-by-probe interactions.</p> <p>Results</p> <p>Gene specific linear mixed models were fit to each of the log<sub>2 </sub>transformed probe intensities on these arrays, using fixed effects for breed, probe, breed-by-probe interaction, and a random effect for array. After surveying the day 25 placental transcriptome, 857 probes with a q-value ≤ 0.05 and |fold change| ≥ 2 for the breed-by-probe interaction were identified as candidates containing SFP. To address the quality of the bioinformatics approach, universal pyrosequencing assays were designed from Affymetrix exemplar sequences to independently assess polymorphisms within a subset of probes for validation. Additionally probes were randomly selected for sequencing to determine an unbiased confirmation rate. In most cases, the 25-mer probe sequence printed on the microarray diverged from Meishan, not occidental crosses. This analysis was used to define a set of highly reliable predicted SFPs according to their probability scores.</p> <p>Conclusion</p> <p>By applying a SFP detection method to two mammalian breeds for the first time, we detected transition and transversion single nucleotide polymorphisms, as well as insertions/deletions which can be used to rapidly develop markers for genetic mapping and association analysis in species where high density genotyping platforms are otherwise unavailable.</p> <p>SNPs and INDELS discovered by this approach have been publicly deposited in NCBI's SNP repository dbSNP. This method is an attractive bioinformatics tool for uncovering breed-by-probe interactions, for rapidly identifying expressed SNPs, for investigating potential functional correlations between gene expression and breed polymorphisms, and is robust enough to be used on any Affymetrix gene expression platform.</p

    Graphene growth on h-BN by Molecular Beam Epitaxy

    The growth of single layer graphene nanometer size domains by solid carbon source molecular beam epitaxy on hexagonal boron nitride (h-BN) flakes is demonstrated. Formation of single-layer graphene is clearly apparent in Raman spectra which display sharp optical phonon bands. Atomic-force microscope images and Raman maps reveal that the graphene grown depends on the surface morphology of the h-BN substrates. The growth is governed by the high mobility of the carbon atoms on the h-BN surface, in a manner that is consistent with van der Waals epitaxy. The successful growth of graphene layers depends on the substrate temperature, but is independent of the incident flux of carbon atoms.Comment: Solid State Communications, 201

    Como apoyo complementario a la metodología de la Inmunogenética en investigaciones el grupos sanguíneos eritrocitarios, la genética bioquímica contribuye con varias leas de trabajo, entre otras: la detección de grupos Serogenéticos referidos a Transferrinas, albúminas e Hemoglobinas. En este aspecto "genético bioquímico" se investigan en Bovinos Criollos de Estación Experimental Agropecuaria de Leales (INTA), Tucumán. los fenotipos de Transferrinas. Albúmina y Hemoglobina, llegando a comprobaciones iniciales de interés por ejemplo el alele Trausferrina E (TfÉ ) no tiene ocurrencia en criollos (en este primer estudio) 1 el alele Albúmina S (AlS ) no aparece en homicigosis, y, en Hemoglobinas se encuentra un franco predominio del alele HbA sobre HbB , con reducida frecuencia del genotipo homocigótico HbB /HbB , no observado por MILLER en Longhorn Americanos. Relativo a la conjunción de los trabajos 1 y 2 es de enfatizar las pautas de indagación sobre los sistemas de grupos sanguíneos y serogenéticos en la secuencia metodológica de la Inmunogenética Animal, recalcando su importancia como Ciencias Aplicada. La utilización de esta metodología de investigación puede contribuir a la certificación individual, fundamentalmente, para la estructuración de rodeos puros constitutivos de grandes reservorios gené- ticos a los efectos de mantener intacta y preservado ese gran patrimonio genético concerniente al Bovino Criollo, ya prácticamente en vías de extinción.As a complementary test to the immunogenetics for the blood groups research, the Biochemical Genetics contributes with several work lines to detect the Seroenetic groups, example, Transferrins, Albumins and Hemoglobins. The investigation is performed in "Bovinos Criollos" of the Estación Experimental Agropecuaria de Leales (INTA), Tucumán, researching the Transferrin, Albumin and Hemoglobin phenotypes. As a first result, in this study we have observed that the TfÉ does not occur in "Criollo", the alele AlbS does not appear as homozygous and the Hemoglobin types gave the major frequency for the HbA with reduced frequency for the homozygous type HbB /HbB . The use of the Animal Immunogenetics with its methods can contribute to the cattle individual certification, fundamentally to structure the "Creole Bovine" herds as large reserves in order to maintain intact and preserved that great patrimony of genes concerning to the "Bovino Criollo" which practically is the way of extintion.Facultad de Ciencias Veterinaria

    Detection of transcriptional difference of porcine imprinted genes using different microarray platforms

    BACKGROUND: Presently, multiple options exist for conducting gene expression profiling studies in swine. In order to determine the performance of some of the existing microarrays, Affymetrix Porcine, Affymetrix Human U133+2.0, and the U.S. Pig Genome Coordination Program spotted glass oligonucleotide microarrays were compared for their reproducibility, coverage, platform independent and dependent sensitivity using fibroblast cell lines derived from control and parthenogenic porcine embryos. RESULTS: Array group correlations between technical replicates demonstrated comparable reproducibility in both Affymetrix arrays. Glass oligonucleotide arrays showed greater variability and, in addition, approximately 10% of probes had to be discarded due to slide printing defects. Probe level analysis of Affymetrix Human arrays revealed significant variability within probe sets due to the effects of cross-species hybridization. Affymetrix Porcine arrays identified the greatest number of differentially expressed genes amongst probes common to all arrays, a measure of platform sensitivity. Affymetrix Porcine arrays also identified the greatest number of differentially expressed known imprinted genes using all probes on each array, an ad hoc measure of realistic performance for this particular experiment. CONCLUSION: We conclude that of the platforms currently available and tested, the Affymetrix Porcine array is the most sensitive and reproducible microarray for swine genomic studies

    Responsiveness of the Eating Disorders Quality of Life Scale (EDQLS) in a longitudinal multi-site sample

    <p>Abstract</p> <p>Background</p> <p>In eating disorders (EDs), treatment outcome measurement has traditionally focused on symptom reduction rather than functioning or quality of life (QoL). The Eating Disorders Quality of Life Scale (EDQLS) was recently developed to allow for measurement of broader outcomes. We examined responsiveness of the EDQLS in a longitudinal multi-site study.</p> <p>Methods</p> <p>The EDQLS and comparator generic QoL scales were collected in person at baseline, and 3 and 6 months from 130 participants (mean age 25.6 years; range 14-60) in 12 treatment programs in four Canadian provinces. Total score differences across the time points and responsiveness were examined using both anchor- and distribution-based methods.</p> <p>Results</p> <p>98 (75%) and 85 (65%) responses were received at 3 and 6 months respectively. No statistically significant differences were found between the baseline sample and those lost to follow-up on any measured characteristic. Mean EDQLS total scores increased from 110 (SD = 24) to 124.5 (SD = 29) at 3 months and 129 (SD = 28) at 6 months, and the difference by time was tested using a general linear model (GLM) to account for repeated measurement (p < .001). Responsiveness was good overall (Cohen's d = .61 and .80), and confirmed using anchor methods across 5 levels of self-reported improvement in health status (p < .001). Effect sizes across time were moderate or large for for all age groups. Internal consistency (Chronbach's alpha=.96) held across measurement points and patterns of responsiveness held across subscales. EDQLS responsiveness exceeded that of the Quality of Life Inventory, the Short Form-12 (mental and physical subscales) and was similar to the 16-dimension quality of life scale.</p> <p>Conclusions</p> <p>The EDQLS is responsive to change in geographically diverse and clinically heterogeneous programs over a relatively short time period in adolescents and adults. It shows promise as an outcome measure for both research and clinical practice.</p

    Some proves about studies in relation to immuno genetic markers and selective process in creole cattle

    Se estudian los tipos sanguíneos eritrocitarios en Bovino Criollo Argentino, encontrándose posibles nuevos fenogrupos del Sistema B en esta raza. La metodología utilizada ha sido "Técnica hemolítica con fijación de complemento". En el análisis de las frecuencias génicas de los Sistemas Sanguíneos de un rodeo seleccionado comparativamente con otro rodeo introducido a la SEEA de Leales, INTA, Tucumán, se observaron diferencias significativas (P1 y Z2 ), no observándose diferencias significativas (hasta el presente) en los Sistemas C, F - V, 5, A, L, J, M y R' - 5' Destacamos la segregación de probables nuevos fenogrupos en el Sistema B tales como BGKO~O', py1 A', E3' G', B2C~ O', BGKC~ Y1E~ O', que significaría el agregado de aleles no detectados anteriormente, con el consiguiente incremento del destacado polimorfismo de Marcadores Genéticos característicos del Bovino Criollo Argentino.We studied Erythrocyte Blood Types in Argentine Creole Cattle, finding new phenogroups of B system. We have used Hemolytic Technics using Rabbit Complement Method-Genic frequency analysis of Blood Systems of a selected Herd comparatively with another Herd introduced to SEEA of Leales, INTA, Tucumán, we observed significant differences (P1 and Z2 alleles) we did not observe significant deferences (until now) in C, F V, 5, A, L, J, M and R' 5' Systems. We point out segregation in probable new phenogroups in B System, such as: BGKC~ O', PY1 A',E3'G', B2 OxO'~ BGKO~YiE3' O', which would mean adding alleles non detected before, increasing the great polimorphism of characteristic Genetic Markers of Argentine Creole Cattle.Facultad de Ciencias Veterinaria
