    Comparison of subdermal and perforator delay techniques on a rat flap model

    Background: In this study, we investigated the subdermal and perforator delay phenomena as a method to improve flap survival. Materials and Methods: In this experimental study, we used 24 rats in three groups. In the control group, the dorsal flaps were elevated and reinserted back to their place. In the experimental groups, we practiced the delay phenomena with two different techniques. In the first experimental group, cranial and lateral side incisions were performed; however, the flaps were not cut-off from the underlying fascia. In the second experimental group, we placed a silicon sheet under the planned flap to cut-off the circulation from the perforator vessels. Four weeks after the delay procedure, the flaps were raised completely and reinserted back to their place. Results: The average of necrotic area in the control group was 21.9% (±7.70). There was no necrosis in both experimental groups (P < 0.0001). Histological examination revealed that collagen density in both of the experimental groups was increased in comparison to the control group, it has only been found a significant first experimental group (P = 0.0315). We have not found any significant difference in lymphocyte density between the groups. Angiographic imaging has showed an increase in the vascular density in the flaps of the first experimental group. Conclusion: We believe that both of these delay techniques can be adapted to clinical applications and used safely to increase flap survival

    Different doses of radiation on agar colony forming development in C6 glioma cells: Assessment by thymidine labeling index, and bromodeoxyuridine labeling index

    WOS: 000254582600002Objective: Gliomas are relatively frequent in adults, and are among the most malignant primary brain tumors. Glioblastoma. multiforme, like many other tumors that exhibit radiation sensitivity in vitro, seems to be very resistant to radiation in vivo, thus suggesting that irradiation may not be a rate-limiting factor for malignant glioma tumor growth. In this study, we aimed to determine the optimal dose of radiation in C6 glioma colony forming assay, which is a valuable tool for antitumor treatment screening. Material and Methods: 10(5) cell/lamella colony forming cells were radiated with 200 cGy, 400 cGy, 800 cGy and 1600 cGy for 10 minutes. Radiosensitivity was measured systematically 24, 48, 72 and 96 hours after the radiation by three methods: soft-agar bilayer assay, thymidinE incorporation, and bromodeoxyuridine (BrdU) incorporation. Results: The soft-agar bilayer assay, which assessed the colony-forming units, showed that the number of colonies in the control group (609, 3 +/- 86.8) were decreased after 200 cGy (8.3 +/- 3.6) and 400 cGy (7.2 +/- 4.3). No colony was detected in 800 cGy and 1600 cGy irradiated cells [3H] Thymidine incorporation was more prominent with higher doses of radiation. BrdU incorporation revealed that even at low doses (200 cGy) of radiation there was a significant decrease of cell proliferation. On higher doses like 1600 cGy it was more prominent. Conclusion: Cell survival, doubling time, and cell phases are parameters of growth kinetics, and the results suggest that C6 glioma cells are radiosensitive and are virtually affected by all radiation doses in our experiment even 200 cGy at 24 hours. Besides, colony forming assay with thymidine labeling index, and BrdU labeling index may be used as new methods for determining radiotherapy doses in clinical applications

    INTRODUCTION: Obliteration of processus vaginalis (PV) has been proposed to result from persistence of smooth muscle which is presented transiently to propel the testis. Sacs associated with inguinal hernia were cultivated to define the cells that are going to proliferate for evaluating the association of inhibition of obliteration of PV and the presence of smooth muscle (SM). The present study does not only provide additional information about the presence of SM in sacs from boys with inguinal hernia, but also provides a new tool for researches directed to define the non- operative treatment of inguinal hernia

    GİRİŞ ve AMAÇ: Processus vaginalisin (PV) kapanmasının, testisin inişini sağlamak için geçici olarak bulunan düz kas hücrelerinin, devamlılığına bağlı olduğu ileri sürülmüştür. Bu amaçla, inguinal herni keseleri, PV kapanması ve düz kas hücre varlığı arasındaki ilişkinin değerlendirilmesi için kültüre edildi. Bu çalışma, sadece inguinal herni keselerinde düz kas hücresi varlığını gösteren ek bilgiler vermekle kalmayıp, aynı zamanda inguinal herninin cerrahi dışı tedavisi için yapılacak çalışmalar için yeni fikirler sunmaktadır. YÖNTEM ve GEREÇLER: Herni keseleri, yaşları 2 ay ile 5 yaş arasında değişen 11 çocuktan inguinal herni ameliyatı sırasında elde edildi. Örnekler uygun şekilde hazırlandı ve kültüre edildi. Hücrelerin morfolojik özellikleri ışık mikroskobu ile değerlendirildi. Hücrelerin hayatiyetleri, tripan blue exclusion metodu ile değerlendirildi. Gelişen hücreler, imminohistokimyasal olarak aktin ve miyozin ile boyandı. BULGULAR: Işık mikroskobu incelemesi ile bu hücrelerin iğ şekilli olduğu ve santral yerleşimli yuvarlak çekirdeklerinin bulunduğu görülmüştür. Tüm flask üreyen hücrelerle dolduğunda, kontakt inhibisyon olmadığından, üst üste çoğalan hücreler tipik tepe-vadi görünümü oluşturmuştur. Hücrelerin canlılığı %95’ in üzerinde bulunmuştur. Gelişen hücrelerin % 80 inin düz kas aktin ve düz kas myosin antikorları ile boyandığı saptanmıştır. TARTIŞMA ve SONUÇ: Herni kesesi dokulardan gelişen hücrelerin büyük çoğunluğunu, düz kas hücreleri oluşturmuştur. Bu bulgu, PV’in inhibisyonu ile düz kas hücresi varlığı arasındaki ilişkiyi desteklemektedir. Bu bilgi inguinal herninin cerrahi dışı tedavisi üzerinde yapılacak çalışmalar için kullanılabilecektir.INTRODUCTION: Obliteration of processus vaginalis (PV) has been proposed to result from persistence of smooth muscle which is presented transiently to propel the testis. METHODS: Hernia sacs were obtained from eleven boys with the ages ranging from two months to five years during operations for inguinal hernia. Samples were prepared and cultivated. Morphologic characteristics of cell populations were examined by light microscopy. Viability was estimated by trypan blue exclusion method. Growing cells were identified via immunohistochemical staining for smooth muscle actin and myosin. RESULTS: Light microscopic images of growing cells displayed characteristic spindle shaped morphology with centrally located round nucleus. When the flasks reached confluence, a hill-valley appearance was observed because of absence of contact inhibition. Cell viability was found more than 95%. Approximately, 80% of growing cell populations was stained positive with actins and myosin antibodies. DISCUSSION and CONCLUSION: In tissue explants of hernia sac, most commonly proliferating cell type is smooth muscle cells. This evidence supports the association of inhibition of PV and the presence of SM. The SM obtained from sacs associated with inguinal hernia may be used for researches directed to establish the non-operative treatment of inguinal hernia
