Capillary Electrophoresis for Sampling Single Nerve Cells
Two micro-capillary electrophoresis techniques are underdevelopment to acquire and determine catecholamines in single nerve cells. The first method involves the analysis of neurotransmitters in a portion of cytoplasm withdrawn from a single cell, and the second method involves the determination
of neurotransmitters in a whole nerve cell. Amperometric detection can be used to determine the easliy oxidized species in both of these methods. A large dopamine-containing cell in the brain of the pond snail, Planorbis corneus, has been used as a model system to evaluate both of these
microsampling techniques. Results for both the acquisition and separation of neurotransmitters in whole nerve cells as well as the analysis of small portions of single cell cytoplasm are presented here
Youth Definitions of Success, Obstacles to Success, and How Significant Others Can Help: Providing Youth a Voice in Their Own Development
Positive Youth Development (PYD) programmes are often conceptualized and led by adults with little or no youth input. The purpose of this study was to better understand youth definitions of success, obstacles they face in achieving success, and how they perceive others can help. Interviews were conducted with 24 youth from three different contexts (swim club, church group, and reform home) in Trinidad and Tobago. The results indicated that youth have a varied understanding of success. Further, many youths’ definitions mirror those prescribed in PYD literature. Youth identified a variety of obstacles in their quest for success, and though similarities existed, several obstacles were context dependent. Finally, youth identified support and assistance as the best mechanisms to propel them to success. Support, both informal and formal, points to the significant role non-parent adults serve in the lives of youth. Implications for youth programmes and future research are discussed
Reduced dimensionality spin-orbit dynamics of CH3 + HCl reversible arrow CH4 Cl on ab initio surfaces
A reduced dimensionality quantum scattering method is extended to the study of spin-orbit nonadiabatic transitions in the CH3 + HCl reversible arrow CH4 + Cl(P-2(J)) reaction. Three two-dimensional potential energy surfaces are developed by fitting a 29 parameter double-Morse function to CCSD(T)/IB//MP2/cc-pV(T+d)Z-dk ab initio data; interaction between surfaces is described by geometry-dependent spin-orbit coupling functions fit to MCSCF/cc-pV(T+d)Z-dk ab initio data. Spectator modes are treated adiabatically via inclusion of curvilinear projected frequencies. The total scattering wave function is expanded in a vibronic basis set and close-coupled equations are solved via R-matrix propagation. Ground state thermal rate constants for forward and reverse reactions agree well with experiment. Multi-surface reaction probabilities, integral cross sections, and initial-state selected branching ratios all highlight the importance of vibrational energy in mediating nonadiabatic transition. Electronically excited state dynamics are seen to play a small but significant role as consistent with experimental conclusions. (C) 2011 American Institute of Physics. [doi:10.1063/1.3592732
The real catecholamine content of secretory vesicles in the CNS revealed by electrochemical cytometry
Resolution of synaptic vesicle neurotransmitter content has mostly been limited to the study of stimulated release in cultured cell systems, and it has been controversial as to whether synaptic vesicle transmitter levels are saturated in vivo. We use electrochemical cytometry to count dopamine molecules in individual synaptic vesicles in populations directly sampled from brain tissue. Vesicles from the striatum yield an average of 33,000 dopamine molecules per vesicle, an amount considerably greater than typically measured during quantal release at cultured neurons. Vesicular content was markedly increased by L-DOPA or decreased by reserpine in a time-dependent manner in response to in vivo administration of drugs known to alter dopamine release. We investigated the effects of the psychostimulant amphetamine on vesicle content, finding that vesicular transmitter is rapidly depleted by 50% following in vivo administration, supporting the "weak base hypothesis'' that amphetamine reduces synaptic vesicle transmitter and quantal size
