    LAPORAN KASUS LEUCOCYTOZOONOSIS PADA AYAM BROILER DI LABORATORIUM PATOLOGI FAKULTAS KEDOKTERAN HEWAN UNIVERSITAS SYIAH KUALA

    ABSTRAK Pengamatan ini bertujuan mengidentifikasi penyakit leucocytozoonosis pada ayam broiler di Laboratorium Patologi  Fakultas  Kedokteran  Hewan  Universitas  Syiah  Kuala.  Sampel  dinekropsi kemudian  diperiksa  secara patologi anatomi. Organ yang mengalami perubahan patologi diambil dan difiksasi dalam neutral buffered formalin (NBF) 10% dilanjutkan dengan pembuatan sediaan histopatologi menggunakan pewarnaan haematoksilin dan eosin (HE). Pengamatan histopatologi dilakukan dengan mikroskop cahaya biokuler, kemudian dilakukan pemotretan dengan fotomikrograf. Data hasil pengamatan dianalisis secara deskriptif. Perubahan makroskopis yang ditemukan adalah bintik perdarahan dengan ukuran yang sangat bervariasi pada kulit, jaringan subkutan, otot, ginjal, hati, paru- paru, dan usus. Hati dan ginjal membengkak dan berwarna merah hitam. Pemeriksaan mikroskopik ditemukan adanya  perdarahan,  skizon  dan  megaloskizon    pada  otot  dada,  ginjal,  paru-paru,  dan  usus.  Dari  perubahan- perubahan tersebut dapat disimpulkan bahwa     ayam positif terserang leucocytozoonosis. Kata kunci : leucocytozoonosis, identifikasi, broiler, histopatolog

    IDENTIFICATION OF COPPER-INDUCIBLE GENES IN SWORDTAIL FISH (Xiphophorus spp.) USING DIFFERENTIAL DISPLAY

    Differential display technique was used to identify differentially expressed gene ofswordtail fish (Xiphophorus spp.) that induced by 1 µg/ml copper for 24 hours. From eightprimers that have been used for differential display, seven of them showed the differentialgenes which were not present in the control. The reamplification of these fragments rangingfrom below 100 to 200 bp. But only two fragments were successfully cloned and sequenced,those are H-AP10 and H-AP11. Sequence analysis showed that both of the sequences revealedhigh similarity with Homo sapiens stress-associated endoplasmic reticulum protein, a signalgenerated that induces apoptosis.Keywords: differential display, swordtail fish, copper pollution, mRNA expression, biomarker

    IDENTIFIKASI PROTEIN IMUNOGENIK TAKIZOIT DAN BRADIZOIT TOXOPLASMA GONDII STRAIN LOKAL UNTUK PENGEMBANGAN KIT DIAGNOSTIK

    Telah dilakukan penelitian untuk mengisolasi dan mengidentifikasi protein 28 kDa (GRA2) dari stadium takizoit dan bradizoit Toxoplasma gondii strain lokal. Cairan asites mencit galur Balb/c digunakan sebagai protein sampel melalui elektroforesis (SDS-PAGE), serum kambing untuk uji antigenitas protein 28 kDa (GRA2) dan serum darah kucing untuk melihat tingkat sensitivitas dan spesifisitas protein 28 kDa (GRA2). Data yang diperoleh dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa protein 28 KDa (GRA2) telah dapat diisolasi dari stadium takizoit dan bradizoit Toxoplasma sp. Nilai OD405 antigenesitas ES adalah sebesar 0,578 sedangkan pada antigen GRA2 (28 kDa) diperoleh nilai OD405 adalah sebesar 0,178. Tingkat sensitivitas protein 28 kDa (GRA2) terhadap antibodi dalam serum darah kucing terinfeksi adalah sebesar 61,53% dan nilai spesifisitasnya adalah 33,33%. Tingkat sensitivitas protein ES terhadap antibodi dalam serum darah kucing terinfeksi adalah 76,92% dan tingkat spesifisitasnya adalah 33,33%

    Histopathological Changes of Erythrocyte and Quantity of immature Erythrocytes in Tegal Duckling (Anas Javanica) Induced by Lead (Pb)

    The aim of this research is to find out the histopathological changes of erythrocyte and the quantity of immature erythrocyte of tegal duckling treated with lead (Pb). Twenty five young female ducks with the age of 7 days and 60.55.07 g body weight alloted into 4 treatment groups and 1 control group (Ko) with 5 replication each. The treatment group fed with K1 (5.7), K2 (11.4), K3 (17.1), and K4 (22.8) mg Pb/kg body weight for 10 days, respectively. Young ducks showed clinical symptoms such as the color of feces were brown to black, green color of diarrhea, anemic and decreasing body weight. Histopathologically, erythrocyte showed that immature and abnormal in shape (like teardrops, dumb bell, and bottles). The result of variant analysis followed by linier regression shows that the higher the dosage of Pb in diet the higher the quantity of immature erythrocyte andabnormal erythrocyte shape found (P0.01)

    The objective of this study was to find out the optimum salinity of media to control Dactylogyrus sp. in catfish. This research was conducted at Marine Biology Laboratory, Marine and Fisheries Faculty on January 2016. This research used non factorial completely randomized design (CRD) with 5 treatments and 4 replications I.e (A) 0 ppt, (B) ppt, (C) 8 ppt, (D) 9 ppt and (E) 10 ppt. The observed parameters were the prevalence and intensity of Dactylogyrus sp. the survival rate of catfish and the water quality of media. The result of this study indicated that the salinity significantly affects the prevalence and intesity of Dactylogyrus, but did not affect the survival rate of catfish. The lower prevalence and intesity of Dactylogyrus sp. was obtained at the treatment of 10 ppt with value 37,50% and 1,00 ind/fish.Penelitian ini bertujuan untuk mengetahui salinitas optimal yang dapat mengendali Dactylogyrus sp. pada ikan patin. Penelitian ini dilaksanakan di Laboratorium Biologi Laut, Fakultas Kelautan dan Perikanan, Universitas Syiah Kuala pada Bulan Januari 2016. Penelitian ini menggunakan rancangan acak lengkap (RAL) non faktorial yang terdiri 5 perlakuan salinitas dan 4 ulangan yaitu (A) 0 ppt, (B) 7 ppt, (C) 8 ppt, (D) 9 ppt, dan (E) 10 ppt. Parameter yang diukur yaitu prevalensi, intensitas Dactylogyrus sp. tingkat kelangsungan hidup ikan patin, serta parameter fisika kimia - kualitas air. Hasil dari penelitian ini menunjukkan bahwa salinitas berpengaruh nyata terhadap prevalensi dan intensitas Dactylogyrus sp., namun tidak berpengaruh nyata terhadap kelangsungan hidup ikan patin. Tingkat intensitas dan prevalensi Dactylogyrus sp. terendah terdapat pada perlakuan 10 ppt dengan nilai 37,50% dan 1,00 ind/ekor

    MERCURY CHLORIDE (HgCl2) EXPOSURE CHANGES THE HISTOPATHOLOGICAL FIGURE OF EYE AND BRAIN OF TILAPIA FISH (Oreochromis mossambicus)

     Mercury pollution brings harmful effects to aquatic animals, the environment and eventually to human health. Mercury accumulates in the liver, kidney, eye lens and brain of fish, resulting in organ damage. This study aimed to determine the effect of HgCl2 exposure on anatomical pathology and histopathology of tilapia fish eye and brain. A total of 36 male tilapia fish were allotted into 4 treatment groups with 3 replications. Fish were exposed to 0.00, 0.25, 0.50, and 0.75 ppm of HgCl2 for 10, 20, and 30 days. Subsequently, the anatomical pathology was observed followed by histopathological examination. Anatomical pathology examination of fish eye on day 30 showed white membrane on the eye lens surface, pupil diminution, and sunken eyes. The brain demonstrated hemorrhage, necrosis, discolorations, and granulated area. The retina showed necrosis, retina pigmentation flexiform layer widened, and cone cell atrophy. Brain depicted structural and cellular damage such as degeneration necrosis and vacuolation. HgCl2 exposure changes the anatomical pathology and histopathology of tilapia fish eye and brain

    Efektivitas Anti Parasit Ekstrak Metanol Daun Pandan Semak Berduri (Pandanus Odoratissimus) Terhadap Mortalitas Lintah Ikan (Piscicola Geometra) Pada Ikan Nila (Oreochromis Niloticus)

    The objective of this study was to determine the effectiveness of Pandanus odoratissimus leaf on Piscicola geometra that infected tilapia fish Oreochromis niloticus. The research was conducted at Laboratory of Marine and Fisheries Faculty, and Laboratory of Chemistry, Mathematics and Natural Sciences Faculty, Syiah Kuala University on June, 2015. This research was designed using non factorial completely randomized design, with seven treatments of concentration (0; 50; 60; 70; 80; 90 dan 100 ppm) and three repetitions. The highest mortality of P. Geometra found at concentration of was 100 ppm. The result of (ANOVA) showed P. Odoratissimus leaf gane the significant effect on the mortality of P. Geometra the optimum concentration of P. O. doratissimus leaf in this study was at 100 ppm and O. Niloticus (P<0.005).     The purpose of this study was to identify and evaluate the prevalence of Aeromonas hydrophila bacteria that attack African catfish to in Aceh Besar district with the microbiological test. This study was conducted on March 2016. Samples were collected from the District recorded have African catfish fish in Aceh Besar district, namely the Baitussalam District. Observations conducted in the laboratory of Bacteria Quarantine Fish Quality Control and Safety of Fishery Class 1 Aceh. This study used a descriptive analytic and the conventional method where the sampling was done by a stratified random sampling method. Further, bacteria was isolated, the morphological of bacteria were observeted, pure cultures included Gram staining, catalase test, test oxidase, biochemical tests include tests Triple Sugar Iron Agar (TSIA), test Lysine Iron Agar (LIA), test citrate, test urea, test Oxidation / fermentative (O/F), Methyl Red test Vogue Proskuer (MRVP), carbohydrate fermentation test, test Motility Indol Ornithine (MIO)and the reading of the identification results were conducted.The result showed that three samples of African catfishfrom concrete pond, were positive infected with 100% prevalance attacked byAeromonas hydrophila. Penelitian ini bertujuan untuk mengidentifikasi serta menghitung nilai prevalensi bakteri Aeromonas hydrophila yang menyerang ikan lele dumbo yang dibudidayakan di Kabupaten Aceh Besar dengan uji mikrobiologi. Penelitian ini dilakukan pada Bulan Maret 2016. Pengambilan sampel dilakukan pada Kecamatan Baitussalam Kabupaten Aceh Besar. Pengamatan bakteri dilakukan di Laboratorium Bakteri Karantina Ikan Pengendalian Mutu dan Keamanan Hasil Perikanan Kelas 1 Aceh. Penelitian ini menggunakan deskriptif analitik dan metode konvensional dimana pengambilan sampel dilakukan dengan metode pengambilan acak terstratifikasi. Selanjutnya dilakukan isolasi bakteri, pengamatan morfologi, kultur murni meliputi uji Gram, uji katalase, uji oksidase, uji biokimia meliputi uji Triple Sugar Iron Agar (TSIA), uji Lysine Iron Agar (LIA), uji sitrat, uji urea, uji Oksidasi/Fermentatif (O/F), uji Methyl Red Vogue Proskuer (MRVP), uji fermentasi karbohidrat, uji Motility Indol Ornithin (MIO), dan pembacaan hasil identifikasi. Hasil penelitian menunjukkan bahwa 3 sampel ikan lele dumbo dari kolam beton positif terinfeksi bakteri Aeromonas hydrophila dengan prevalensi yang menyerang ikan lele dumbo kolam beton 100%

    The Purpose of this study were to identify the type of endoparasite and to find out the prevalance of endoparasite infected digestive tract of Colossoma macropomum. The study was conducted at parasitology Laboratory of Veterinary Medicine Faculty, Syiah Kuala University in May - June 2015. The sample used were collected randomly. As many as 10% of total fish in a population. The exa mination of endoparasite were carried out using smear method. Parameter measured were prevalence and physic – chemical of water. The results showed that type of endoparasite found in digestive tract of Colossoma macropomum fish was Capillaria sp. with the prevalence of 15%. Tujuan penelitian ini untuk mengidentifikasi jenis endoparasit yang menginfeksi dan mengetahui prevalensi endoparasit yang menginfeksi saluran pencernaan ikan bawal air tawar. Penelitian dilakukan di Laboratorium Parasitologi Fakultas Kedokteran Hewan, Universitas Syiah Kuala pada Bulan Mei dan Juni 2015. Penelitian menggunakan metode smear method dan penyamplingan sampel dilakukan secara acak. Sampel yang digunakan dalam penelitian sebanyak 10% dari total ikan dalam satu populasi. Parameter yang diukur yaitu prevalensi dan parameter fisika – kimia air. Hasil dari penelitian menunjukkan bahwa jenis endoparasit yang teridentifikasi pada ikan bawal air tawar adalah Capillaria sp.
