Due to its rapid development of insecticide resistance, conventional insecticides are not effective to thrips control, thus requiring an alternative control method. RNA interference (RNAi)-based control strategy has been developed to control various phytophagous chewing pests. However, no successful case of RNAi-based control was reported for sucking pests including thrips. In this study, the ingested amount of nuclei and plastids following time course in the two thrips (F. occidentalis and F. intonsa) along with two reference sucking pests (Tetranychus urticae, a cell feeder, and Nilaparvara, a cap feeder) were quantified by quantitative PCR (qPCR). The ingested amount of plastids was significantly greater than those of nuclei in the thrips and mites species. However, substantially lower and greater ratio between ingestion amount of nuclei and plastids than those in intact kidney bean leaves was identified in the two thrips and mite species, respectively, suggesting that thrips and mite has different preference for nuclei and plastids. In contrast, no plant subcellular fractions were detected in brown planthopper. These findings provide the first proof-of-concept that hairpin RNA expressed in the nucleus can be delivered to mesophyll sucking feeders such as thrips and mites. With this in mind, a total of 57 candidate genes were selected from the transcriptome data of F. occidentalis, and the double-stranded RNAs (dsRNA), targeting each candidate gene, were delivered to a susceptible strain of F. occidentalis via the leaf disc-feeding method. The dsRNA of toll-like receptor 6 (TLR6), apolipophorin (apoLp), coatomer protein subunit epsilon (COPE), and sorting and assembly machinery component 50 (SAM50) resulted in the highest mortality in both insecticide-susceptible strain and -resistant strain. The dsRNA-fed thrips showed 46%, 47%, 37%, and 22% reduced transcription levels of TLR6, apoLp, COPE and SAM50 in insecticide-susceptible strain and 71%, 36%, 27%, and 50% in -resistant strain, respectively. This result demonstrates that the observed mortality following dsRNA ingestion was due to RNAi. The efficacy of ingestion RNAi of the lethal genes was also confirmed against an insecticide-resistant strain of F. occidentalis, indicating that RNAi-based control would be effective against both susceptible and resistant populations of F. occidentalis. Development of transgenic tomato plants expressing dsRNA targeting these lethal genes is currently in progress as a practical tool to control thrips in the field. ๊ทธ ๊ฒฐ๊ณผ, Toll-like receptor 6 (TLR6), apolipophorin (apoLp), COPE (coatomer protein subunit epsilon) ๋ฐ SAM50 (sorting and assembly machinery component 50)์˜ dsRNA๋Š” ์‚ด์ถฉ์ œ ๊ฐ์ˆ˜์„ฑ ๋ฐ ์‚ด์ถฉ์ œ ์ €ํ•ญ์„ฑ ๊ณ„ํ†ต ๋ชจ๋‘์—์„œ ๋†’์€ ์น˜์‚ฌ์œจ์„ ๋ณด์—ฌ์ฃผ์—ˆ๋‹ค. dsRNA๋ฅผ ์„ญ์‹ํ•œ ๊ฝƒ๋… CONTENTS ABSTRACT i LIST OF TABLES vi LIST OF FIGURES vii CHAPTER 1. Thyroid Isthmus Length and Iodine Turnover as Predictors of Successful Radioactive Iodine Therapy in Patients with Graves' Disease

Radioactive iodine (RAI) therapy is an effective treatment option for Graves' disease. However, predicting treatment failures after RAI therapy remains controversial. The objective of this study was to investigate the factors associated with the success rate of RAI therapy for treatment of Graves' hyperthyroidism. Thyroid functional outcome, pre-RAI ultrasonographic features, and clinical parameters were evaluated retrospectively in 98 patients followed up for at least 12 months after RAI (mean RAI dose was 11.7โ€‰ยฑโ€‰1.8โ€‰mCi). Hypothyroidism was achieved in 59 patients (60.2%), and euthyroidism in 16 patients (16.3%), while 23 patients (23.5%) remained hyperthyroid. Age, sex, body mass index, pre-RAI thyroid function, or thyroid-stimulating immunoglobulin levels were not associated with treatment outcome. Length of thyroid isthmus (p = 0.028) and 2- to 24-hour iodine uptake ratios (p = 0.002) were significantly associated with treatment failure, which was defined as a persistent hyperthyroid status after RAI therapy. Patients with a longer isthmus had a higher risk of remaining hyperthyroid, with a threshold for isthmus length of 5.2โ€‰mm, with a sensitivity of 69.6% and specificity of 70.3% for treatment success. Measuring the length of the thyroid isthmus can be a simple and useful way to predict RAI treatment outcome.ope

Localization of oncogenic osteomalacia by systemic venous sampling of fibroblast growth factor 23

PURPOSE: Tumor-induced osteomalacia (TIO) is characterized by hypophosphatemia caused by a phosphaturic mesenchymal tumor. While surgical resection of the tumor leads to a cure, identification of the responsible tumor is challenging. Recently, several studies showed that systemic sampling of fibroblast growth factor 23 (FGF23) is helpful for localization of tumors. The present study aimed to evaluate the clinical utility of this method in Korean patients. MATERIALS AND METHODS: Six patients compatible with TIO who were admitted to our hospital between 2006 and 2015 were analyzed. Systemic venous sampling of FGF23 was performed to detect blind lesions or to confirm a suspicious lesion identified in previous imaging studies. RESULTS: Venous sampling helped confirming the tumor in five of the six patients. Three patients underwent surgery after sampling, and in two patients, the lesions were detected after 3 years by means of โถโธGa-DOTATOC positron emission tomography with computed tomography. In one patient, there was a local elevation of serum FGF23 without any related lesion on additional imaging. CONCLUSION: Our data strengthened the value of venous sampling of FGF23 in predicting the location of tumors and suggested that it can be more effective in the presence of the relevant lesion in subsequent imaging analyses.ope

Untreated Congenital Adrenal Hyperplasia with 17-ฮฑ Hydroxylase/17, 20-lyase Deficiency Presenting as Massive Adrenocortical Tumor

Congenital adrenal hyperplasia (CAH) with 17ฮฑ-hydroxylase/17,20-lyase deficiency is usually characterized by hypertension and primary amenorrhea, sexual infantilism in women, and pseudohermaphroditism in men. hypertension, and sexual infantilism in women and pseudohermaphroditism in men. In rare cases, a huge adrenal gland tumor can present as a clinical manifestation in untreated CAH. Adrenal cortical adenoma is an even more rare phenotype in CAH with 17ฮฑ-hydroxylase/17,20-lyase deficiency. A 36-year-old female presented with hypertension and abdominal pain caused by a huge adrenal mass. Due to mass size and symptoms, left adrenalectomy was performed. After adrenalectomy, blood pressure remained high. Based on hormonal and genetic evaluation, the patient was diagnosed as CAH with 17ฮฑ-hydroxylase/17,20-lyase deficiency. The possibility of a tumorous change in the adrenal gland due to untreated CAH should be considered. It is important that untreated CAH not be misdiagnosed as primary adrenal tumor as these conditions require different treatments. Adequate suppression of adrenocorticotropic hormone (ACTH) in CAH is also important to treat and to prevent the tumorous changes in the adrenal gland. Herein, we report a case of untreated CAH with 17ฮฑ-hydroxylase/17,20-lyase deficiency presenting with large adrenal cortical adenoma and discuss the progression of adrenal gland hyperplasia due to inappropriate suppression of ACTH secretion.ope

Hepatic Sarcoidosis in a Patient with Chronic Hepatitis B Virus Infection

    Get PDF
Sarcoidosis is a systemic inflammatory granulomatous disease affecting multiple organs, including liver, spleen, heart, eyes, and skin. Liver involvement is reported in 11.5% of cases and many studies have reported on the association between hepatitis C virus infection and sarcoidosis. However, the role of hepatitis B virus (HBV) infection as a trigger for sarcoidosis has never been reported. We describe a case of hepatic sarcoidosis in a patient with chronic hepatitis B infection, with a possible link between the two. It is the first case report of a patient with interferon-ฮฑ-naรฏve chronic HBV infection presenting with hepatic sarcoidosis accompanied by portal hypertension and liver cirrhosis
