    Expression of the Soluble Human Fas Ligand in Dictyostelium discoideum

    [中文文摘]用PCR扩增从激活的人中性粒细胞中得到的编码可溶性Fas配体胞外区中第14 1个到第2 81个氨基酸的cDNA ,将其与hCG_β信号肽片段融合到质粒MB12neo中,随后导入到盘基网柄菌AX3细胞中,得到分泌性表达hFasL的重组菌AX3_H3。为提高shFasL的表达量,对质粒pMB12neo作了改造,得到衍生质粒pMB74。利用质粒pMB74克隆表达shFasL ,得到高通量表达shFasL的重组菌AX3_pLu8。在复杂培养基HL_5C中,重组菌的细胞密度可达(1 5~2 )×10 7 mL ,AX3_H3及AX3_pLu8分泌的shFasL浓度分别为2 3 5 μg L及2 0 6 μg L。利用合成培养基SIH培养重组菌AX3_H3及AX3_pLu8,细胞密度均达到(4~5 )×10 7 mL ,shFasL浓度则分别达到111μg L和4 2 0 μg L。[英文文摘]An expression system is described for high-yield production of recombinant soluble human FasL (shFasL) in Dictyostelium discoideum cells. DNA encoding amino acids 141~281 of hFasL was PCR amplified from cDNA derived from activated human neutrophils. The resulting product was fused with a DNA fragment encoding hCG-β signal peptide and cloned in the expression vector pMB12neo. Dictyostelium strain AX3 was transfected with this plasmid, yielding a recombinant strain called AX3-pCESFL95-H3. In order to mprove the shFasL expression level , pMB12neo was optimized by replacing its transcriptional terminatorPpolyadenylation segment of the 2H3 gene with an actin8 terminatorPpolyadenylation segment , yielding derived expression vector pMB741 The recombinant Dictyostelium strain called AX3-pLu8 was generated with this new plasmid. When the recombinant cells were cultivated in a complex HL-5C medium, a cell density of (115~2) ×107PmL was reached , and the shFasL level expressed by strains AX3-pCESFL95-H3 and AX3-pLu8 was 2315 μgPL and 206μgPL , respectively. By using a newly developed synthetic medium called SIH as culture medium, higher cell density of (4~5) ×107PmL was achieved. Correspondently, 111 μgPL and 420 μgPL shFasL were secreted by recombinant strains AX3-pCESFL95-H3 and AX3-pLu8 ,respectively.国家自然科学基金资助项目(No.20306025,30370039

    Traumatic superficial temporal artery aneurysm:analysis of 7 cases

    目的探讨外伤性颞浅动脉瘤的诊断和治疗方法。方法回顾性分析7例外伤性颞浅动脉的临床表现、手术方法和治疗结果。所有病例均因外伤后额部或颞部进行性增大肿物就诊,无颞浅动脉瘤破裂出血病例,全部病例在局部麻醉下切除动脉瘤囊肿,其中1例为分叉部梭形动脉瘤。结果病理报告均为假性动脉瘤,动脉外膜存在。无术后头皮缺血、坏死发生。结论根据外伤史、颞浅动脉走行上搏动性肿物的特点,颞浅动脉瘤即可明确诊断。动脉壁中膜弹力层断裂,局部因动脉血流搏动冲击膨出形成可能是颞浅动脉瘤形成的另一个机制。动脉瘤切除术是治疗颞浅动脉瘤的有效方法。Objective To explore the diagnosis and treatment for traumatic superficial temporal artery pseudoaneurysm.Methods Clinical manifestations, operation and prognosis of 7 patients with traumatic superficial temporal artery pseudoaneurysm were analyzed retrospectively.There was the same complaint of progressive swelling mass on the frontal or temporal area post brunt injury, but no complaint of rupture of the aneurysm in all the patients.All the patients underwent resections of the aneurysm capsule under local anesthesia, including one fusiform aneurysm at the bifurcation of the superficial temporal artery in 1 case.Results All the cases were pseudoaneurysm with intact outlayer of the artery.There was no complication of scalp ischemia or necrosis.Conclusion Traumatic superficial temporal artery pseudoaneurysm can be diagnosed according to the history of head injury and progressive pulsatile swelling or mass on the line of the superficial temporal artery.It may be another mechanism of forming pseudoaneurysm that breakdown of the elastic layer in the tunica media of the arterial wall and beating of arterial blood flow arterial lead to lumen enlargement on the local point.Resection of the aneurysm is an effective way of the treatment

    本文首次对厦门西海域表层沉积物中碱性磷酸酶活力的分布、动态进行了检测和分析,并在同步检测该沉积物各种形态磷含量的基础上对该酶与各形态磷的关系进行了探讨.结果表明,厦门西海域表层沉积物中碱性磷酸酶的活力分布与陆源排污对水体的污染有明显的相关性;酶活力与总磷、总无机磷含量相关性较大,与弱吸附可交换磷相关性不大,与总有机磷含量则呈显著不相关性.并认为,就厦门西海域而言,碱性磷酸酶活力的水平可作为小区域水体富营养化程度的重要参考指标之一.国家自然科学基金;福建省自然科学基

The structure of C50H10 is identified through NMR, mass spectrometry, and IR and Raman spectroscopy as a D5h symmetric closed-cage molecule with five pairs of fused pentagons stabilized by ten hydrogen atoms. UV/Vis and fluorescence spectrometric analyses disclose its optical properties as comparable with those of its chloride cousin (C50Cl10).     在现有自适应粒子群优化算法的研究基础上本文引入1种反弹机制(rEbOund MECHAnISM),提出了1种改进的粒子群算法——反弹自适应粒子群优化算法。rAPSO能在搜索过程中充分利用粒子的飞行速度和方向等信息(下文称为动量信息),维持粒子的多样性以提升算法的搜索性能。通过比较,本文提出的rAPSO在一定程度上改进了现有的自适应粒子群算法的优化性能。运用rAPSO对催化裂化装置进行优化试验,其结果表明无论在单变量优化还是在多变量优化中,该装置的转化率都得到了一定程度的提高。This paper presents a new method named Rebound Mechanism for improving traditional Particle Swarm Optimization(PSO)algorithm.Based on this method and a kind of Adaptive PSO algorithm this paper proposed a modified PSO algorithm named Rebound Adaptive Particle Swarm Optimization(RAPSO)algorithm.The proposed approach can maintain the population diversity and improve the PSO algorithm by taking full advantage of the momentum information,such as flight speed and orientation of particles.According to comparison,the RAPSO improved existing PSO algorithm in some degree.The results of a Fluid Catalytic Cracking(FCC)unit optimization experiment applied RAPSO indicated that the conversion rate increased a lot regardless of applying single-objective optimization or multiple-objective optimization.中央高校基本科研业务费专项资金(2010121047

    催化裂化装置(fluId CATAlyTIC CrACkIng unIT,fCCu)对炼油厂的经济效益至关重要,本文主要探讨了人工神经网络在催化裂化装置建模中的应用。利用实际的工业数据分别采用lMbP,rbf_PlS神经网络对某工厂的催化裂化装置进行了建模试验。将它们的拟合与泛化结果、学习速度以及参数调整进行了比较,其结果显示rbf_PlS神经网络在收敛速度以及预测性能等方面均优于lMbP神经网络。此外,本文在神经网络模型的基础上对其进行了最小二乘校正,得到了比较满意的结果。This paper discussed the application problems of artificial neural network(ANN) modeling of industrial fluid catalytic cracking unit (FCCU).Two neural network,the Levenberg-Marquardt Back Propagation(LMBP) neural network and the Radial Basis Function(RBF) neural network using the partial least square(PLS) algorithm were adopted in this work.Use the data from an Industrial FCCU,the training rate and the generalization capability of these two neural network models were compared.Online least square correction(LSC) was lead in this study to test the online weight modification of the RBF neural network model.The results show that RBF_PLS neural network has a better performance in generalization capability and other aspects such as the training time and parameters determination.Besides,when LSC was used,the results became more satisfied.中央高校基本科研业务费专项资金(2010121047
