    Circular RNA CircHIPK3 Promotes NCI-H1299 and NCI-H2170 Cell Proliferation through miR-379 and its Target IGF1

    背景与目的已有的研究证明:环状RNA是一类在哺乳动物中普遍存在的具有稳定闭合环状结构的内源性RNA分子。环状RNA; circHIPK3(circular RNA HIPK3, circHIPK3)在肝细胞癌(hepatocellular carcinoma,; HCC)中表达水平较高,促进肝癌细胞生长。但是其在非小细胞肺癌(non-small cell lung cancer,; NSCLC)中的作用及其调控机制尚无文献报道。本研究拟探讨环状RNA; circHIPK3对NSCLC细胞系NCI-H1299和NCI-H2170细胞增殖的影响,并进一步研究其调控的分子机制。方法Real-time; PCR法检测circHIPK3在NSCLC各细胞系中的表达水平。CCK-8实验和克隆形成实验检测过量表达和干扰circHIPK3对细胞增殖的影响; 。双荧光素酶报告基因实验分别检验miR-379与circHIPK3及miR-379与IGF1 mRNA的结合情况。Western; blot和ELISA检测细胞内外的IGF1蛋白表达水平。结果环状RNA; circHIPK3在6株NSCLC细胞株中均有表达,其中H1299表达最低,H2170表达最高,通过转染过表达的circHIPK3可显著促进NC; I-H1299细胞增殖,干扰circHIPK3可显著抑制NCI-H2170细胞增殖。miR-379可与circHIPK3及IGF1; mRNA直接结合。过表达circHIPK3导致IGF1表达量上调,干扰circHIPK3能够下调IGF1表达水平,转入miR-379; mimics恢复了circHIPK3稳转细胞株IGF1表达水平的上调及细胞增殖表型。结论环状RNA; circHIPK3在NSCLC细胞系NCI-H1299及NCI-H2170中可通过miR-379调控IGF1表达促进细胞增殖,环状RNA; circHIPK3可能成为非小细胞肺癌治疗的新靶点。Background and objective It has been proven that the circular RNA,; possessing a stable covalently closed continuous loop, is a type of RNA; molecule which is expressed widespread in mammals. The circular RNA; circHIPK3 is abundantly expressed in hepatocellular carcinoma (HCC) and; promotes tumourgenesis. However, a role for circHIPK3 has not been; systematically examined in non-small cell lung cancer (NSCLC). In this; study, we investigated whether circHIPK3 has an effect on cell; proliferation in the NSCLC cell lines NCI-H1299 and NCI-H2170 and the; underlying molecular mechanisms. Methods The expression of circHIPK3 was; measured by real-time PCR in NSCLC cell lines. Nuclear mass separation; experiment was used to detect the location of circHIPK3 in cells. The; gain and loss function experiments were used to examine the; proliferation of NCI-H1299 and NCI-H2170 cells by Cell Counting Kit-8; (CCK-8) and the colony formation assays. Then, circHIPK3 was cloned into; the downstream of the luciferase reporter gene which activity was; detected to verify whether miR-379 could bind with circHIPK3 or IGF1; mRNA. The protein level of IGF1 was detected by Western blot and ELISA; in circHIPK3 overexpressed/knock-down NCI-H1299 and NCI-H2170 cells.; Results CircHIPK3 was generally expressed in six kinds of NSCLC cells; lines we detected, and the expression level was highest in H2170 and; lowest in H1299. Overexpression of circHIPK3 obviously promoted; NCI-H1299 cell proliferation and knock-down of circHIPK3 inhibited; NCI-H2170 cell proliferation. In the luciferase assay, miR-379 was; observed to sequester circHIPK3 and IGF1 mRNA with potential binding; sites. Furthermore, we found that the overexpression of circHIPK3 could; increase the expression levels of IGF1 and knockdown reduced it.; Moreover, up-regulation of miR-379 rescued the phenotype induced by; overexpression of circHIPK3. Conclusion CircHIPK3 could promote cell; proliferation by a circHIPK3/miR-379 pathway in NCI-H1299 and NCI-H2170; cells and might be a potential tumor biomarker for NSCLC.国家自然科学基金项

    New Distribution Records of One Genus and Six Plants from Coastal Zones,Fujian Province

    在福建海岸带与海岛生物多样性调查中,发现了1个福建省被子植物新记录属:山黧豆属(Lathyrus),6个新记录种(含变种):蓝花子(Raphanus sativus var.raphanistroides)、海滨山黧豆(Lathyrus japonicus)、大狼毒(Euphorbia jolkinii)、冬青卫矛(Euonymus japonicus)、大叶胡颓子(Elaeagnus macrophylla)、滨艾(Artemisia fukudo)。凭证标本均存放于厦门大学植物标本馆(Au)。One newly recorded genus,Lathyrus,and six species(varity),namely Raphanus sativus var. raphanistroides,Lathyrus japonicas,Euphorbia jolkinii,Euonymus japonicas,Elaeagnus macrophylla and Artemisia fukudo,were recorded in Fujian Province during plant resource survey on coasts and islands.     Iron and manganese oxides enhance electron output efficiency of Clostridium pasteurianum

    [Background] Fermentative dissimilatory metal-reducing bacteria, which are capable of reducing metallic oxides, get energy from fermentation. Little is known about how metallic oxides affect electron output efficiency of fermentative dissimilatory metal-reducing microorganisms. [Objective] This study was conducted to explore the influence of iron and manganese oxides (Fe_2O_3/MNO_2) on electron output efficiency. [Methods] Different concentrations of Fe_2O_3/MNO_2 were added into fermented system containing glucose and inoculated 5% C. pasteurianum. Electrochemical activity of C. pasteurianum was detected. The concentrations of Fe(Ⅱ) and Mn(Ⅱ) were measured by ferrozine spectrophotometry and formaldoxime method. Fermentation substrate and metabolites of C. pasteurianum were detected by gas chromatography and high performance liquid chromatography. Lastly, we calculated the electron output efficiency. [Results] The current density peaked with the value of about 0.93 mA/m~2. The concentrations of Fe(Ⅱ) and Mn(Ⅱ) gradually accumulated. The consumption of glucose was increased by 9.4%/7.7%, Meanwhile, acetate production was increased by 37.5%/25.0%, and butyrate production was increased by 22.7%/6.8%. Additionally, hydrogen production was increased by 21.6%/9.8%, and the total electron output efficiency was increased by 24.27%/10.82%, respectively. The pH values between experimental group and control are no significant difference. [Conclusion] This study shows that iron and manganese oxides can improve the electron output efficiency of C. pasteurianum by increasing glucose consumption and buffering pH value. The results provide evidence for revealing the effects of multivalent metal oxides on the electron output of fermentative dissimilatory metal-reducing bacteria, and further expand our understanding of the interaction mechanism between multivalent metal oxides and fermentative dissimilatory metal-reducing bacteria.</p

    “动物酵素营养液”对母猪便秘的影响

    本试验选择28日龄的仔猪,喂养动物酵素营养液至70日龄,结果表明:酵素组的仔猪中,康复率为75%,28.6%完全康复,57.1%转为轻微患病,14.3%处于严重状态,在对照组中只有40%完全康复,严重时有1头死亡,4头转为轻微患病,说明动物酵素营养液对仔猪呼吸道疾病有较好的促进康复的效果

    动物酵素营养液对繁殖母猪生产性能的研究

    为验证“动物酵素营养液“对母猪生产性能的影响,选120头母猪进行饲养试验。结果表明,“动物酵素营养液“能够提高母猪群产仔率、成活率,平均窝产仔最多12.5头,成活率最高可达98%~99%,仔猪出生重为1.60千克/头,断奶后仔猪体重为11.2千克/头。猪无腹泻现象,长势快,皮毛光滑,整齐度提高,母猪妊娠期正常,产程时间少,无三炎,母猪健康,可在产后3~4天开始发情,配种率与受胎率均为100%,每年可产2.4窝,每头母猪与对照组的经济差额为4 520元

    弹道靶上的跨音速实验

    本文介绍用弹道靶作跨音速试验的新方法,在二级轻气炮上采用了二个特殊的新技术:充气放炮能发射模型达到亚音速,低充填条件能发射模型达到跨、超音速。由于初始加速度较小,发射的模型外形完整,姿态稳定;加上没有支撑干扰和小洞壁效应的优点,使弹道靶设备在跨音速实验方面有特殊的优越性,用上述技术得到了圆球跨音速流场的清晰照片,其中击波脱体距离、分离点位置、颈部宽度、尾迹形状与超音速、高超音速流动比较有明显变化。文中M≈1.010和M≈0.99的二张全流场照片是十分有意义的。最后提出一个想法,用靶场加压、降温的方法可以提高雷诺数直到10~7以上
