    Genetic Diversity Analysis of Five Populations of Paphia undulata

    采用形态特征和微卫星分子标记对采自菲律宾(PH)、泰国(TH)、福建云霄(YX)、海南儋州(HN)和广西北海(BH)的5个地理群体的波纹巴非蛤(Paphia undulata)进行遗传多样性分析.形态差异分析结果显示,国外群体和国内群体形态差异明显:2个国外群体(PH和TH)形态接近,聚为一支;国内支系中,YX和HN群体的形态较近,先聚为一支,而BH群体的外部形态与前两者差异较大.Hardy-Weinberg平衡检验检测到所有群体在5个位点上均出现不同程度地偏离平衡.5个群体间遗传分化指数(FST,0.000 5~0.183 1)、基因流值(Nm,2.230 46~1 066.23)以及分子方差分析(AMOVA,群体间的变异贡献率为10.21%,p<0.000 1)结果表明,群体间存在中等程度的分化.基于遗传距离的非加权组平均法(UPGMA)聚类分析显示:2个国外群体和3个国内群体分别聚为一支,再聚为一个整体;国内支系中,YX和BH群体先聚为一体,再与HN群体聚类.研究结果揭示了波纹巴非蛤较高的遗传多样性水平和中等程度的遗传分化水平,同时也暗示其现有的遗传分化程度受到人类生产活动的干扰.Genetic diversity of five populations of Paphia undulata sampled from Philippines( PH),Thailand( TH),Fujian Yunxiao( YX),Hainan Danzhou( HN) and Guangxi Beihai( BH) was studied using the morphological variation analysis and 8 micro-satellite markers.The morphological variations showed significant differences between the Chinese group and the exotic group.Within the Chinese group,the samples from YX and HN populations were rather similar in morphology,whereas BH population differed greatly from the above two populations.Meanwhile,significant departures from Hardy-Weinberg equilibrium were observed in 5 of the 8 microsatellite loci in all the 5 populations.The lines of FST( 0. 000 5 to 0. 183 1),Nm( 2. 230 46 to 1 066. 23) and AMOVA analysis( 10. 21% of genetic variation was distributed within groups with the significant value,p<0. 000 1) across all populations indicated that there was medium level of divergence among the5 P. undulata populations.The UPGMA clustering tree based on genetic distance demonstrated that the populations of PH and TH clustered into one group,and the other three Chinese populations clustered into one other group.Within the Chinese group,the HN population clustered into one group,while the populations of YX and BH clustered together,which indicated that the YX population was more closely related to the BH population.In this study,high levels of genetic diversity within populations and moderate levels of genetic differentiation among populations were presented for P. undulata.Our data also revealed that the genetic pattern of P. undulata was likely to be disturbed by human-mediated passive dispersal via aquaculture activities.These results would provide scientific basis for the conservation and reasonable utilization of natural resources of P. undulata.海洋公益性行业科研专项(201205021-2);; 国家贝类产业技术体系项目(CARS-48

    三七总苷对二甲基亚硝胺致肝纤维化大鼠胶原代谢的影响

    目的:考察三七总苷对二甲基亚硝胺(DMN)致肝纤维化模型大鼠胶原代谢的影响。方法:用DMN复制中度肝纤维化模型,随机分为三七总苷组、干扰素组、三七总苷配合干扰素组、阴性对照组,分别以相应的药物连续治疗8周,检测各组大鼠血清肝功能、透明质酸及肝组织羟脯氨酸(Hyp)含量,结合常规病理HE、苦味酸-天狼星红染色,评价肝组织炎症及纤维化程度。结果:中度肝纤维化大鼠模型经过连续8周的三七总苷治疗,Hyp含量明显少于模型对照组,差异显著(P<0.05),其血清学指标、肝纤维化分级等方面均出现了不同程度的改善。结论:三七总苷能够降低大鼠胶原沉积,减少DMN所致肝纤维化大鼠胶原含量

    旨在建立基于大肠杆菌表达系统的高效可溶性表达人鳞状上皮细胞癌抗原(SCCAg)方法,获得具有较好活性的重组SCCAg抗原并应用于建立抗原检测方法; 。基于pGEX-6P-l载体和大肠杆菌E. coli; ER2566菌株开展重组SCCAg抗原可溶性表达纯化方法研究,评价纯化抗原活性,筛选特异性单克隆抗体,初步建立并评价SCCAg抗原检测方法。结果; 显示,pGEX-6P-l载体和E coli; ER2566菌株可用于建立较高效的可溶性表达和纯化SCCAg抗原的方法,获得了具有较高纯度和活性的重组SCCAg抗原,筛选获得特异性单克隆抗体并; 初步建立了 SCCAg管式化学发光检测方法。建立了有效的基于大肠杆菌表达系统的可溶性表达和纯化SCCAg的方法。The aims of this study are to establish a method for efficient soluble; expression of human squamous cell carcinoma antigen(SCCAg ) based on; Escherichia coli expression system and obtain the recombinant SCCAg; antigen in fine activity, then apply it in the detection method; establishment of antigen. The study on the method of soluble expression; and purification of recombinant SCCAg antigen was conducted based on; pGEX-6P-l vector and E. coli ER2566 strain. The activity of the purified; antigen was evaluated by Abbott Kit and the specific monoclonal antibody; was screened by indirect ELISA. It was proved that PGEX-6P-1 vector and; E. coli strain ER2566 could be used to establish efficient soluble; expression and purification method for recombinant SCCAg antigen.; Moreover, the recombinant SCCAg antigen was proved to be in high purity; and activity. Thus,the SCCAg detection method of chemical luminous tube; was established with the specific monoclonal antibodies.     Development of detection kit for T cell infected with Mycobacterium tuberculosis based on fullautomatic chemiluminescence immune analyzer

    目的建立基于化学发光平台的人结核感染T细胞检测方法。方法将一株γ-干扰素单抗标记在磁微粒上,另一株γ-干扰素单抗标记在吖啶酯上,然后将检测体系与已有的细胞刺激培养体系相结合。结果本研究成功建立基于化学发光平台的人结核感染T细胞检测方法。以ElISA平台检测试剂的检测结果作为参考,该方法的灵敏度为98.3%,特异性为99.2%,总体符合率达到98.8%。结论该方法具有更高的分析灵敏度(可达0.27 Pg/M l)、更宽的线性范围(1 Pg/M l~5 000 Pg/M l)、更好的重复性(批内与批间变异系数均<6.0%)及更易实现高通量检测,为临床诊断结核感染提供了有力的工具。Objective To develop the detection kit for T cell infected with Mycobacterium tuberculosis based on full-automatic chemiluminescence immune analyzer.Mehtods An anti IFN-γ Mab was coated on the surface of microparticle.Another anti IFN-γ Mab was labelled to acridinium ester.After that, the detection system was combined with the in vitro cell culture system.Results This research developed the detection kit for T cell infected with Mycobacterium tuberculosis based on full-automatic chemiluminescence immune analyzer.Compared with the testing results of the ELISA kit, the sensitivity, specificity and total matching ratio of the CLIA kit was 98.3%, 99.2% and 98.8%, respectively.Conclusion The CLIA kit has better sensitivity(0.27 pg/m L), wilder linear range(1 pg/m L ~ 5 000 pg/m L), and better repeatability(intra and inter coefficient of variation < 6.0%).It makes a high throughput detection available.It will contribute to the clinical diagnosis of Mycobacterium tuberculosis.国家高技术研究发展计划(863计划)(2011AA02A101
