IMPDH2 promotes migration and invasion in cancer cells by sumoylation

IMPDH2是GTP从头合成代谢的关键酶，在多种肿瘤组织和分生活跃的组织中高表达。IMPDH2可以通过SUMO化修饰调节自身酶活。在癌细胞中，IMPDH2的SUMO化促进酶活并通过调节细胞内GTP的含量发挥生物学功能。细胞内含有众多的小GTP蛋白(smallGTPase)，这些蛋白通过与GTP结合从而处于活化状态。活化的小GTP蛋白可以与其下游效应蛋白结合，启动或者传递信号以发挥生物学功能。其中，Rho蛋白家族的主要成员RhoA、cdc42等参与了肌动蛋白的聚合以及与之相关的生物学进程，比如黏附，迁移与侵袭。 本文通过一系列实验发现，SUMO化的IMPDH2可以通过提高自身酶活，在肿瘤细胞内...IMPDH2 is the rate-limiting enzyme of GTP de novo synthesis,which is overexpressed in many kinds of tumor tissues and normal tissues with high proliferation.
