IgM-producing tumors in the BALB/c mouse: a model for B-cell maturation

Five adjuvant induced BALB/c tumors producing IgM—McPc 1748, W 3469, TEPC 183, McPc 774, and Y 5781—were characterized morphologically by electron microscopy, analysis of the distribution of surface-bound and intracytoplasmic IgM using immunofluorescence, and by biochemical study of IgM synthesis, turnover, and secretion. The cells of different tumors appear to represent different stages in B-cell maturation when compared to normal, lipopolysaccharide-stimulated B cells. Thus, McPc 1748 tumor cells resemble 10–25-h stimulated normal B cells, 3469 cells resemble 20–35-h stimulated B cells, TEPC 183 cells resemble 45–65-h stimulated B cells, Y 5781 cells resemble 80–110-h stimulated B cells, and McPc 774 cells resemble 100–130-h stimulated B cells

Shared Decision Making door Arts, Patiënt en Geestelijk Begeleider. Een verkenning van de mogelijke bijdrage van de geestelijk begeleider aan het gezamenlijk besluitvormingsproces in de medische praktijk

Shared Decision Making (SDM) is een besluitvormingsmodel dat wordt aangemerkt als de ideale manier om tot een gezamenlijk besluit te komen, maar dat moeizaam geïmplementeerd blijkt te worden in de medische praktijk. Dit afstudeeronderzoek gaat in op de vraag welke bijdrage de geestelijk begeleider kan leveren aan het bevorderen van het ideaal van SDM in de medische praktijk. Aan de hand van belemmerende en bevorderende factoren voor de implementatie van SDM wordt inzicht verschaft in een mogelijke verbinding tussen SDM en geestelijke begeleiding om zowel de implementatie van SDM te bevorderen alsook een aanvulling op de werkzaamheden van de geestelijk begeleider te introduceren. Op basis van een literatuurstudie en Nederlandse casuïstiek wordt aannemelijk gemaakt dat de geestelijk begeleider zowel in theorie als in praktijk een positieve bijdrage kan leveren aan het bevorderen van SDM. Uit de resultaten blijkt onder meer dat een bijdrage van de geestelijk begeleider aan het individuele keuzeproces, waar de focus van dit onderzoek op ligt, niet los gezien kan worden van de rol die geestelijke begeleiding speelt op instellingsniveau. Ter validatie en specificatie van de onderzoeksresultaten wordt vervolgonderzoek aangeraden

Murine CD8+ T cells that specifically delete autologous CD4+ T cells expressing Vβ8 TCR: a role of the Qa-1 molecule

SummaryInteractions mediated by TCRs expressed on different T cell subsets may play a role in immunoregulation. To investigate this idea, we studied the regulation of superantigen-induced TCR Vβ-restricted responses. We asked whether the in vivo regulation of CD4+ Vβ8+ T cells following SEB injection is controlled by CD8+ T cells. We found that in mice deficient in CD8+ T cells, the down-regulation of CD4+ Vβ8+ T cells below baseline is not observed. Moreover, following SEB administration, CD8+ T cells emerge that preferentially kill subpopulations of activated CD4+ Vβ8+ but not CD4+ Vβ8- T cells in vitro. This TCR Vβ-specific cytotoxicity is dependent on β2-microglobulin and is inhibited by antisera specific for Qa-1 but not by antibody to MHC class la. These data suggest the idea that the specificity of immune regulation may involve CD8+ T cell recognition of TCR Vβ determinants and Qa-1 molecules expressed on CD4+ T cells
