    Developing vocational practice in the jewellery sector through the incubation of a new “project-object”

    This paper analyses a work placement scheme established to create the conditions to: (i) incubate new designs in the jewellery sector in Birmingham; (ii) support a jewellery company compete more effectively in the global market; (iii) assist a newly qualified graduate jeweller to enter the jewellery sector. It introduces a new theoretical framework based on concepts from: Cultural-Historical Activity Theory-'project-object'; Workplace Learning-'vocational practice'; Philosophy of Mind-'space of reasons'; to analyse individual and organisational contributions to workplace learning in this scheme. It identifies the strategies and tactics used by: (i) the organisations involved with the scheme to facilitate the incubation of the new designs, and (ii) an aspiring jewellery designer to create a new product range. The paper concludes with a discussion of the implications of this conceptual framework for debates in workplace learning about: (i) the 'front-loaded' versus 'practice-based' conceptions of vocational education and (ii) the role of epistemic objects and practices in the development of vocational practice. © 2008 Elsevier Ltd.     'We are trying to reproduce a crafts apprenticeship': from Government Blueprint to workplace-generated apprenticeship in the knowledge economy

    Recent research has shown that the UK's Advanced Apprenticeship Programme (AAP) struggles to develop the forms of ‘vocational practice’, that is, a combination of knowledge, skill and judgement, employers are looking for in the creative and cultural sector. Employers' reluctance to get involved with the AAP does not mean that they are uninterested in training. They are concerned that the UK's Department of Education and Skills promotes the AAP to serve ‘educational’ goals (i.e. route for academic progression), rather than functioning as a genuine attempt to develop the sector-specific vocational knowledge and skill that they feel it is important for apprentices to develop. To understand why many employers distance themselves from the AAP, the paper compares and contrasts the AAP with the ‘Technical Apprenticeship’, which has been developed by Birmingham Repertory Theatre, with regard to the different notions of skill formation, skill transfer and employability. It concludes by raising a number of questions and issues as regards: (1) the future development of apprenticeship in the creative and cultural sector in the UK and internationally, and (2) the concept of vocational practice in Vocational Education and Training. © 2007 The Vocational Aspect of Education Ltd

    Response Collector: A Video Learning System for Flipped Classrooms

    The flipped classroom has become famous as an effective educational method that flips the purpose of classroom study and homework. In this paper, we propose a video learning system for flipped classrooms, called Response Collector, which enables students to record their responses to preparation videos. Our system provides response visualization for teachers and students to understand what they have acquired and questioned. We performed a practical user study of our system in a flipped classroom setup. The results show that students preferred to use the proposed method as the inputting method, rather than naive methods. Moreover, sharing responses among students was helpful for resolving individual students' questions, and students were satisfied with the use of our system.Comment: The 2018 International Conference On Advanced Informatics: Concepts, Theory And Application (ICAICTA2018

    The AAP gene family for amino acid permeases contributes to development of the cyst nematode Heterodera schachtii in roots of Arabidopsis

    The beet cyst nematode Heterodera schachtii is able to infect Arabidopsis plants and induce feeding sites in the root. These syncytia are the only source of nutrients for the nematodes throughout their life and are a nutrient sink for the host plant. We have studied here the role of amino acid transporters for nematode development. Arabidopsis contains a large number of different amino acid transporters in several gene families but those of the AAP family were found to be especially expressed in syncytia. Arabidopsis contains 8 AAP genes and they were all strongly expressed in syncytia with the exception of AAP5 and AAP7, which were slightly downregulated. We used promoter::GUS lines and in situ RT-PCR to confirm the expression of several AAP genes and LHT1, a lysine- and histidine-specific amino acid transporter, in syncytia. The strong expression of AAP genes in syncytia indicated that these transporters are important for the transport of amino acids into syncytia and we used T-DNA mutants for several AAP genes to test for their influence on nematode development. We found that mutants of AAP1, AAP2, and AAP8 significantly reduced the number of female nematodes developing on these plants.     Software Quality Assurance

    Telecom networks are composed of very complex software-controlled systems. In recent years, business and technology needs are pushing vendors towards service agility where they must continuously develop, deliver, and improve such software over very short cycles. Moreover, being critical infrastructure, Telecom systems must meet important operational, legal, and regulatory requirements in terms of quality and performance to avoid outages. To ensure high quality software, processes and models must be put in place to enable quick and easy decision making across the development cycle. In this chapter, we will discuss the background and recent trends in software quality assurance. We will then introduce BRACE: a cloud-based, fully-automated tool for software defect prediction, reliability and availability modeling and analytics. In particular, we will discuss a novel Software Reliability Growth Modeling (SRGM) algorithm that is the core of BRACE. The algorithm provides defect prediction for both early and late stages of the software development cycle. To illustrate and validate the tool and algorithm, we also discuss key use cases, including actual defect and outage data from two large-scale software development projects from telecom products. BRACE is being successfully used by global teams of various large-scale software development projects

    Gateway-Compatible CRISPR-Cas9 Vectors and a Rapid Detection by High-Resolution Melting Curve Analysis

    CRISPR-Cas9 system rapidly became an indispensable tool in plant biology to perform targeted mutagenesis. A CRISPR-Cas9-mediated double strand break followed by non-homologous end joining (NHEJ) repair most frequently results in a single base pair deletion or insertions (indels), which is hard to detect using methods based on enzymes that detect heteroduplex DNA. In addition, somatic tissues of the T1 generation inevitably contain a mosaic population, in which the portion of cells carrying the mutation can be too small to be detected by the enzyme-based methods. Here we report an optimized experimental protocol for detecting Arabidopsis mutants carrying a CRISPR-Cas9 mediated mutation, using high-resolution melting (HRM) curve analysis. Single-base pair insertion or deletion (indel) can be easily detected using this method. We have also examined the detection limit for the template containing a one bp indel compared to the WT genome. Our results show that <5% of mutant DNA containing one bp indel can be detected using this method. The vector developed in this study can be used with a Gateway technology-compatible derivative of pCUT vectors, with which off-target mutations could not be detected even by a whole genome sequencing

    Insect-induced daidzein, formononetin and their conjugates in soybean leaves.

    In response to attack by bacterial pathogens, soybean (Gylcine max) leaves accumulate isoflavone aglucones, isoflavone glucosides, and glyceollins. In contrast to pathogens, the dynamics of related insect-inducible metabolites in soybean leaves remain poorly understood. In this study, we analyzed the biochemical responses of soybean leaves to Spodoptera litura (Lepidoptera: Noctuidae) herbivory and also S. litura gut contents, which contain oral secretion elicitors. Following S. litura herbivory, soybean leaves displayed an induced accumulation of the flavone and isoflavone aglycones 4',7-dihyroxyflavone, daidzein, and formononetin, and also the isoflavone glucoside daidzin. Interestingly, foliar application of S. litura oral secretions also elicited the accumulation of isoflavone aglycones (daidzein and formononetin), isoflavone 7-O-glucosides (daidzin, ononin), and isoflavone 7-O-(6'-O-malonyl-β-glucosides) (malonyldaidzin, malonylononin). Consistent with the up-regulation of the isoflavonoid biosynthetic pathway, folair phenylalanine levels also increased following oral secretion treatment. To establish that these metabolitic changes were the result of de novo biosynthesis, we demonstrated that labeled (13C9) phenylalanine was incorporated into the isoflavone aglucones. These results are consistent with the presence of soybean defense elicitors in S. litura oral secretions. We demonstrate that isoflavone aglycones and isoflavone conjugates are induced in soybean leaves, not only by pathogens as previously demonstrated, but also by foliar insect herbivory
