Linefish resources annual report for the year 2000. Part 1: fisheries assessment
This work reflects the activities of line and trap fishing in Southern Mozambique in 2000. The catch in line fishing has been estimated at 441 mt, according to the DNAP records. The same sources indicated that 1767 days were spent at sea and the estimated catch rate was 250 Kg per boaticlay. Most of the line fishing effort shifted away from Maputo and moved to Inhambane region. The monthly analysis of fishing vessels, stricter controls over catch and effort data submission, development of long-term research programme and the continuation of the on board sampling to improve the data collection are the recommendation for line fishing. The catch of trap vessels increases from 30mt in 1997 to 172 mt in 2000, during which the total number of traps increased from 25 to 300. During this time the number of fishing days has remained relatively constant, as well the soak time. These data sets are thus not compatible with each other, reflecting an increase in daily catch from 243 Kg to 791 kg. The species composition is mainly dominated by P. coeruleopunctatus, C. puniceus, C. nufar and E. andersoni

“Facing Our Fears”: Using facilitated film viewings to engage communities in HIV research involving MSM in Kenya
Kenya is a generally homophobic country where homosexuality is
criminalised and people who engage in same sex sexuality face stigma and
discrimination. In 2013, we developed a 16Â min documentary entitled "Facing Our Fears"
that aimed at sharing information on how and why men who have sex with
men (MSM) are involved in on-going KEMRI HIV prevention research, and
associated community engagement. To consider the film's usefulness as a
communication tool, and its perceived security risks in case the film
was publicly released, we conducted nine facilitated viewings with 122
individuals representing seven different stakeholder groups. The
documentary was seen as a strong visual communication tool with
potential to reduce stigma related to homosexuality, and facilitated
film viewings were identified as platforms with potential to support
open dialogue about HIV research involving MSM. Despite the potential,
there were concerns over possible risks to LGBT communities and those
working with them following public release. We opted-giving emphasis to
the "do no harm" principle-to use the film only in facilitated settings
where audience knowledge and attitudes can be carefully considered and
discussed. The results highlight the importance of carefully assessing
the range of possible impacts when using visuals in community
engagement.</p
Transcriptional signatures of somatic neoblasts and germline cells in <i>Macrostomum lignano</i>
The regeneration-capable flatworm <i>Macrostomum lignano</i> is a powerful model organism to study the biology of stem cells in vivo. As a flatworm amenable to transgenesis, it complements the historically used planarian flatworm models, such as <i>Schmidtea mediterranea</i>. However, information on the transcriptome and markers of stem cells in <i>M. lignano</i> is limited. We generated a de novo transcriptome assembly and performed the first comprehensive characterization of gene expression in the proliferating cells of <i>M. lignano</i>, represented by somatic stem cells, called neoblasts, and germline cells. Knockdown of a selected set of neoblast genes, including <i>Mlig-ddx39</i>, <i>Mlig-rrm1</i>, <i>Mlig-rpa3</i>, <i>Mlig-cdk1</i>, and <i>Mlig-h2a</i>, confirmed their crucial role for the functionality of somatic neoblasts during homeostasis and regeneration. The generated <i>M. lignano</i> transcriptome assembly and gene expression signatures of somatic neoblasts and germline cells will be a valuable resource for future molecular studies in <i>M. lignano</i>
Caspofungin Weight-Based Dosing Supported by a Population Pharmacokinetic Model in Critically Ill Patients
The objective of this study was to develop a population pharmacokinetic model and to determine a dosing regimen for caspofungin in critically ill patients. Nine blood samples were drawn per dosing occasion. Fifteen patients with (suspected) invasive candidiasis had one dosing occasion and five had two dosing occasions, measured on day 3 (±1) of treatment. Pmetrics was used for population pharmacokinetic modeling and probability of target attainment (PTA). A target 24-h area under the concentration-time curve (AUC) value of 98 mg·h/liter was used as an efficacy parameter. Secondarily, the AUC/MIC targets of 450, 865, and 1,185 were used to calculate PTAs for Candida glabrata, C. albicans, and C. parapsilosis, respectively. The final 2-compartment model included weight as a covariate on volume of distribution (V). The mean V of the central compartment was 7.71 (standard deviation [SD], 2.70) liters/kg of body weight, the mean elimination constant (Ke) was 0.09 (SD, 0.04) h-1, the rate constant for the caspofungin distribution from the central to the peripheral compartment was 0.44 (SD, 0.39) h-1, and the rate constant for the caspofungin distribution from the peripheral to the central compartment was 0.46 (SD, 0.35) h-1. A loading dose of 2 mg/kg on the first day, followed by 1.25 mg/kg as a maintenance dose, was chosen. With this dose, 98% of the patients were expected to reach the AUC target on the first day and 100% of the patients on the third day. The registered caspofungin dose might not be suitable for critically ill patients who were all overweight (≥120 kg), over 80% of median weight (78 kg), and around 25% of lower weight (≤50 kg). A weight-based dose regimen might be appropriate for achieving adequate exposure of caspofungin in intensive care unit patients
Autonomous decision-making against induced seismicity in deep fluid injections
The rise in the frequency of anthropogenic earthquakes due to deep fluid
injections is posing serious economic, societal, and legal challenges to
geo-energy and waste-disposal projects. We propose an actuarial approach to
mitigate this risk, first by defining an autonomous decision-making process
based on an adaptive traffic light system (ATLS) to stop risky injections, and
second by quantifying a "cost of public safety" based on the probability of an
injection-well being abandoned. The ATLS underlying statistical model is first
confirmed to be representative of injection-induced seismicity, with examples
taken from past reservoir stimulation experiments (mostly from Enhanced
Geothermal Systems, EGS). Then the decision strategy is formalized: Being
integrable, the model yields a closed-form ATLS solution that maps a risk-based
safety standard or norm to an earthquake magnitude not to exceed during
stimulation. Finally, the EGS levelized cost of electricity (LCOE) is
reformulated in terms of null expectation, with the cost of abandoned
injection-well implemented. We find that the price increase to mitigate the
increased seismic risk in populated areas can counterbalance the heat credit.
However this "public safety cost" disappears if buildings are based on
earthquake-resistant designs or if a more relaxed risk safety standard or norm
