    Does teachers' pedagogical content knowledge affect their fluency instruction?

    The relation is studied between teachers' pedagogical content knowledge of reading and the quality of their subsequent classroom behaviour in teaching fluent reading. A confirmatory factor analysis model with two latent variables is tested and shows adequate goodness-of-fit indices. Contrary to our expectations, the results of structural equation modelling reveal a small but significant gamma-value of .29, indicating that only 8% of the variance in teachers' classroom behaviour in teaching fluent reading is accounted for by teachers' pedagogical content knowledge of reading. Presumably teacher knowledge is not as stable and conclusive as one might think. More research is needed in determining the factors that work restricting for teachers in putting their knowledge into classroom practice. It is recommended that preservice and in-service teacher training should not be limited to transfer of knowledge, but should also assist teachers in designing and performing effective fluent reading instruction

    Genetic diversity of the dengue vector Aedes aegypti in Australia and implications for future surveillance and mainland incursion monitoring

    In February 2004, the discovery of an incursion of the dengue vector Aedes aegypti into the town of Tennant Creek in the Northern Territory caused concern for the Northern Territory health authorities who proceeded to implement a Commonwealth-funded eradication program. To determine the origin of the incursion, we performed a genetic analysis on Ae. aegypti from several Queensland and overseas localities. A comparison of DNA sequences from the mitochondrial cytochrome oxidase 1 gene indicated that the incursion was probably from Cairns or Camooweal. This genetic marker was also useful in identifying a separate Townsville haplotype population and another population on Thursday Island in the Torres Strait that is genetically distant to the mainland populations. The possible use of this marker as a surveillance tool for identifying the origins of local and overseas incursions is discussed

    Don’t look back in anger: the rewarding value of a female face is discounted by an angry expression

    The modulating effect of emotional expression on the rewarding nature of attractive and nonattractive female faces in heterosexual men was explored in a motivated viewing paradigm. This paradigm, which is an indicator of neural reward, requires the viewer to expend effort to maintain or reduce image-viewing times. Males worked to extend the viewing time for happy and neutral attractive faces but to reduce the viewing time for the attractive angry faces. Attractive angry faces were rated as more aesthetically pleasing than the nonattractive faces; however, the males worked to reduce their viewing time to a level comparable with the nonattractive neutral and happy faces. Therefore, the addition of an angry expression onto an otherwise attractive face renders it unrewarding and aversive to potential mates. Mildly happy expressions on the nonattractive faces did little to improve their attractiveness or reward potential, with males working to reduce viewing time for all nonattractive faces

    Evaluation of European Land Data Assimilation System (ELDAS) products using in site observations

    Three land-surface models with land-data assimilation scheme (DA) were evaluated for one growing season using in situ observations obtained across Europe. To avoid drifts in the land-surface state in the models, soil moisture corrections are derived from errors in screen-level atmospheric quantities. With the in situ data it is assessed whether these land-surface schemes produce adequate results regarding the annual range of the soil water content, the monthly mean soil moisture content in the root zone and evaporative fraction (the ratio of evapotranspiration to energy available at the surface). DA considerably reduced bias in net precipitation, while slightly reducing RMSE as well. Evaporative fraction was improved in dry conditions but was hardly affected in moist conditions. The amplitude of soil moisture variations tended to be underestimated. The impact of improved land-surface properties like Leaf Area Index, water holding capacity and rooting depth may be as large as corrections of the DA systems. Because soil moisture memorizes errors in the hydrological cycle of the models, DA will remain necessary in forecast mode. Model improvements should be balanced against improvements of DA per se. Model bias appearing from persistent analysis increments arising from DA systems should be addressed by model improvement

    Hunting before herding:A zooarchaeological and stable isotopic study of suids (Sus sp.) at Hardinxveld-Giessendam, the Netherlands (5450–4250 cal BC)

    Suids (Sus sp.) played a crucial role in the transition to farming in northern Europe and, like in many regions, in the Netherlands pig husbandry became an important subsistence activity at Neolithic sites. Yet little is known about wild boar palaeoecology and hunting in the Late Mesolithic Netherlands with which to contextualize this transition. This paper presents the first multi-proxy analysis of archaeological suid remains in the Netherlands. It explores human-suid interactions at the Swifterbant culture sites of Hardinxveld-Giessendam Polderweg and De Bruin (5450–4250 BC) through biometric analysis, estimation of age-at-death, and stable carbon and nitrogen isotope analysis. The results reveal targeted hunting of adult wild boar in the Late Mesolithic (5450–4850 BC), with a possible shift over time towards more juveniles. The wild boar in this period are demonstrated to be of comparably large size to contemporary northern European populations and exhibiting a wide range of dietary regimes. In the final occupational period (4450–4250 BC), small suids are present, possibly domestic pigs, but there is no evidence of pig management. This study demonstrates that the nature of human-suid interactions varied over time, which may have been connected to changing environmental conditions, human mobility, and wild boar behaviour. This study also contributes the first biometric and dietary baseline for mid-Holocene wild boar in the Netherlands

    Seasonal evaluation of the land surface scheme HTESSEL against remote sensing derived energy fluxes of the Transdanubian region in Hungary

    The skill of the land surface model HTESSEL is assessed to reproduce evaporation in response to land surface characteristics and atmospheric forcing, both being spatially variable. Evaporation estimates for the 2005 growing season are inferred from satellite observations of the Western part of Hungary and compared to model outcomes. Atmospheric forcings are obtained from a hindcast run with the Regional Climate Model RACMO2. Although HTESSEL slightly underpredicts the seasonal evaporative fraction as compared to satellite estimates, the mean, 10th and 90th percentile of this variable are of the same magnitude as the satellite observations. The initial water as stored in the soil and snow layer does not have a significant effect on the statistical properties of the evaporative fraction. However, the spatial distribution of the initial soil and snow water significantly affects the spatial distribution of the calculated evaporative fraction and the models ability to reproduce evaporation correctly in low precipitation areas in the considered region. HTESSEL performs weaker in dryer areas. In Western Hungary these areas are situated in the Danube valley, which is partly covered by irrigated cropland and which also may be affected by shallow groundwater. Incorporating (lateral) groundwater flow and irrigation, processes that are not included now, may improve HTESSELs ability to predict evaporation correctly. Evaluation of the model skills using other test areas and larger evaluation periods is needed to confirm the results. <br><br> Based on earlier sensitivity analysis, the effect of a number of modifications to HTESSEL has been assessed. A more physically based reduction function for dry soils has been introduced, the soil depth is made variable and the effect of swallow groundwater included. However, the combined modification does not lead to a significantly improved performance of HTESSEL

    Comparative susceptibility of mosquito populations in North Queensland, Australia to oral infection with dengue virus.

    Dengue is the most prevalent arthropod-borne virus, with at least 40% of the world's population at risk of infection each year. In Australia, dengue is not endemic, but viremic travelers trigger outbreaks involving hundreds of cases. We compared the susceptibility of Aedes aegypti mosquitoes from two geographically isolated populations to two strains of dengue virus serotype 2. We found, interestingly, that mosquitoes from a city with no history of dengue were more susceptible to virus than mosquitoes from an outbreak-prone region, particularly with respect to one dengue strain. These findings suggest recent evolution of population-based differences in vector competence or different historical origins. Future genomic comparisons of these populations could reveal the genetic basis of vector competence and the relative role of selection and stochastic processes in shaping their differences. Lastly, we show the novel finding of a correlation between midgut dengue titer and titer in tissues colonized after dissemination

    Patch-Seq Protocol to Analyze the Electrophysiology, Morphology and Transcriptome of Whole Single Neurons Derived From Human Pluripotent Stem Cells

No use, distribution or reproduction is permitted which does not comply with these terms.The human brain is composed of a complex assembly of about 171 billion heterogeneous cellular units (86 billion neurons and 85 billion non-neuronal glia cells). A comprehensive description of brain cells is necessary to understand the nervous system in health and disease. Recently, advances in genomics have permitted the accurate analysis of the full transcriptome of single cells (scRNA-seq). We have built upon such technical progress to combine scRNA-seq with patch-clamping electrophysiological recording and morphological analysis of single human neurons in vitro. This new powerful method, referred to as Patch-seq, enables a thorough, multimodal profiling of neurons and permits us to expose the links between functional properties, morphology, and gene expression. Here, we present a detailed Patch-seq protocol for isolating single neurons from in vitro neuronal cultures. We have validated the Patch-seq whole-transcriptome profiling method with human neurons generated from embryonic and induced pluripotent stem cells (ESCs/iPSCs) derived from healthy subjects, but the procedure may be applied to any kind of cell type in vitro.
