    Interactive drama in real and virtual worlds.

    How do we resolve the paradox of computer-supported interactive drama - that the human participant requires the very freedom to interact that the authored narrative structure denies them? This paper reports work around the concept of Emergent Narrative - the development of narrative structure through interaction itself. We cover both systems using a virtual world and those using a virtually-augmented real world, exploring how far reworking narrative structure as a loop between the causal (plot) and affective (character) can produce engaging experiences for participants. We discuss the key role of a cognitive-affective architecture for charac-ters and the process of cognitive appraisal as an engine for both in-character and in-role dramatic action

    All the choice and all the responsibilities: an exploration of the agency perceived by women with children around their childbearing

    Findings from an exploratory study of agency around childbearing perceived by women with children from a feminist perspective are presented. Contemporary women‘s childbearing has confusedly been portrayed as chosen but constrained and also a duty. Most recently women have been considered to have a duty to reproduce to stimulate economic growth. Furthermore, a plethora of fertility theories have been put forward to explain recent declining fertility but these have found to be incapable of explaining fertility trends and the complexity of childbearing negotiations. Amongst these deliberations women‘s interests tend to get lost. This research attempts to reverse that tendency. Taking part in this research were 26 women each of whom had at least one child of nine years of age or younger from in and around Orange, NSW. Data was collected using in-depth semi-structured interviews and focus groups. As agency is ill-defined within sociology, seven criteria for recognising agency were constructed from a review of sociological theory. These criteria were used as a series of lenses through which the data were viewed; allowing for the explanatory powers of theories to be compared. The findings show that women‘s agency around childbearing was intermittent and imperfect but not completely absent. Important to the discourse of choice prevalent in the popular media and expounded by economic rationalists, the concept of choice was shown to be simplistic. Rather the women described complex negotiations between biological factors, social influences and personal preferences. This research demonstrates that despite making some progress in workforce participation, women‘s expected role in the home curtailed what they could achieve. The thesis furthers understanding of women‘s childbearing agency, has implications for public policy, provides insights into the relevance of sociological theories to women who have children and provides a novel methodological approach for assessing agency

    Expanded Parameters in the Self-Organized Critical Forest Fire Model

    The forest fire model has been used to test the theory of Self-Organized Criticality as a model of complexity. The goal is to search for scale invariance in randomly generated forest fires using a computer simulation. In a previous model by B. Drossel and F. Schwabl,1 power-law behavior was seen when the nearest neighbors to a tree on fire catch on fire, and it has been assumed that if further trees also catch fire, then it will still exhibit self-organized criticality, showing scale invariance. Testing this assumption aids to the exploration of the applicability of self-organized criticality because the model is the most useful when it applies to a large range of systems, as closely related to nature as possible. 1 B.     Citrullination of synovial proteins in murine models of rheumatoid arthritis

    Item does not contain fulltextOBJECTIVE: Antibodies directed to citrulline-containing proteins are highly specific for rheumatoid arthritis (RA) and can be detected in up to 80% of patients with RA. Citrulline is a nonstandard amino acid that can be incorporated into proteins only by posttranslational modification of arginine by peptidylarginine deiminase (PAD) enzymes. The objective of this study was to investigate the presence of anticitrulline antibodies, PAD enzymes, and citrullinated antigens in mouse models of both acute and chronic destructive arthritis: streptococcal cell wall (SCW)-induced arthritis and collagen-induced arthritis (CIA), respectively. METHODS: Synovial tissue biopsy specimens were obtained from naive mice, mice with CIA, and mice with SCW-induced arthritis. The expression of messenger RNA (mRNA) for PAD enzymes was analyzed by reverse transcriptase-polymerase chain reaction; the presence of PAD proteins and their products (citrullinated proteins) was analyzed by Western blotting and by immunolocalization. The presence of anticitrullinated protein antibodies was investigated by an anti-cyclic citrullinated peptide (anti-CCP) enzyme-linked immunosorbent assay (ELISA) and an ELISA using in vitro citrullinated fibrinogen. RESULTS: In both mouse models, PAD type 2 (PAD2) mRNA was present in the synovium but was not translated into PAD2 protein. In contrast, PAD4 mRNA, although absent from healthy synovium, was readily transcribed and translated by polymorphonuclear neutrophils infiltrating the synovial tissue during inflammation. As a consequence, several synovial proteins were subjected to citrullination. One of these proteins was identified as fibrin, which has been reported to be citrullinated also in synovium of patients with RA. Although generation of citrullinated antigens during synovial inflammation in the mice was eminent, no anti-CCP antibodies could be detected. CONCLUSION: Citrullination of synovial antigens is an active process during joint inflammation in both mice and humans, but the induction of autoantibodies directed to these proteins is a more specific phenomenon, detectable only in human RA patients
