    Recolección de especies silvestres de papa en Guatemala.

    El objetivo de este trabajo fue recolectar germoplasma de especies silvestres de papa, para su conservación, e incremento para ponerlas a disposición a nivel internacional, y reunir datos de campo para continuar con los estudios taxonómicos. De Solanum agrimonifolium se efectuaron 13 recolectas en los departamentos de Huehuetenango, San Marcos, Quetzaltenango, Totonicapan, Sololá y Chimaltenango en altitudes comprendidas de 2.340 a 3.300 msnm; de S. bulbocastanum subsp. partitum se obtuvieron seis recolectas en los departamentos de Huehuetenango y Baja Verapaz; de S. clarum se obtuvieron 17 recolectas en los departamentos de Huehuetenango, San Marcos, Sololá y Totonicapan, en altitudes de 3,000 a 3,500 msnm; de S. demissum solo se encontró una localidad en Totonicapan; y de S. morelliforme se obtuvieron seis recolectas de los departamentos de Huehuetenango, San Marcos y Totonicapan en alturas de 2.900 a 3.050 msnm. Los hábitats, donde estas especies crecen, están muy deteriorados por la deforestación y el cambio de uso del suelo. Se plantean dos áreas para la conservación in situ por la concentración de especies silvestres de papa y porque aún mantienen determinadas áreas casi naturales: 1) La Cumbre de María Tecún en el departamento de Totonicapan, y 2) áreas dispersas en la Sierra de los Cuchumatanes

    AFLP analysis reveals a lack of phylogenetic structure within Solanum section Petota

    <p>Abstract</p> <p>Background</p> <p>The secondary genepool of our modern cultivated potato (<it>Solanum tuberosum </it>L.) consists of a large number of tuber-bearing wild <it>Solanum </it>species under <it>Solanum </it>section <it>Petota</it>. One of the major taxonomic problems in section <it>Petota </it>is that the series classification (as put forward by Hawkes) is problematic and the boundaries of some series are unclear. In addition, the classification has received only partial cladistic support in all molecular studies carried out to date.</p> <p>The aim of the present study is to describe the structure present in section <it>Petota</it>. When possible, at least 5 accessions from each available species and 5 individual plants per accession (totally approx. 5000 plants) were genotyped using over 200 AFLP markers. This resulted in the largest dataset ever constructed for <it>Solanum </it>section <it>Petota</it>. The data obtained are used to evaluate the 21 series hypothesis put forward by Hawkes and the 4 clade hypothesis of Spooner and co-workers.</p> <p>Results</p> <p>We constructed a NJ tree for 4929 genotypes. For the other analyses, due to practical reasons, a condensed dataset was created consisting of one representative genotype from each available accession. We show a NJ jackknife and a MP jackknife tree. A large part of both trees consists of a polytomy. Some structure is still visible in both trees, supported by jackknife values above 69. We use these branches with >69 jackknife support in the NJ jackknife tree as a basis for informal species groups. The informal species groups recognized are: Mexican diploids, Acaulia, Iopetala, Longipedicellata, polyploid Conicibaccata, diploid Conicibaccata, Circaeifolia, diploid Piurana and tetraploid Piurana.</p> <p>Conclusion</p> <p>Most of the series that Hawkes and his predecessors designated can not be accepted as natural groups, based on our study. Neither do we find proof for the 4 clades proposed by Spooner and co-workers. A few species groups have high support and their inner structure displays also supported subdivisions, while a large part of the species cannot be structured at all. We believe that the lack of structure is not due to any methodological problem but represents the real biological situation within section <it>Petota</it>.</p

    Relevance of Titin Missense and Non-Frameshifting Insertions/Deletions Variants in Dilated Cardiomyopathy

    Recent advancements in next generation sequencing (NGS) technology have led to the identification of the giant sarcomere gene, titin (TTN), as a major human disease gene. Truncating variants of TTN (TTNtv) especially in the A-band region account for 20% of dilated cardiomyopathy (DCM) cases. Much attention has been focused on assessment and interpretation of TTNtv in human disease; however, missense and non-frameshifting insertions/deletions (NFS-INDELs) are difficult to assess and interpret in clinical diagnostic workflow. Targeted sequencing covering all exons of TTN was performed on a cohort of 530 primary DCM patients from three cardiogenetic centres across Europe. Using stringent bioinformatic filtering, twenty-nine and two rare TTN missense and NFS-INDELs variants predicted deleterious were identified in 6.98% and 0.38% of DCM patients, respectively. However, when compared with those identified in the largest available reference population database, no significant enrichment of such variants was identified in DCM patients. Moreover, DCM patients and reference individuals had comparable frequencies of splice-region missense variants with predicted splicing alteration. DCM patients and reference populations had comparable frequencies of rare predicted deleterious TTN missense variants including splice-region missense variants suggesting that these variants are not independently causative for DCM. Hence, these variants should be classified as likely benign in the clinical diagnostic workflow, although a modifier effect cannot be excluded at this stage.Peer reviewe

    Trivial, Critical and Near-critical Scaling Limits of Two-dimensional Percolation

    It is natural to expect that there are only three possible types of scaling limits for the collection of all percolation interfaces in the plane: (1) a trivial one, consisting of no curves at all, (2) a critical one, in which all points of the plane are surrounded by arbitrarily large loops and every deterministic point is almost surely surrounded by a countably infinite family of nested loops with radii going to zero, and (3) an intermediate one, in which every deterministic point of the plane is almost surely surrounded by a largest loop and by a countably infinite family of nested loops with radii going to zero. We show how one can prove this using elementary arguments, with the help of known scaling relations for percolation. The trivial limit corresponds to subcritical and supercritical percolation, as well as to the case when the density p approaches the critical probability, p_c, sufficiently slowly as the lattice spacing is sent to zero. The second type corresponds to critical percolation and to a faster approach of p to p_c. The third, or near-critical, type of limit corresponds to an intermediate speed of approach of p to p_c. The fact that in the near-critical case a deterministic point is a.s. surrounded by a largest loop demonstrates the persistence of a macroscopic correlation length in the scaling limit and the absence of scale invariance.Comment: 15 pages, 3 figure

    Gastrointestinal biomarkers and their association with feeding in the first five days of pediatric critical illness

    OBJECTIVES: Predicting the patients' tolerance to enteral nutrition (EN) would help clinicians optimize individual nutritional intake. This study investigated the course of several gastrointestinal (GI) biomarkers and their association with EN advancement (ENA) longitudinally during pediatric intensive care unit (PICU) admission.METHODS: This is a secondary analysis of the PEPaNIC RCT. EN was started early and increased gradually. The cholecystokinin (CCK), leptin, glucagon, intestinal fatty acid-binding protein 2 (I-FABP2), and citrulline plasma concentrations were measured upon PICU admission, day three and day five. ENA was defined as kcal EN provided as % of predicted resting energy expenditure (pREE). The course of the biomarkers and ENA was examined in patients with samples on all time points using Friedman and Wilcoxon signed-rank tests. The association of ENA with the biomarkers was examined using a two-part mixed-effects model with data of the complete population, adjusted for possible confounders.RESULTS: For 172 patients, median age 8.6 years (first quartile (Q1); third quartile (Q3): 4.2; 13.4), samples were available, of which 55 had samples on all time points. The median ENA was 0 (0; 0) on admission, 14.5 (0.0; 43.8) on day 3 and 28.0 (7.6; 94.8) on day 5. During PICU stay, CCK and I-FABP2 concentrations decreased significantly, whereas glucagon concentrations increased significantly, and leptin and citrulline remained stable. None of the biomarkers was longitudinally associated with ENA.CONCLUSIONS: Based on the current evidence, CCK, leptin, glucagon, I-FABP2, and citrulline appear to have no added value in predicting ENA in the first five days of pediatric critical illness.</p

    The Frequency of Binary Stars in the Core of 47 Tucanae

    Differential time series photometry has been derived for 46422 main-sequence stars in the core of 47 Tucanae. The observations consisted of near-continuous 160-s exposures alternating between the F555W and F814W filters for 8.3 days in 1999 July with WFPC2 on the Hubble Space Telescope. Using Fourier and other search methods, eleven detached eclipsing binaries and fifteen W UMa stars have been discovered, plus an additional ten contact or near-contact non-eclipsing systems. After correction for non-uniform area coverage of the survey, the observed frequencies of detached eclipsing binaries and W UMa's within 90 arcseconds of the cluster center are 0.022% and 0.031% respectively. The observed detached eclipsing binary frequency, the assumptions of a flat binary distribution with log period and that the eclipsing binaries with periods longer than about 4 days have essentially their primordial periods, imply an overall binary frequency of 13 +/- 6 %. The observed W UMa frequency and the additional assumptions that W UMa's have been brought to contact according to tidal circularization and angular momentum loss theory and that the contact binary lifetime is 10^{9} years, imply an overall binary frequency of 14 +/- 4 %. An additional 71 variables with periods from 0.4 - 10 days have been found which are likely to be BY Draconis stars in binary systems. The radial distribution of these stars is the same as that of the eclipsing binaries and W UMa stars and is more centrally concentrated than average stars, but less so than the blue straggler stars. A distinct subset of six of these stars fall in an unexpected domain of the CMD, comprising what we propose to call red stragglers.Comment: Accepted for publication in the Astrophysical Journal, 65 pages including 26 figure

    What's in a name; Genetic structure in Solanum section Petota studied using population-genetic tools

    Background - The taxonomy and systematic relationships among species of Solanum section Petota are complicated and the section seems overclassified. Many of the presumed (sub)species from South America are very similar and they are able to exchange genetic material. We applied a population genetic approach to evaluate support for subgroups within this material, using AFLP data. Our approach is based on the following assumptions: (i) accessions that may exchange genetic material can be analyzed as if they are part of one gene pool, and (ii) genetic differentiation among species is expected to be higher than within species. Results - A dataset of 566 South-American accessions (encompassing 89 species and subspecies) was analyzed in two steps. First, with the program STRUCTURE 2.2 in an 'unsupervised' procedure, individual accessions were assigned to inferred clusters based on genetic similarity. The results showed that the South American members of section Petota could be arranged in 16 clusters of various size and composition. Next, the accessions within the clusters were grouped by maximizing the partitioning of genetic diversity among subgroups (i.e., maximizing Fst values) for all available individuals of the accessions (2767 genotypes). This two-step approach produced an optimal partitioning into 44 groups. Some of the species clustered as genetically distinct groups, either on their own, or combined with one or more other species. However, accessions of other species were distributed over more than one cluster, and did not form genetically distinct units. Conclusions - We could not find any support for 43 species (almost half of our dataset). For 28 species some level of support could be found varying from good to weak. For 18 species no conclusions could be drawn as the number of accessions included in our dataset was too low. These molecular data should be combined with data from morphological surveys, with geographical distribution data, and with information from crossing experiments to identify natural units at the species level. However, the data do indicate which taxa or combinations of taxa are clearly supported by a distinct set of molecular marker data, leaving other taxa unsupported. Therefore, the approach taken provides a general method to evaluate the taxonomic system in any species complex for which molecular data are available
