    Laser interference lithography with highly accurate interferometric alignment

    Three dimensional photonic crystals, e.g. for obtaining the so-called woodpile structure, can, among others, be fabricated by vertical stacking of multiple gratings. One of the requirements for obtaining a full photonic bandgap in such a photonic crystal is an accurate angular and lateral alignment of the successive gratings. Using laser interference lithography at 266 nm wavelength, we fabricated gratings in silicon with periods down to 300 nm. We present a method for aligning further grating exposures with respect to this grating with a 0.001 degree angular and a few nanometers lateral resolution

    Correction regarding Sulfolobus acidocaldarius strain GA06 Upon resequencing we discovered that strain GA06 was not, as published (van Wolferen et al., 2013), a deletion of the upsX ORF and a 40 bp upstream region, but was a deletion starting 55 bp upstream and ending 35 bp downstream of upsX. Since this could affect the genes downstream of UpsX, a clean deletion mutant of the upsX promoter (all 261 bp upstream of upsX) was created and analysed by qRT-PCR studies as well as by UV-induced aggregation assays. qRT-PCR experiments showed that in this strain, upsE, F, A and B were upregulated normally upon UV-induction, and aggregation experiments showed that the deletion mutant of the upsX promoter aggregated in a similar manner as the WT strain after treatment with UV-light. These results are similar to the results obtained for the deletion of the upsX gene (MW115, ΔupsX, Figures 4 and 5). Therefore, the promoter in front of upsX is not the primary promoter of the ups-operon and is also not essential for transcription of the other ups-genes. Additionally, we discovered that the EM image in Figure 6 (GA06) was accidently replaced by an image of strain (ΔupsEF). Both strains did not show any UV induced pili, but the specific image shown was not obtained for this strain. All other strains and EM pictures and the conclusions obtained with these strains are correct

    Large-scale production of LGR5-positive bipotential human liver stem cells

    Background and Aims: The gap between patients on transplant waiting lists and available donor organs is steadily increasing. Human organoids derived from leucine‐rich repeat‐containing G protein‐coupled receptor 5 (LGR5)–positive adult stem cells represent an exciting new cell source for liver regeneration; however, culturing large numbers of organoids with current protocols is tedious and the level of hepatic differentiation is limited. Approach and Results: Here, we established a method for the expansion of large quantities of human liver organoids in spinner flasks. Due to improved oxygenation in the spinner flasks, organoids rapidly proliferated and reached an average 40‐fold cell expansion after 2 weeks, compared with 6‐fold expansion in static cultures. The organoids repopulated decellularized liver discs and formed liver‐like tissue. After differentiation in spinner flasks, mature hepatocyte markers were highly up‐regulated compared with static organoid cultures, and cytochrome p450 activity reached levels equivalent to hepatocytes. Conclusions: We established a highly efficient method for culturing large numbers of LGR5‐positive stem cells in the form of organoids, which paves the way for the application of organoids for tissue engineering and liver transplantation

    Participatory Budgeting and Local Government in a Vertical Society: A Japanese Story

    This article examines a case of participatory budgeting in Japanese local government. The article demonstrates how cultural values interact with stages of budgeting (in our case, the co-planning or consultation phase of budgeting). We find three key stakeholders – councillors, administrators and citizens – have varying degree of participation in the budget process. While direct citizen participation has been limited and challenging, we find that local associations and councillors work as lobbyists to influence the budget less publicly. The budget desk led by the mayor plays the dominant role. This article contributes to the broader debate on local government reforms and their translation into varied contexts by problematising such a linear adoption of knowledge from a cultural perspective

    Long-Term Adult Feline Liver Organoid Cultures for Disease Modeling of Hepatic Steatosis.

    Hepatic steatosis is a highly prevalent liver disease, yet research is hampered by the lack of tractable cellular and animal models. Steatosis also occurs in cats, where it can cause severe hepatic failure. Previous studies demonstrate the potential of liver organoids for modeling genetic diseases. To examine the possibility of using organoids to model steatosis, we established a long-term feline liver organoid culture with adult liver stem cell characteristics and differentiation potential toward hepatocyte-like cells. Next, organoids from mouse, human, dog, and cat liver were provided with fatty acids. Lipid accumulation was observed in all organoids and interestingly, feline liver organoids accumulated more lipid droplets than human organoids. Finally, we demonstrate effects of interference with β-oxidation on lipid accumulation in feline liver organoids. In conclusion, feline liver organoids can be successfully cultured and display a predisposition for lipid accumulation, making them an interesting model in hepatic steatosis research

    Damage accumulation in thin ruthenium films induced by repetitive exposure to femtosecond XUV pulses below the single shot ablation threshold

    The process of damage accumulation in thin ruthenium films exposed to multiple femtosecond XUV free electron laser FEL pulses below the critical angle of reflectance at the Free electron LASer facility in Hamburg FLASH was experimentally analyzed. The multi shot damage threshold is found to be lower than single shot damage threshold. Detailed analysis of the damage morphology and its dependence on irradiation conditions justifies the assumption that cavitation induced by the FEL pulse is the prime mechanism responsible for multi shot damage in optical coating
