    Data-driven Urban Design: Conceptual and Methodological Interpretations of Negroponte’s ‘Architecture Machine’

    Nicholas Negroponte and MIT’s Architecture Machine Group speculated in the 1970s about computational processes that were open to participation, incorporating end-user preferences and democratizing urban design. Today’s ‘smart city’ technologies, using the monitoring of people’s movement and activity patterns to offer more effective and responsive services, might seem like contemporary interpretations of Negroponte’s vision, yet many of the collectors of user information are disconnected from urban policy making. This article presents a series of theoretical and procedural experiments conducted through academic research and teaching, developing user-driven generative design processes in the spirit of ‘The Architecture Machine’. It explores how new computational tools for site analysis and monitoring can enable datadriven urban place studies, and how these can be connected to generative strategies for public spaces and environments at various scales. By breaking down these processes into separate components of gathering, analysing, translating and implementing data, and conceptualizing them in relation to urban theory, it is shown how data-driven urban design processes can be conceived as an open-ended toolkit to achieve various types of user-driven outcomes. It is argued that architects and urban designers are uniquely situated to reflect on the benefits and value systems that control data-driven processes, and should deploy these to deliver more resilient, liveable and participatory urban spaces

    The Architecture Machine Revisited: Experiments exploring Computational Design-and- Build Strategies based on Participation

    This article summarises a series of experiments at the Architectural Association between 2011 and 2017, which explore the intellectual notion of ‘the architecture machine’ as introduced by Nicholas Negroponte and the Architecture Machine Group at MIT in 1967. The group explored automated computational processes that could assist the process of generating architectural solutions by incorporating much greater levels of complexity at both large and small scales. A central idea to the mission of the Architecture Machine Group was to enable the future inhabitants to participate in the decision-making process on the spatial configurations. The group aimed to define architecture as a spatial system that could directly correlate with human social activities through the application of new computer technologies. Our research presented here focuses on technologies and workflows that trace and translate human activities into architectural structures in order to continue the research agenda set out by Negroponte and others in the 1970s. The research work discusses new scenarios for the creation of architectural structures, using mobile and low-cost fabrication devices, and generative design algorithms driven by sensory technologies. The research question focuses on how architects may script individual and unique processes for generating structures using rule-sets that organise materiality and spatial relationships in order to achieve a user-driven outcome. Our explorations follow a renewed interest in the paradigm where the architect is a ‘process designer’, aiming to generate emergent outcomes where the inherent complexity of the project is generated towards specific performance criteria related to human activities and inhabitation

    ' Pay-as-you-go City ': New Forms of Domesticity in a Technological Society

    Ongoing urbanization, combined with market fundamentalism as the prevailing mode of political management, is leading to the spatial and social segregation of economic classes in cities. The housing market, being driven by economic interests rather than public policy, favors inflexible forms of ownership or tenancy that are increasingly incompatible with the more diverse forms of live-work patterns and family structures occurring in the society. This paper presents a research-by-design project that explores a speculative future scenario of housing, based on current developments in digital technologies and their impact on the mobility and accessibility to services enjoyed by urban residents. It references technology platforms that underpin the 'sharing economy' or 'gig economy', such as 'pay-as-you-go' car and bike sharing programs or internet and smartphone-based services for taxis or temporary accommodation. The study explores how new forms of participation in the housing market could circumvent the current segregation of different communities across the city. It describes a speculative system of distributed residential spaces, accessible to all on a 'pay-for-time-used' basis. By offering freedom of choice across domestic functions of greater range and accessibility than found within existing housing or hotel accommodation, the system would enable opportunistic or nomadic forms of living linked to the dynamic spatio-temporal occurrences of social, cultural or economic opportunities. The research references how new forms of social networking create new challenges and opportunities to participate in communities and explores how new technologies, applied to housing, can help to find a 'sense of belonging' within the technological society

    Digital common(s): the role of digital gamification in participatory design for the planning of high-density housing estates

    “Digital Commons” explores the intersection between participatory design, digital gamification, and community engagement, contextualised in the planning of high-density housing estates in Hong Kong. The research project investigates how digital gamified participatory design can be applied in decision-making processes for the planning of public facilities in high-density housing estates. Focusing on community engagement methods, the project has engaged with residents of a case study housing estate, Jat Min Chuen in the Shatin Wai area of Hong Kong, to facilitate collective planning discussions about the past, present, and future of community facilities. Using a digital community game approach, it has collected opinions and needs from public housing residents, promoted collaborative design thinking processes, and provided a platform for participants to increase their understanding of the complexity of planning problems through 3D visualisation tools. The experiences documented in this study demonstrate how 3D interactivity, real-time engagement, and bottom-up perspectives may enhance the potential of using immersive digital twins during collective decision-making. The gaming outcomes show a high similarity across all teams in close relationship to users’ daily life routines, demonstrating a new powerful role for urban designers as a coordinator of interactive and collaborative planning processes

    A versatile assay for protein tyrosine phosphatases (PTP) employing 3-nitrophosphotyrosine containing peptidic substrates is described. These therapeutically important phosphatases feature in signal transduction pathways. The assay involves spectrophotometric detection of 3-nitrotyrosine production from 3-nitrophosphotyrosine containing peptidic substrates, which are accepted by many PTPs. Compared to conventional chromogenic phosphate derivatives, the more realistic peptidic substrates allow evaluating substrate specificity. The assay’s applicability is demonstrated by determining kinetic parameters for several PTP-substrate combinations and inhibitor evaluation, as well as detection of PTP activity in lysates. The convenient new assay may assist further adoption of PTPs in drug development

    A novel strategy to prepare bisubstrate based inhibitors for histone acetyltransferases is presented. To obtain these, azido peptides derived from histone H3 incorporating either a serine or a phosphoserine residue were connected to a propargyl coenzyme A derivative through copper catalyzed click chemistry. The resulting inhibitors were tested with therapeutically relevant acetyltransferase PCAF. Increased potency of the phosphoserine containing inhibitor was observed. The synthetic strategy presented may be used for developing bisubstrate based inhibitors against any acetyltransferase

    A high-throughput method for the determination of the kinetics of protein tyrosine phosphatase (PTP) activity in a microarray format is presented, allowing real-time monitoring of the dephosphorylation of a 3-nitro-phosphotyrosine residue. The 3-nitro-phosphotyrosine residue is incorporated in potential PTP substrates. The peptide substrates are immobilized onto a porous surface in discrete spots. After dephosphorylation by a PTP, a 3-nitrotyrosine residue is formed that can be detected by a specific, sequence-independent antibody. The rate of dephosphorylation can be measured simultaneously on 12 microarrays, each comprising three concentrations of 48 clinically relevant peptides, using 1.0–5.0 μg of protein from a cell or tissue lysate or 0.1–2.0 μg of purified phosphatase. The data obtained compare well with solution phase assays involving the corresponding unmodified phosphotyrosine substrates. This technology, characterized by high-throughput (12 assays in less than 2 h), multiplexing and low sample requirements, facilitates convenient and unbiased investigation of the enzymatic activity of the PTP enzyme family, for instance by profiling of PTP substrate specificities, evaluation of PTP inhibitors and pinpointing changes in PTP activity in biological samples related to diseases
