Molecular simulation and spectroscopic studies on the interaction between perfluorohexadecanoic acid and human serum albumin

    Get PDF
185-192In the present study, the interaction between Perfluorohexadecanoic acid (PFHxDA) and human serum albumin (HAS) was studied by fluorescence spectroscopy, molecular docking, dynamic simulation and circular dichroism (CD). The interaction character and the effect on human serum albumin conformation were measured by simulating the physiological condition (pH= 7.4). Experiments and simulation results revealed that PFHxDA molecules and HSA have regular fluorescence quenching, and the quenching mechanism is static quenching and non-radiative energy transfer. Thermodynamic analysis revealed the binding behavior was mainly governed by hydrophobic forces. Specific binding site experiments showed that the binding site of PFHxDA was a site I of HSA. The results from the CD spectrum demonstrated that PFHxDA changed the molecular conformation of HSA, which is consistent with the results obtained by molecular docking and dynamic simulation

Horizontal structure of convergent wind shear associated with sporadic E layers over East Asia

At present, the main detection instruments for observing sporadic E (Es) layers are ground-based radars, dense networks of ground-based global navigation satellite system (GNSS) receivers, and GNSS radio occultation, but they cannot capture the whole picture of the horizontal structure of Es layers. This study employs the Whole Atmosphere Community Climate Model with thermosphere and ionosphere eXtension model (WACCM-X 2.1) to derive the horizontal structure of the ion convergence region (HSICR) to explore the shapes of the large-scale Es layers over East Asia for the period from June 1 to August 31, 2008. The simulation produced the various shapes of the HSICRs elongated in the northwest−southeast, northeast−southwest, or composed of individual small patches. The close connection between Es layer critical frequency (foEs) and vertical ion convergence indicates that the HSICR is a good candidate for revealing and explaining the horizontal structure of the large-scale Es layers

In order to study the effect of germinated mung bean powder on the quality and nutrition of yogurt, different varieties of mung bean were selected in this paper, germinated mung bean sprout powder and pure milk as raw materials,Lactobacillus bulgaricus and Streptococcus thermophilus as fermentation strains, solidified germinated mung bean yoghurt was developed by single factor and orthogonal test; The contents of vitexin, isovitexin and cellulose in the germination mung bean powder were determined by high performance liquid chromatography and enzyme-weight method to explore the effects of adding germination mung bean powder on the quality and nutrition of yogurt. The results showed that the addition of germinated mung bean powder could strengthen the tissue structure of yogurt, increase the number of viable lactic acid bacteria, and give the yogurt a unique mung bean flavor; Moreover, the addition of germinated mung bean powder could enhance the antioxidant properties of yogurt, increase soluble dietary fiber, beneficial ingredients such as vitexin and isovitexin, and give yogurt physiological health functions. The best process for coagulated germination mung bean yogurt was:adding 2% germination mung bean powder,7% sucrose,0.3% CMC stabilizer, adding starter at the ratio of 1.5 g/L,and incubating at 42 ℃ for 5 hours.Under these conditions the germinated mung bean yogurt had a stable structure and good flavor and taste

Clearance of Free Silica in Rat Lungs by Spraying with Chinese Herbal Kombucha

The effects of spraying with kombucha and Chinese herbal kombucha were compared with treatments with tetrandrine in a rat silicosis model. Silica dust (50 mg) was injected into the lungs of rats, which were then treated with one of the experimental treatments for a month. The rats were then killed and the effects of the treatments were evaluated by examining the extent and severity of the histopathological lesions in the animals’ lungs, measuring their organ coefficients and lung collagen contents, determining the dry and wet weights of their lungs, and measuring the free silica content of the dried lungs. In addition, lavage was performed on whole lungs taken from selected rats, and the numbers and types of cells in the lavage fluid were counted. The most effective treatment in terms of the ability to reduce lung collagen content and minimize the formation of pulmonary histopathological lesions was tetrandrine treatment, followed by Chinese herbal kombucha and non-Chinese herbal kombucha. However, the lavage fluid cell counts indicated that tetrandrine treatment had severe adverse effects on macrophage viability. This effect was much less pronounced for the kombucha and Chinese herbal kombucha treatments. Moreover, the free silica levels in the lungs of animals treated with Chinese herbal kombucha were significantly lower than those for any other silica-exposed group. These preliminary results indicate that spraying with Chinese herbal kombucha preparations can effectively promote the discharge of silica dust from lung tissues. Chinese herbal kombucha inhalation may thus be a useful new treatment for silicosis and other pneumoconiosis diseases

Ultrasound measurement of vastus lateralis and vastus medialis muscle parameters to identify chronic thyrotoxic myopathy

Introduction: Chronic thyrotoxic myopathy (CTM) is a common, easily neglected complication of hyperthyroidism. There are currently no standard diagnostic criteria for CTM, and the ultrasonic characteristics of CTM-affected skeletal muscle remain unclear. Herein, we aimed to evaluate hyperthyroid patients for CTM by ultrasound and identify ultrasonic muscle parameter cutoffs for CTM diagnosis. Materials and methods: Each participant underwent ultrasonography. The original (muscle thickness (MT), pennation angle (PA), and cross-sectional area (CSA)) and corrected (MT/height (HT), MT/body mass index (BMI), CSA/HT, and CSA/BMI) parameters of the vastus lateralis and vastus medialis (VM) were evaluated. The diagnostic effectiveness of ultrasound for predicting CTM was determined using receiver operating characteristic (ROC) curve analysis. Our study included 203 participants: 67 CTM patients (18 males, 49 females), 67 non-CTM patients (28 males, 39 females) and 69 healthy controls (20 males, 49 females). Results: The CTM group had lower muscular ultrasonic and anthropometric parameters, higher thyroid hormone and thyroid-stimulating hormone receptor antibody (TRAb) levels, and a longer duration of hyperthyroidism than the non-CTM group (P < 0.05). The VM-PA, VM-CSA, VM-CSA/HT, and VM-CSA/BMI were lower in females than in males (P < 0.05). Free thyroxine (FT4) and TRAb both showed significant negative correlations with VM-MT, VM-MT/HT, VM-CSA, and VM-CSA/HT (P < 0.05). VM-MT/BMI and VM-CSA/HT, respectively, best predicted male and female CTM (AUC = 0.84, 0.85; cutoff ≤ 0.07, < 4.01). Conclusion: Ultrasound measurement of muscular parameters, especially in the VM, is a valid and feasible way of diagnosing and characterizing possible CTM in hyperthyroidism
