    Serological prevalence of leptospiral infection in cattle from ladang angkat and Taman Pertanian Universiti, Universiti Putra Malaysia

    The study was conducted to determine the serological prevalence of leptospiral infection in cattle from ladang angkat and Taman Pertanian Universiti, Universiti Putra Malaysia (UPM). Leptospirosis is known as an important disease in Malaysia which causes economic losses to the farmers as well as human infection. Previous studies revealed that there were high prevalence rates of leptospiral infection in Malaysia. One hundred fifty serum samples were collected from ladang angkat and Taman Pertanian Universiti, UPM. The samples were then tested for antibodies against 16 Leptospira serovars (Australis, Autumnalis, Ballum, Bataviae, Canicola, Celledoni, Djasiman, Hardjobovis, Hardjo, Hebdomadis, Hurstbridge, Icterohaemorrhagiae, Javanica, Pomona, Pyrogenes and Sejroe) using the microscopic agglutination test (MAT). Thirty (20%) of the 150 samples were positive on MAT. Of the 30 MAT-positive serum samples, 18 (12%) were positive against serovar Hardjo, 6 (4%) were positive against serovar Pomona, 2 (1.33%) were positive against serovars Bataviae and Autumnalis, and 1 (0.6%) was positive for serovars Celledoni, Hardjobovis, and Hebdomadis. Kedah-Kelantan breed was shown to have a high prevalence rate. Intensive system showed the highest prevalence rate when compared to the other management system. The age group of 1-2 years old also had the highest prevalence rate when compared to other age groups. Statistical analysis indicated a significant difference (p<0.05) in serological prevalence among different breeds, farms, management systems, and age groups of cattle examined

    Herbal-Based Formulation Containing <i>Eurycoma longifolia</i> and <i>Labisia pumila</i> Aqueous Extracts: Safe for Consumption?

    A combined polyherbal formulation containing tongkat ali (Eurycoma longifolia) and kacip fatimah (Labisia pumila) aqueous extracts was evaluated for its safety aspect. A repeated dose 28-day toxicity study using Wistar rats was conducted where the polyherbal formulation was administered at doses 125, 500 and 2000 mg/kg body weight to male and female treatment groups daily via oral gavage, with rats receiving only water as the control group. In-life parameters measured include monitoring of food and water consumption and clinical and functional observations. On day 29, blood was collected for haematological and biochemical analysis. The rats were necropsied and the organs were collected for histopathological examination. This study showed that the combined formulation did not induce any significant toxicity effect at any dose level in terms of morbidity, mortality, behaviour, functional observation, body weight, food and water consumption, whole blood haematology and serum biochemistry. However, there were some microscopic changes in the histopathological examinations of some organs given 2000 mg/kg body weight, which may suggest an early response to the polyherbal formulation. From this study, the no observed adverse effect level is estimated to be more than 500 mg/kg body weight but not exceeding 2000 mg/kg body weight. The observed effects at the highest dose indicate the need for further study of longer dosing duration

    Bioavailability of Eurycomanone in Its Pure Form and in a Standardised Eurycoma longifolia Water Extract

    Eurycoma longifolia is one of the commonly consumed herbal preparations and its major chemical compound, eurycomanone, has been described to have antimalarial, antipyretic, aphrodisiac, and cytotoxic activities. Today, the consumption of E. longifolia is popular through the incorporation of its extract in food items, most frequently in drinks such as tea and coffee. In the current study, the characterisation of the physicochemical and pharmacokinetic (PK) attributes of eurycomanone were conducted via a series of in vitro and in vivo studies in rats and mice. The solubility and chemical stability of eurycomanone under the conditions of the gastrointestinal tract environment were determined. The permeability of eurycomanone was investigated by determining its distribution coefficient in aqueous and organic environments and its permeability using the parallel artificial membrane permeability assay system and Caco-2 cultured cells. Eurycomanone&rsquo;s stability in plasma and its protein-binding ability were measured by using an equilibrium dialysis method. Its stability in liver microsomes across species (mice, rat, dog, monkey, and human) and rat liver hepatocytes was also investigated. Along with the PK evaluations of eurycomanone in mice and rats, the PK parameters for the Malaysian Standard (MS: 2409:201) standardised water extract of E. longifolia were also evaluated in rats. Both rodent models showed that eurycomanone in both the compound form and extract form had a half-life of 0.30 h. The differences in the bioavailability of eurycomanone in the compound form between the rats (11.8%) and mice (54.9%) suggests that the PK parameters cannot be directly extrapolated to humans. The results also suggest that eurycomanone is not readily absorbed across biological membranes. However, once absorbed, the compound is not easily metabolised (is stable), hence retaining its bioactive properties, which may be responsible for the various reported biological activities

    The Effect of Cement and wrapping on the Decomposition rate of the Rabbit Carcasses

    Taphonomic study was first time conducted in Kuala Lumpur, capital city of Malaysia. In this research model, male Oryctolagus cunicullus weighed 1.8-2.6 kg each were killed by Dolethal intravenously at the ear region. In Phase 1 study, eight subjects were wrapped in a sack and cemented within a container to be compared with the other four controls at each location labelled A (ground) and B (manmade freshwater pond). Phase 2 involved duplicating set of 15 cemented samples comparable to 1 control made up to total of 32 subjects which were all put into a compact polytank containing freshwater to simulate a case study. Taphonomic changes were observed and scored using TBS system incorporating of fresh stage, early decomposition stage, advanced decomposition stage and skeletonisation stage. The ambient temperature of surroundings with 28.81C&plusmn;4.21&deg;C and 29.21&deg;C&plusmn;4.57&deg;C (mean&plusmn;S.D.) while relative humidity of air with 74.49 %&plusmn;14.61% and 79.15%&plusmn;16.32% (mean&plusmn;S.D.) were recorded for Phase 1 and Phase 2 study respectively. Time taken for ground control exposed and wrapped carcasses to reach first sign of skeletonised stage were four days and five days respectively. Whilst freshwater controls reached initial skeletonised stage within one week for exposed carcass and two weeks for wrapped carcass. Within the control and cemented sample carcasses, TBS scores increased from initial stages of decomposition and become plateau after advanced decomposition. The cementing factor have superseded the wrapping factor due to its stronger physical barrier effect to slow down the decomposition more than half compared to controls based on Multiple way ANOVA test. Phase 2 study has demonstrated more accurately on the decomposition rate of the cemented samples. It showed that wrapping and cementing factors have delayed the decomposition process of the rabbit carcasses about 4 times to reach the initial stage of skeletonisation compared to the control carcasses. There was absence of insect activity within the cemented samples, hence the microorganism activity would be the only contributor to the decaying process within the cemented samples at slower rate comparing to the exposed or wrapped controls
