The mean field infinite range p=3 spin glass: equilibrium landscape and correlation time scales

We investigate numerically the dynamical behavior of the mean field 3-spin spin glass model: we study equilibrium dynamics, and compute equilibrium time scales as a function of the system size V. We find that for increasing volumes the time scales τ\tau increase like lnτV\ln \tau \propto V. We also present an accurate study of the equilibrium static properties of the system.Comment: 6 pages, 9 figure

4D Spin Glasses in Magnetic Field Have a Mean Field like Phase

By using numerical simulations we show that the 4D J=±1J=\pm 1 Edwards Anderson spin glass in magnetic field undergoes a mean field like phase transition. We use a dynamical approach: we simulate large lattices (of volume VV) and work out the behavior of the system in limit where both tt and VV go to infinity, but where the limit VV \to \infty is taken first. By showing that the dynamic overlap qq converges to a value smaller than the static one we exhibit replica symmetry breaking. The critical exponents are compatible with the ones obtained by mean field computations.Comment: Physrev format, 5 ps figures include

Disrupting HIV-1 capsid formation causes cGAS sensing of viral DNA

    Get PDF
Detection of viral DNA by cyclic GMP-AMP synthase (cGAS) is a first line of defence leading to the production of type I interferon (IFN). As HIV-1 replication is not a strong inducer of IFN, we hypothesised that an intact capsid physically cloaks viral DNA from cGAS. To test this, we generated defective viral particles by treatment with HIV-1 protease inhibitors or by genetic manipulation of gag. These viruses had defective Gag cleavage, reduced infectivity and diminished capacity to saturate TRIM5α. Importantly, unlike wild-type HIV-1, infection with cleavage defective HIV-1 triggered an IFN response in THP-1 cells that was dependent on viral DNA and cGAS. An IFN response was also observed in primary human macrophages infected with cleavage defective viruses. Infection in the presence of the capsid destabilising small molecule PF-74 also induced a cGAS-dependent IFN response. These data demonstrate a protective role for capsid and suggest that antiviral activity of capsid- and protease-targeting antivirals may benefit from enhanced innate and adaptive immunity in vivo

Extending Hybrid CSP with Probability and Stochasticity

Probabilistic and stochastic behavior are omnipresent in computer controlled systems, in particular, so-called safety-critical hybrid systems, because of fundamental properties of nature, uncertain environments, or simplifications to overcome complexity. Tightly intertwining discrete, continuous and stochastic dynamics complicates modelling, analysis and verification of stochastic hybrid systems (SHSs). In the literature, this issue has been extensively investigated, but unfortunately it still remains challenging as no promising general solutions are available yet. In this paper, we give our effort by proposing a general compositional approach for modelling and verification of SHSs. First, we extend Hybrid CSP (HCSP), a very expressive and process algebra-like formal modeling language for hybrid systems, by introducing probability and stochasticity to model SHSs, which is called stochastic HCSP (SHCSP). To this end, ordinary differential equations (ODEs) are generalized by stochastic differential equations (SDEs) and non-deterministic choice is replaced by probabilistic choice. Then, we extend Hybrid Hoare Logic (HHL) to specify and reason about SHCSP processes. We demonstrate our approach by an example from real-world.Comment: The conference version of this paper is accepted by SETTA 201

Present-Day Surface Deformation in North-East Italy Using InSAR and GNSS Data

    Get PDF
Geodetic data can detect and estimate deformation signals and rates due to natural and anthropogenic phenomena. In the present study, we focus on northeastern Italy, an area characterized by similar to 1.5-3 mm/yr of convergence rates due to the collision of Adria-Eurasia plates and active subsidence along the coasts. To define the rates and trends of tectonic and subsidence signals, we use a Multi-Temporal InSAR (MT-InSAR) approach called the Stanford Method for Persistent Scatterers (StaMPS), which is based on the detection of coherent and temporally stable pixels in a stack of single-master differential interferograms. We use Sentinel-1 SAR images along ascending and descending orbits spanning the 2015-2019 temporal interval as inputs for Persistent Scatterers InSAR (PSI) processing. We apply spatial-temporal filters and post-processing steps to reduce unrealistic results. Finally, we calibrate InSAR measurements using GNSS velocities derived from permanent stations available in the study area. Our results consist of mean ground velocity maps showing the displacement rates along the radar Line-Of-Sight for each satellite track, from which we estimate the east-west and vertical velocity components. Our results provide a detailed and original view of active vertical and horizontal displacement rates over the whole region, allowing the detection of spatial velocity gradients, which are particularly relevant to a better understanding of the seismogenic potential of the area. As regards the subsidence along the coasts, our measurements confirm the correlation between subsidence and the geological setting of the study area, with rates of similar to 2-4 mm/yr between the Venezia and Marano lagoons, and lower than 1 mm/yr near Grado

Multidisciplinary Conservative Treatment of Difficult Bile Duct Stones: A Real Alternative to Surgery

    Get PDF
56 patients with large CBD or intrahepatic stones underwent endoscopic and/or percutaneous treatment followed by extracorporeal shock wave lithotripsy. Percutaneous access to the biliary tract was chosen when an endoscopic approach was not possible (hepaticojejunostomy in 5 patients, 1 juxtapapillary diverticulum and I inflammatory bile duct stricture). Visualization of stones was achieved radiologically in 32 patients and by ultrasound in 24. The procedure was successful in 47 of 56 treated patients (83.9%). Clearance of the biliary tract was obtained in 25 cases (53%), whereas in 22 cases (47%) complete clearing of biliary tract was obtained only after endoscopic extraction of fragments (17 cases) or percutaneous (5 cases). The median number of shock waves in each session was 1725 (range 300–3166), which were applied during one (n=30), two (n=22) or three sessions (n=4). The only complications were 1 case of symptomatic hyperamylasemia and 3 cases of macrohematuria. In conclusion, extracorporeal lithotripsy combined with endoscopic and/or percutaneous treatment is a real alternative to surgery for difficult stones

SARS-CoV-2 sensing by RIG-I and MDA5 links epithelial infection to macrophage inflammation

    Get PDF
SARS-CoV-2 infection causes broad-spectrum immunopathological disease, exacerbated by inflammatory co-morbidities. A better understanding of mechanisms underpinning virus-associated inflammation is required to develop effective therapeutics. Here we discover that SARS-CoV-2 replicates rapidly in lung epithelial cells despite triggering a robust innate immune response through activation of cytoplasmic RNA-sensors RIG-I and MDA5. The inflammatory mediators produced during epithelial cell infection can stimulate primary human macrophages to enhance cytokine production and drive cellular activation. Critically, this can be limited by abrogating RNA sensing, or by inhibiting downstream signalling pathways. SARS-CoV-2 further exacerbates the local inflammatory environment when macrophages or epithelial cells are primed with exogenous inflammatory stimuli. We propose that RNA sensing of SARS-CoV-2 in lung epithelium is a key driver of inflammation, the extent of which is influenced by the inflammatory state of the local environment, and that specific inhibition of innate immune pathways may beneficially mitigate inflammation-associated COVID-19
